A pályázat kutatási célja, hogy a magas vércukorszint következtében kialakuló vesebetegségekre patológiás biomarkereket találjunk. A vérben nem enzimatikusan glikálódó és a vizeletben is kiválasztott fehérjék- főleg a szérum albumin- analízise ezt lehetővé teszik. Kellő számú minta proteomikai analízisével összefüggést kaphatunk a betegség típusa, illetve a fehérjék glikáltsági szintje között, mely egyben egy eddig le nem írt biomarkere lehet a betegségnek. A kutatás további célja, hogy a vérben elsődlegesen glikálódó, valamint a vizeletben kiválasztott fehérjék módosulásainak összehasonlításával információt nyerjünk a vesében bekövetkező oxidatív folyamatokról. A betegek szérumából, illetve vizeletéből izolált, majd emésztett fehérjeminták analízise micro-HPLC-MS módszerrel történik, de munkánk további metodikai célja, hogy egyébb elválasztástechnikai módszerekkel (on-és off-line CE-MS, SPE-off-line MS technikák) is tanulmányozzuk a nem enzimatikus glikációt. Mivel biológiai mintákban a glikált fehérje mellett jelentős mennyiségben nem glikált fehérjék is előfordulnak, céljaink között szerepel a glikált fehérjék, majd ezek emésztését követően a glikált peptidek affinitás kromatográfiával történő szelektív dúsítása, megfelelő affinitás kromatográfiás fázis előállítása.
Summary
The aim of this tender is to explore pathological biomarkers for those renal diseases evolved by the high level of glucose in diabetes.Analysis of proteins (mainly serum albumin) glycated non enzimatically primarily in blood serum and of those excreted and isolated from urine enables us to study extensively this question. By means of analysis of appropriate number of samples an important relationship must be explained between the type of disease and the level of glycation that can be a possible biomarker. We are also interested in comparing the proteins glycated in blood serum with those excreted to urine in order to get information about oxidative processes occuring in case of renal failures.The proteins will be isolated, from human urine and blood, digested by trypsin and finally, the digests will be subjected to HPLC-MS analysis. Besides using mainly HPLC-MS method for the analysises we are going to study the non-enzymatic glycation with other tecniques such as on- and off-line CE-MS and SPE-off line MS. Due to the presence of non modyfied protein in biological samples, the enrichment of glycated proteins/peptides are required, therefore efforts will be made to develop and use affinity chromatographic phases .

• Új mintaelőkészítési eljárást dolgoztunk ki nem-enzimatikusan glikálódott fehérjék triptikus emésztményének különböző szilikagél alapú állófázisokon (C18, C30, C60(10)-, C60(30)- , C60(100)-fullerénszilika) történő frakcionálására és dúsítására. • Elsőként térképeztük fel a fibrinogén lehetséges glikációs helyeit. • Optimalizáltuk a glikált fehérje emésztmények szelektív dúsítására alkalmas boronát-affinitáskromatográfiás körülményeket. • Meghatároztuk a diabetes mellitus-ban szenvedő betegek vérszérumából nyert HSA (Human Serum Albumin) glikáltsági szintjét. • Különbséget véltünk felfedezni a beteg és kontrol csoport szérum és vizelet HSA glikáltsági szintje között. A várakozásoknak megfelelően több lehetséges glikációs hely módosult a diabetes mellitus-ban szenvedő betegek esetében. • A HSA 414K és a 428R glikációs helyei biomarker jelentőségűek lehetnek. • Különbséget mutattunk ki MALDI-TOF technikával Shigella zonei mutáns törzsek endotoxiunjainak (LPS) struktúráiban a teljes core és Lipid A szerkezetek megadásával. • Potenciális biomarkereket sikerült folyadékkromatografiás és tömegspektrometriás módszerekkel Crohn beteg vizeletéből azonosítani a II. számú Belgyógyászati Klinika együttműködésével. Ezen a fehérje biomarkerek mennyisége a betegség különböző stádiumaiban eltérő.

• New sample preparation method was developed for the fractionation on silica based materials (C18, C30, C60(10)-, C60(30)-, C60(100)-fullerene silica) and selective enrichment of the in vitro glycated tryptic peptides. • Possible glycated sites of the fibrinogen were described first. • The crucial circumstances of the boronate affinity chromatography were optimized for the selective enrichment of the glycated tryptic peptide. • The glycation level of the HSA (Human Serum Albumin) collected from patients suffering from type 2 diabetes was determined. • A difference was discovered between the glycation level of the serum and urine of the patients suffering from diabetes mellitus and the healthy volunteers. • 414K and 428R, glycation sites of the HSA might serve as a biomarker of diabetes mellitus • Distinction was established in the structure of mutant Shigella zonei exotoxins (LPS) by the whole structure of the core and Lipid A with MALDI-TOF mass spectrometer. • A potential biomarkers were determined from urine of a patient suffering from Crohn disease with liquid chromatograpy and mass spectrometric methods collaborating with the 2nd Department of Internal Medicine and Nephrology Centre. The amount of these proteins varies in different statuses of the disease.
