A javasolt projekt komplex megközelítéssel vizsgálná a központi idegrendszeri de- és remyelinsatioval összefüggő microRNS, génexpressziós és immunológiai változásokat sclerosis multiplexben és állatmodelljeiben. Az alkalmazott tiszta de/remyelinisatiós modell és szövetizolálási technika a miRNS és génexpresszió speciális és precíz elemzését teszi lehetővé. A microRNS és génexpressziós profilt ugyanazon mintákban elemeznénk, így lehetőség lenne a de- és remyelinisatióra jellemző egyes reguláló miRNS-ek és gének közötti összefüggések vizsgálatára. A demyelinisatiót követő remyelinisatio során differenciáltan expresszálódó miRNS-eket és géneket vizsgálnánk fizikológiás myelinisatio során is újszülött állatokban, a két folyamat kapcsolatának elemzésére. A tiszta de- és remyelinisatio során azonosított. miRNS és géneket a sclerosis multiplex autoimmun gyulladásos modelljében, myelin oligodendrocyta glycoprotein T sejt receptor transzgén egérben is vizsgálnánk kísérletes autoimmun agyvelőgyulladás során . Amennyiben a differenciáltan expresszálódó gének effektor proteineket kódolnak, további vizsgálatokat tervezünk az in situ jelenlét igazolására, illetve annak elemzésére, hogy hiányuk és adagolásuk hogyan befolyásolja a de- és remyelinisatiót in vivo és in vitro modellekben.
A nyert adatokat sclerosis multiplexre extrapolálnánk: az azonosított géneket és potenciális miRNS-eket MS plakkokban is vizsgálnánk. Interferon-béta által miRNS indukálható makrofágokban, ezért IFN-bétával és egyéb immunmoduláló szerekkel kezelt SM betegek mononukleáris sejtjeiben a miR-155 és az előzetes adataink alapján differenciáltan expresszálódó let-7d expressziót külön elemeznénk. Szintén vizsgálnánk IFN-béta hatását a kísérletes demyelinisatiót kísérő miRNS és génexpresszióra, klinikumára szövettani és 9T MRI mérésekkel is.
A javasolt tervezet szorosan kapcsolódik a klinikánkon jelenleg zajló, regulátoros RNA kísérletekhez. A költségek csökkentése érdekében a kísérletek egy részét külföldi kollaborációban végeznénk.
A javasolt kísérletek fontos adatokat nyújthatnak a de- és remyelinisatio molekuláris mechanizmusaira, és új, terápiás potenciállal rendelkező molekulák azonosítását eredményezhetik.
Summary
Here, we propose a complex approach to explore microRNAs, gene expression and inflammation related to de- and remyelination in the CNS.
MicroRNA and gene expression will be tested in experimental de- and remyelination. The model and tissue sampling provide a careful setting for miRNA/gene analysis. By using the same samples for miRNA and gene expression microarray profiling, we may identify regulatory connections between particular miRNAs and genes related to de- and remyelination. Genes and miRNAs specific to remyelination will be checked in physiological myelination in newborn animals to examine whether these two processes are functionally different. The identified miRNA and genes related to pure de- and remyelination will be next examined in the autoimmune inflammatory model of multiple sclerosis (MS), using myelin oligodendrocyte glycoprotein T cell receptor transgenic mice. If the identified genes encode for effector proteins (eg. apoptosis, immune molecules), further in vitro and in vivo experiments are planned.
The data will be extrapolated to MS: the identified genes and potential miRNAs will be checked in MS plaques. MiRNAs, which can be induced in vitro by IFN-beta and have been found differentially expressed in MS in preliminary experiments , will be examined in MS patients treated with different protolcols with IFN-beta and glatiramer acetate. The effect of IFN-beta on experimental demyelination will be also tested using histology and 9T MRI and alteration in miRNAs/gene expression will be analyzed.
The proposal is related to ongoing experiments regarding regulatory RNAs in our department. To reduce expenses, the planned experiments will be partly done in foreign collaboration.
Our data will provide important data about molecular mechanisms of de- and remyelination, and we may identify molecules with therapeutic potential in MS.

A projekt folyamán de- és remyelinisatio gén expresszióját és ennek poszt-traszkripciós szabályozását vizsgáltuk állatmodellben. Array és kvantitatív RT-PCR alkalmazásával 3 miRNS differenciált expresszióját észleltük a de- és remyelinisatio során: egy miRNS funkciója eddig nem volt ismert. Ez a miRNS a fejlődő idegrendszer corpus callosumában is differenciáltan expresszálódott, ami a myelinisatio szabályozásában játszott szerepére utalhat. A prediktív target géneket 5 adatbázis elemzésével vizsgáltuk, majd útvonalelemzést végeztünk. Microarray alkalmazásával vizsgáltuk a génexpressziót a de- és remyelinisalt corpus callosumban. Beállítottuk az in situ hybridisatiot a miRNS detektálására; jelenleg zajlik a csendesítés oligodendrocyta sejtvonalon. Az eredmények a de- és remyelinisatio géenexpressziós szabályozásának molekuláris mechanizmusát, génexpressziós és miRNS által szabályozott útvonalakat tárhatnak fel. Szintén zajlik génexpressziós array-ek eredményeinek összehasonlítása a demyelinisalt és a gyulladásos állatmodellekben, illetve sclerosis multiplex agyi plakkokból: ez degeneratív és gyulladásos biomarkerek azonosítását célozza, melyet az SM hosszú távú prognózis predikciójára alkalmaznánk.
Results in English
We have examined de- and remyelination-related gene expression and its post-transcriptional regulation in an animal model of MS. By using array and quantitative RT-PCR, we have identified differential expression of 3 miRNAs during de- and remyelination: function of one miRNA has not been known. This particular miRNA was also differentially expressed during developmental myelination. We also screened for predictive target genes using 5 different databases, and performed pathway analysis. Furthermore gene expression in the de- and remyelinated corpus callosum has been also examined by microarray. In situ hybridisation has been established for detection of miRNAs, and we have started silencing experiments by establishing oligodendrocyte cell lines. Our results may reveal molecular pathways of gene expression and gene expression regulation related to de- and remyelination. We have also started comparing gene expression in degenerative and inflammatory models of MS, as well a sin MS plaques. These experiments may result in identification of novel degenerative and inflammatory biomarkers, which we can translate back to human MS and use in prediction of long-term prognosis.
