A burok nélküli, pozitív egyszálú RNS (+ssRNS) genomú vírusok - melyek evolúciós szinten prokarióta mRNS molekulákat utánoznak – a legszámosabbak a planétán; 27 víruscsaládja ismert, melyek közül 9 víruscsalád emlősök vírusai, és a számuk gyorsan emelkedik a korszerű metagenomikai módszerek segítségével. A burok nélküli vírusok az Astroviridae, Caliciviridae, Hepeviridae és a Picornaviridae családokban számos közös és jellegzetes tulajdonsággal rendelkeznek.
A kutatással megvalósítható, hogy (1) ígéretes mintákból, szisztematikusan, RT-PCR módszerekkel keressük és mutassuk ki a burok nélküli +ssRNS genomú vírusok új fajait és nemzetségeit (vírus diverzitás) a vírusok nagyon konzervatív genetikai motívumaira elméletileg lehetséges nukleotid variációkat figyelembe vevően tervezett konzervatív (és degenerált) primerek kombinációival; (2) „genome-walking” és korszerű metagenomikai módszerekkel (pl.: 454-pyroszekvenálás) meghatározhassuk az ígéretesen új vírus(ok) teljes genetikai állományát(-ait); és (3) molekuláris módszerekkel kimutassuk és jellemezzük az újonnan azonosított vírus variáns(ok), faj(ok) vagy nemzetség(ek) tagjait (beleértve a újonnan azonosított potenciálisan új picornavírus nemzetséget a „Hungarovirus”-t) a lehető legtöbb gazdafajból (gazdafaj diverzitás) így emberből is. A kutatással kitölthetjük és folytathatjuk azt a virtuális táblázatot, ahol a sorok a burok nélküli +ssRNS vírusok nemzetségeit és fajait, az oszlopok ezek gazdafajait tartalmazzák.
Summary
Viruses with positive, single-stranded RNA (+ssRNA) genome – which evolutionally imitate prokaryote mRNA molecule - are the most plentiful on this planet; 27 families have been recognized, including 9 families which infect mammals, and this number is increasing rapidly using up-to-date metagenomic methods. Non-enveloped +ssRNA viruses in families Astroviridae, Caliciviridae, Hepeviridae and Picornaviridae have many common and characteristic features.
In this research project there will be a (1) systematic search for identification of novel (species or genus of) non-enveloped viruses with +ssRNA genome (virus diversity) using a combination of conserved (and degenerate) primers designed for highly conserved genetic motifs based upon the theoretically possibly nucleotide variations from promising samples by RT-PCR methods; (2) characterization of the complete genome of novel promising virus(es) by genome-walking and up-to-date viral metagenomics methods (like 454-pyrosequencing); and (3) detection and characterization of the identified novel virus variant(s), species or genus (including the recently identified potential new picornavirus genus “Hungarovirus”) by molecular methods in as many animal host species as possible (host species diversity) including human. With this research, we can fill up and continue the virtual table where the rows represent the genera or species of non-enveloped +ssRNA viruses and the columns represent the range of its host species.

Az (re)emerging fertőző betegségek folyamatos fenyegetést jelentenek a házi és vadon élő állatokra és az emberre. A kutatásaink alátámasztják feltételezésünket, hogy új, burok nélküli, pozitív, egyszálú RNS (+ssRNS) genomú vírusokat lehet azonosítani a már ismert rokon vírusok genom-jellegzetességei figyelembe vételével a gazdafajok szisztematikus vizsgálatával. A négy kutatási év alatt több új vírusgeno-/szerotípus mellett összesen 19 taxonómiailag is új vírusfajt és ebből egyben 13 potenciálisan új névadó vírusnemzetséget (név szerint a Picornaviridae családban: Avisivirus, Gallivirus, Hunnivirus, Megrivirus, ”Kunsagivirus”, ”Sakobuvirus”, ”Falcovirus”, ”Orivirus”, ”Rafivirus”, ”Mesivirus”, ”Phacovirus”; a Picornavirales rendben: ”Halastavi árva RNA virus”, ”Fisavirus”) sikerült felfedezni és teljes genomjukat meghatározni 14 gazdafajból (emlősökből, madarakból, hüllőből és halból). A munka eredményei hozzájárultak a +ssRNS genomú vírusok diverzitásának megismeréséhez, a vírustaxonómia fejlődéséhez, a vírusok közötti mélyebb (valósabb) genetikai és evolúciós összefüggések, gazdafaj-specificitás és váltás (gazdafaj diverzitás), a rezervoir szerep, valamint a faji-határátlépések összefüggéseinek megértéséhez és erősítette az ember-állat-környezet egységében, a „One Health” koncepcióban való gondolkodást, együttműködést. Az eredményeket 36 elfogadott, illetve benyújtott „peer-reviewed” közleményben (+2 angol és 1 magyar nyelvű felkért könyvfejezetben) publikáltuk (ΣIF: 97,475)
Results in English
The (re)emerging infectious diseases pose a continuous threat to domestic and wild animals and humans. This study confirms our hypothesis that it is possible to discover novel, non-enveloped viruses with positive-sense, single-stranded RNA (+ssRNA) genome if we take account of the viral genome characteristics of known relatives and by systematic investigation of host species. During the 4-research-years, beside several novel viral geno-/serotypes, we were discovered and completely characterized a total of 19 taxonomically novel virus species including 13 potential novel, name founder viral genera (namely in family Picornaviridae: Avisivirus, Gallivirus, Hunnivirus, Megrivirus, ”Kunsagivirus”, ”Sakobuvirus”, ”Falcovirus”, ”Orivirus”, ”Rafivirus”, ”Mesivirus”, ”Phacovirus”; in order Picornavirales: ”Halastavi árva RNA virus”, ”Fisavirus”) from 14 host species (in mammals, birds, reptile and fish). The results of this study contributes to better recognition the diversity, progress of virus taxonomy and to understand deeper (real) genetic and evolutional relationship, host-species specificity and switch (host species diversity), reservoir role and cross-species transmission of +ssRNA viruses as well as it improved the integrative thinking and collaboration related to human-animal-environment in “One Health” conception. Results were accepted or submitted for publication in 36 peer-reviewed articles (+2 English and 1 Hungarian invited book chapters) (ΣIF: 97.475).
