A vasklorózis meszes/lúgos talajok esetén általános növekedési probléma, mely számos mediterrán területen lévő ültetvényt befolyásol. Súlyos elemhiány esetén a növények fejlődése és termőképessége erősen csökken, és a növény elpusztul. A vashiány leginkább az őszibarack, körte, kivi és citrusfélék termesztését befolyásolja. A vashiány megszüntetésére jelenleg a leghatásosabb módszer a szintetikus Fe-kelátok alkalmazása, jóllehet ez igen magas költségeket (több, mint 70 millió euró évente, csak Európában) jelent és emellett legtöbb esetben a talajba juttatott vegyületek lassan bomlanak le. Természetes komplexképzők alkalmazása jelenleg kidolgozás alatt áll, mivel ezek olcsóbb alternatívát jelentenek, és hamarabb lebomlanak. Ezek az anyagok egyben más mikroelemek hiányának megszüntetésében is hatékonyak lehetnek.
A különféle komplexek lehetőséget adnak, hogy a növények mikroelem szükségleteit biztosítsuk, de ezek hatásmechanizmusa és hatékonyságukat meghatározó tényezők máig nem ismertek, és az erre vonatkozó, eddig közölt eredmények igen eltérőek. A 57Fe Mössbauer-spektroszkópia alkalmas eszköz a Fe-kelátok speciációjának, szerkezetének vizsgálatára, ezáltal segíthet az egyes Fe-kelátok és -komplexek növényélettani hatásainak felderítésében.
Jelen projekt céljai tehát a következők: i) természetes Fe-komplexek szerkezetének és speciációjának tanulmányozása különböző kísérleti körülmények között; ii) Fe-kelátok és Fe-komplexek talajjal történő kölcsönhatásának vizsgálata; iii) az I. vasfelvételi stratégiát követő növényekben található Fe-vegyületek és a vas-kelát reduktáz enzim szubsztrátjaként szolgáló Fe-vegyületek vizsgálata.
Summary
Fe chlorosis in plants is a common nutritional disorder occurring in calcareous and/or alkaline soils,
that affects to many crops in the Mediterranean basin. Under severe deficiency conditions size and
productivity of plants is seriously reduced, and plant death can occur. High added value crops can be
affected by Fe deficiency, the most significant being fruit trees like peach, pear, kiwifruit and Citrus.
Currently synthetic chelates are the most effective way to solve Fe chlorosis, though they are expensive
(only in Europe, more than 70 millions € are spent yearly in Fe chelates) and in most of cases degrade very
slowly. As an alternative natural complexes, cheaper and more degradable than synthetic chelates, are
under consideration. Complexes are less specific and cheaper than chelates, so they can be used to solve
other micronutrient deficiencies, and are suitable to be applied on low add value crops that currently can
not be treated with chelates.
Complexes have a potential ability to provide Fe and other micronutrients to plants, but at the present
stage of knowledge the aspects that determine their effectiveness (Fe speciation, complexing sites, pH,
molecular size, etc.) remain unclear and very variable results are obtained. 57Fe Mössbauer spectroscopy is
a powerful tool to elucidate Fe speciation and Fe environment, and it can be very useful to understand Fe
complexes behaviour. Thus, the main aims of this project are: i) to study Fe environment and speciation in
natural complexes under different experimental conditions; ii) to elucidate Fe chelates and complexes
behaviour (sorption, Fe splitting and desorption processes) after interaction with soils and soil materials;
iii) to evaluate the species formed during Fe uptake by efficient and sensitive Strategy I plants, and
determine if Fe complexes can be used as substrate for Fe3+ reduction by the root plasma ferric membrane
reductase enzyme.

A pályázat keretében a spanyol Universidad Autónoma de Madrid egyetemmel együttműködésben vizsgáltuk azokat a szempontokat, melyek meghatározzák a természetes eredetű vaskomplexek vasellátásban mutatott hatékonyságát. A munka során analitikai (Mössbauer-spektroszkópia, FTIR, HPLC, ICP-MS, AAS, XRD) és biológiai (vas-kelát reduktáz aktivitás, klorofill fluoreszcencia, fluoreszcencia leképezés, PAGE) módszereket alkalmaztunk. 6 lignoszulfonát, Na-glukonát és leonardit esetén, különböző előállítási körülmények (pH, Fe:ligandum arány) mellett vizsgáltuk a vaskomplexek szerkezetét és a vas speciációját. Vizsgáltuk továbbá a felsorolt vaskomplexek stabilitását meszes talajon. A természetes vaskomplexekre kapott eredményeket összehasonlítottuk az egyik legstabilabb szintetikus vaskelát, a Fe3+-o,o-EDDHA esetében kapott eredményekkel. A felsorolt vaskomplexek és 4 vaskelát esetében két tesztnövény (uborka, szója) segítségével mértük a vas-kelát reduktáz aktivitást és a vasfelvételt. A vaskomplexek hatékonyságát vizsgáltuk vízkultúrán nevelt növényeken, mind a tápoldaton keresztül, mind a leveleken keresztül történő vasellátás esetén. A tanulmányozott vaskomplexek hatékonynak bizonyultak a kloroplasztiszok vastartalmának, illetve a fotoszintetikus rendszer működésének helyreállításában. Az eredményeket referált tudományos folyóiratokban és nemzetközi konferenciákon közöltük és közöljük. A tudományos kooperáció igen sikeres volt a hazai kutatók külföldi tapasztalatszerzése terén is.
Results in English
In the framework of the project, the aspects that determine the effectiveness of natural complexes having potential ability to provide Fe to plants, were studied with analytical (Mössbauer spectroscopy, FTIR, HPLC, ICP-MS, AAS, XRD) and biological (FCR activity tests, chlorophyll fluorescence, fluorescence imaging, PAGE) methods in cooperation with the Universidad Autónoma de Madrid, Madrid, Spain. We have investigated the Fe speciation in Fe complexes prepared with 6 different lignosulfonates, Na-gluconate and leonardite at different pH and Fe:ligand ratios. The stability of the Fe complexes under calcareous soil conditions and the interaction with 4 soil materials were studied. The results were compared with those obtained for one of the most stable synthetic Fe chelate, Fe3+-o,o-EDDHA. Ferric reductase activity with these Fe complexes and with four synthetic Fe chelates was measured together with Fe uptake using two model plants: cucumber (efficient plant) and soybean (inefficient plant). The ability of Fe complexes to overcome iron chlorosis was tested in both plants when the complexes were applied in nutrient solution and through foliar sprays. The Fe complexes were found to be effective in restoring chloroplast iron content and the photosynthetic apparatus. The results have been and being published at international conferences and in SCI journals. The scientific cooperation has been very successful regarding the experience with the others’ methodologies, too.
