A dUTPáz a dUTP pirofoszforolízisét katalizálja, és ezzel szabályozza a sejtekben az uracil DNS-be való beépülésének mértékét. A DNS-be épülő nagy mennyiségű uracil következménye sejthalál is lehet. Ezért nagy tudományos érdeklődésre tart számot a dUTPáz gátlásán alapuló új rákellenes, antivirális és antituberkulózis gyógymódok kutatása. Jelen pályázat fókuszában a Mycobacterium tuberculosis (MTB) dUTPáz áll, amelynek fiziológiás működése feltehetően élet-halál kérdés a baktérium számára a dTTP szintézisben betöltött központi szerepe miatt. A pályázat fő célkitűzései 1) a Mycobacterium túlélésének vizsgálata a dUTPáz működésének hiányában, 2) az MTB dUTPáz enzimatikus mechanizmusának felderítése különös tekintettel a humán (gazda) és MTB (patogén) enzimek közötti különbségekre, 3) az MTB és humán dUTPázokat leginkább megkülönböztető két szerkezeti elem, a C-terminális kar és a központi csatorna, katalízisben betöltött szerepének meghatározása. A fenti célok megvalósításához számos gyorskinetikai valamint egyensúlyi enzimológiai és spektroszkópiai módszert alkalmazunk vad típusú és helyspecifikusan módosított MTB dUTPáz enzimeken. A dUTPáz funkcióvesztés (feltehetően letális) hatását irányított géncserével történő génkiütéssel egy nem-patogén Mycobacterium modellben vizsgáljuk. A tervezett kísérletek egyértelműen felmérik az MTB dUTPáz, mint antituberkulózis célpont jelentőségét egy potenciális újgenerációs gyógymódban, és jelentősen hozzájárulnak a dUTPázok alapkutatásához.
Summary
dUTPase is the unique enzyme that catalyses the pyrophosphorolysis of dUTP, thus regulating the extent of uracil incorporation into DNA. Massive uracil incorporation may lead to cell death, dUTPase has therefore been recognized as a high-potential drug target in cancer, viral and bacterial disease control. The present proposal focuses on the Mycobacterium tuberculosis (MTB) dUTPase that also plays a central role in the mycobacterial dTTP biosynthesis and thus may be essential for the viability of MTB. The proposal has three specific aims all directed towards the evaluation of MTB dUTPase as a valid tool in fighting tuberculosis: 1) study the effect of dUTPase functional ablation on the viability of Mycobacterium, 2) elucidation of the enzymatic mechanism of MTB dUTPase with regards to the mechanistic differences between the human (host) and MTB (pathogen) dUTPases, 3) determine the catalytic role of the two structural elements that may be species-specifically targeted in dUTPase. To address the above issues, several transient kinetics and equilibrium enzymology as well as spectroscopy methods will be employed using wild-type and mutant MTB dUTPase enzymes. The physiological effect of dUTPase functional ablation will be investigated in a non-pathogenic Mycobacterium model subjected to dUTPase gene replacement. Besides the utility of this study in tuberculosis research, the expected result will also significantly contribute to the field of dUTPase enzymology.
