A gerinctelen állatok idegrendszere viszonylag egyszerű, kevés-sejtes felépítésük miatt kíváló lehetőséget nyújt egyes, a magatartást meghatározó szabályozó folyamatok sejtszintű eseményeinek vizsgálatára. Jelen pályázatunk célja, hogy az összehasonlító, gerinctelen neurobiológiai kutatások egyes modell állataiban, csigákban (Helix pomatia, Limax maximus) elemezzük azokat a szerveződési elveket, melyek a gerincesekével analóg szenzo-motoros információfeldolgozás strukturális és funkcionális alapjait meghatározzák. Kutatásaink tárgya a szaglórendszer, melyben vizsgáljuk az érző (szagló) bemenetek perifériás és központi integrációs rendszereinek felépítését és mintázatait, valamint az ezek által kialakított lokális és a viselkedést meghatározó motoros válaszokhoz tartozó interneuronális és ideg-izom kapcsolatok ultrastrukturális (szinaptológia), neurokémiai (transzmitterek és modulátorok) és fiziológiai (receptorok/ion csatornák) jellemzőit. Az intercelluláris kapcsolatok kialakulását mind a szinapszisok, mind az ioncsatornák és receptorok expressziója szintjén in vitro körülmények között (sejttenyészet) modellezzük, ahol idegnövekedési és adhéziós molekulák szerepét is vizsgáljuk. Kutatási eredményeink alapkutatás jellegűek, de egyúttal rávilágíthatnak az élővilág környezetünket meghatározó öko- és populációs stratégiai folyamatainak infraindividuális hátterére, illetve egyes vonatkozásai a gyógyszerkutatás területén is felhasználhatók lehetnek.
Summary
Because of the relative simple organization of the nervous system of invertebrates, consisting of only a small number of neurons, it provides an excellent comparative model for studying the cellular basis of regulatory processes underlying behavior. The aim of our current proposal is to analyze the organization principles determining the structural and functional basis of senso-motor information processing, analogous with that of vertebrates, in model species, snails (Helix, Limax), of invertebrate neurobiology. Object of our research is the olfactory system, in which we shall investigate the organization and arrangement of peripheral and central processing of sensory (olfactory) inputs, as well as the ultrastructural, (synaptology), neurochemical (transmitters, modulators) and physiological (ion channels/receptors) characteristics of interneuronal and neuromuscular contacts responsible for local and motor responses. Modeling of development and re-establishment of intercellular contacts, including the expression of ion channels and receptors, will be performed under in vitro conditions (cell culture), in the course of which the influence of nerve growth factors and cell adhesion molecules will also be studied. Beyond basic research, our results might shed light on the infraindividual background of eco- and population strategic processes determining environment, and they also could be used potentially in generating novel pharmaceutic compounds.

Nyelesszemű csigák (Helix, Limax) szaglórendszerét vizsgáltuk az olfaktórikus integrációs központ celluláris és neurokémiai szerveződése illetve a környezeti szaginformációkat lokalizáló tapogatók innervációja szintjén. A szaginformációk központi feldolgozásáért felelős procerebrum (PC) sejtes rétegében axo-szomatikus kapcsolatokat és lokális neuropil területeken komplex szinaptikus konfigurációkat tártunk fel, melyek szerepe oszcillációs illetve memória folyamatok szabályozásában feltételezhető. Az extrinsic 5-HTerg elemek innervációs rendszerét és szinaptológiáját is leírtuk; igazoltuk az 5-HT általános moduláló szerepét axo-szomatikus folyamatokban. Leírtuk a lokális field potenciálok (LFP) peptiderg (FMRFamid) szabályozásának elektrofiziológiai és molekuláris alapjait, továbbá a szagmemória folyamatokat befolyásoló NOerg rendszer ultrastruktúrális lokalizációját és intracelluláris cascade rendszerét. Na, Ca, és K feszültségfüggő ioncsatornák eltérő expresszióját és funkcióját igazoltuk a PC sejtekben, továbbá a Kv2.1, Kv3.4, Kv4.3 csatornák előfordulását a teljes CNS-ben is leírtuk. Azonosítottuk a tapogatók térbeli pozicionálását szabályozó flexor izmokat, leírtuk azok központi eredetű innervációját, neuromuszkuláris kapcsolatait, azonosítottuk transzmitterüket (acetilkolin) és aminerg (5-HT, dopamin) modulációjukat. Eredményeink a szaglás evolúciója és sejtszintű mechanizmusai általános törvényszerűségeinek részletesebb megismerését teszik lehetővé.

The olfactory system of stylommatophoran snails (Helix, Limax) was investigated at the level of the cellular and neurochemical organization of the integrative center, and the innervation of the tentacles responsible for the localization of odor stimuli. In the cell body layer of the procerebrum (PC), which is the site of olfactory information processing, axo-somatic contacts and complex synaptic configurations in local neuropils were demonstrated. Their role in the regulation of oscillation hence in olfactory memory is suggested. The innervation pattern and synaptology of the extrinsic 5-HTergic system as well as its general modulatory role were also described. The electrophysiological and molecular basis of the peptidergic (FMRFamide) regulation of the local field potentials, as well as the ultrastructural localization and intracellular cascade system of the NOergic system influencing odor memory were revealed. Differential expression and function of voltage dependent Na, Ca, és K channels were demonstrated in the PC cells, in addition to the localization of Kv2.1, Kv3.4, Kv4.3 channels in the entire CNS. Three flexor muscles responsible for the spatial positioning of the tentacles possessing the „nose” were identified. Their central innervation, neuromuscular contacts, fast transmitter (ACh) and aminergic (5-HT, dopamine) modulation were cleared. Our results contribute to a more detailed insight in the evolution and inter- and intracellular mechanisms of olfaction.
