Az obligát intracelluláris baktérium, a Chlamydia trachomatis a szexuálisan terjedő fertőző betegségek és a fertőzéses eredetű vakság fő okozója. A gyakran tünetnélküli nőkben a krónikus fertőzés infertilitáshoz vezethet. A Chlamydia pneumoniae gyakori okozója a légúti megbetegedéseknek. A fertőzés itt is gyakran válik krónikussá, amely növeli az atheroszklerózis és az asztma kialakulásának kockázatát.
A ChlamyTrans egy újonnan alakult kezdeményezés amely négy kutatóhely és két üzleti partner által jött létre, egyesítve a tapasztalatokat a transzkriptomika és pathogenomika területén. A ChlamyTrans célja olyan diagnosztikus kit kifejlesztése, amely a krónikus chlamydia fertőzést képes kimutatni , és olyan peptidmolekulák tervezése amellyel a krónikus fertőzéses állapotot kezelni lehet. A partnerek olyan módszereket alkalmaznak mint a nagy-áteresztőképességű genom szintű transzkriptomika és interakciós vizsgálatok a későbbi diagnosztikus és terápiás chlamydia targetek kiválasztására. Az üzleti partnerek gyártó és marketingtapasztalatokkal segítik a fennt említett célok megvalósítását.
Summary
The obligate intracellular bacterium Chlamydia trachomatis is the leading cause for sexually transmit-
ted diseases and preventable infectious blindness. In asymptomatic females establishment of chronic
disease may result in infertility. Urogenital chlamydial disease is the main cause for female infertility.
C. pneumoniae is a frequent cause of acute respiratory infection. It has a strong propensity to produce chronic infection, which may predispose to arteriosclerosis and asthma.
ChlamyTrans is a newly formed consortium of two commercial partners (biotech; assay manufacture)
and four biomedical research groups including a clinical partner that will integrate their expertise in
transcriptome-based techniques and pathogenomics of chronic chlamydial disease. ChlamyTrans is
devoted to produce commercially exploitable diagnostic kits for detecting chronic chlamydial infection,
susceptibility to it, and to design therapeutic small inhibitory peptides to combat it. The joint effort al-
lows use of a novel strategy of high throughput transcriptomics for parallel analysis of the complete
open reading frames (ORFs) of C. trachomatis and C. pneumoniae as the core technique. The pro-
gress of ChlamyTrans is closely attended by an unfunded commercial partner with longstanding ex-
perience in producing and marketing diagnostic assays for both pathogens.

A ChlamyTrans project keretében sikerült kidolgoznunk egy új típusú, PCR amplifikáción alapuló Chlamydiális mRNS amplifikációs módszert. A módszer szenzitivitását és specificitását részletesen teszteltük QPCR módszerrel. Az amplifikációs módszert Chlamydia pneumoniae-val fertőzött egértüdőben lezajló bakteriális génexpresszió monitorozására használtuk fel. Több olyan gént-géncsoportot azonosítottunk, amelyek in vivo a baktérium aktív vagy perzisztens növekedésében szerepet játszhatnak. A bakteriális génexpresszió kutatása mellett a Chlamydia fertőzés hatására bekövetkezett gazdasejti génexpressziót is monitoroztuk. Olyan géneket azonosítottunk, amelyek szerepet játszhatnak a Chlamydia fertőzések által indukált szöveti pathológia kialakításában. Végezetül olyan módszerfejlesztésekben is részt vettünk, amelyekkel a Chlamydia fajok funkcionális genetikáját lehet részletesen tanulmányozni.
Results in English
In the framework of ChlamyTrans project, we developed a novel PCR-based Chlamydial mRNA amplification method. The sensitivity and specificity of the method was extensively tested using the QPCR method. The amplification was used to monitor Chlamydial gene expression in Chlamydia pneumoniae infected mouse lung tissue. The gene expression screen identified Chlamydial genes that were involved in the active and persistent growth of C. pneumoniae in vivo. Besides Chlamydial gene expression profiling we monitored the host gene expression during Chlamydia infection. We could identify host genes that could potentially participate in Chlamydia-indued tissue pathology. Finally, we developed two methods with which we can characterize the functional genetics of Chlamydiae.
