A TPPP/p25 (Tubulin Polymerization Promoting Protein) egy nemrég felfedezett, rendezetlen fehérje, ami a mikrotubuláris rendszer dinamikáját és stabilitását befolyásolja. Multinukleáris NMR mérésekkel a közelmúltban igazoltuk, hogy a rendezetlen régiók a TPPP/p25 fehérje N- és C-terminális részein egy flexibilis régiót fognak közre; továbbá a fehérje köti és specifikusan hidrolizálja a GTP-t. Mivel a TPPP/p25 teljes asszignációja sikertelen volt, ezért mutáns, N- és/vagy C-terminális mentes csonkolt fehérjéket, továbbá a TPPP/p25 homológjait tervezzük használni a következő ligandok/partnerek kötőmotívumainak feltérképezésére: GTP és egyéb nukleotidok; kétértékű kationok, mint a fiziológiásan releváns Zn2+; valamint a fiziológiásan vagy patológiásan kölcsönható fehérjék. Néhány kötőmotívumot, mint a GTP, NAD+, Zn2+ és a tubulin esetében, szekvenciahomológia alapján jósultunk. Továbbá előzetes adataink alapján a mikrotubulusok dinamikáját és stabilitását befolyásoló tubulinacetilációt a TPPP/p25 jelentősen növeli, valószínűleg a deacetiláz enzimek gátlása révén. A TPPP/p25 kölcsönhat az alfa-synucleinnel és felhalmozódik a szinukleinopátiákra jellemző zárványtestekben, azonban nemrégen kimutattuk a fehérje és a beta-amyloid kölcsönhatását is. Ezért a pályázatban célul tűztük ki i) a TPPP/p25 ligandumok és makromolekulák kötődéséért felelős kötőmotívumainak azonosítását; ii) a TPPP/p25 és a tubulindeacetilációban szerepet játszó, lehetséges fiziológiás partnerei (HDAC6 and SIRT2) közötti kölcsönhatás jellemzését; iii) a TPPP/p25 és a patológiás zárványtestek kialakulásában részt vevő, lehetséges patológiás partnerei (alfa-synuclein és beta-amyloid) közötti kölcsönhatás jellemzését. Céljaink eléréséhez in vitro és in vivo szerkezeti és funkcionális vizsgálatokat végzünk izolált fehérjékkel/peptidekkel, valamint endogén TPPP/p25-öt, tubulindeacetilázokat, alfa-synucleint vagy amyloid precursor proteint expresszáló sejtvonalakon, pl. oligodendrocitákon, CG-4, HeLA, neuroblasztóma és indukálható CHO sejteken.
A heteroasszociációkat cirkuláris dichroizmus, fluoreszcencia, felületi plazmon rezonancia, ELISA és izotermális kalorimetria segítségével jellemezzük; néhány esetben multinukleáris NMR spektroszkópiával kiegészítve. A kölcsönhatások funkcionális következményeit TPPP/p25 indukált tubulinpolimerizációval és GTPáz aktivitásméréssel vizsgáljuk. Sejt szinten ún. egy-sejtes kísérletekben tanulmányozzuk a kölcsönhatásokat, a fiziológiás és patológiás partnerek és a TPPP/p25 együttes expressziójának hatását vizsgálva a tubulinacetiláció mértékére, a mikrotubuláris rendszer dinamikájára és stabilitására, a sejtek morfológiájára, életképességére, a mitokondriális membrán polarizációjára, az oligodendrociták differenciációjára, a fehérjeaggregációra, aggreszómaképződésre fluoreszcens/konfokális mikroszkópiával.
A kutatások eredménye értékes új információt szolgáltat a TPPP/p25 kölcsönhatásainak szerepéről a speciális funkciójú mikrotubuláris struktúrák/ultrastruktúrák kialakulásában, segít a fiziológiásan és patológiásan releváns komplexek kontaktfelszínének azonosításában, így új utat nyit specifikus gyógyszermolekulák tervezésére különböző neurológiai betegségek kezelésében. Ezért a pályázat sikeres teljesítése értékes alapkutatási adatokat szolgáltat, emellett azonban innovatív jelentőséggel is bír.

Összefoglaló laikusok számára

A Sejtarchitektúra kutatócsoport kutatási területe az eukarióta sejtváz egyik fő komponensének, a mikrotubuláris rendszernek és az azzal összefüggő dinamikus, ultrastruktúrális és funkcionális hatásainak az atomi, molekuláris- és sejtszintű vizsgálata. Bizonyították, hogy egy általuk izolált és azonosított fehérjének, a Tubulin Polymerization Promoting Proteinnek (TPPP/p25), amely szerkezet nélküli fehérje, jelentős szerepe van a mikrotubuláris rendszer dinamikájának és stabilitásának szabályozásában. A fehérje aktív részvételét bizonyos idegrendszeri rendellenségek kialakulásában igazolták, így pl. a Parkinson-kór esetén a zárványtestek kialakulásában, illetve a szklerózis multiplexben, amely a mielinhüvely károsodásának következménye.
A jelen pályázat fő célkitűzései: i) a TPPP/p25 ligandumok és makromolekulák kötődéséért felelős kötőmotívumainak azonosítása; ii) a TPPP/p25 és a tubulindeacetilációban szerepet játszó, lehetséges fiziológiás partnerei (HDAC6 and SIRT2) közötti kölcsönhatás jellemzése; iii) a TPPP/p25 és a patológiás zárványtestek kialakulásában részt vevő, lehetséges patológiás partnerei (alfa-synuclein és beta-amyloid) közötti kölcsönhatás jellemzése.

Tubulin Polymerization Promoting Protein (TPPP/p25) is a recently discovered disordered protein which modulates the dynamics and stability of the microtubule system. Our recent multinuclear NMR analysis provided the first evidence that extended unstructured segments of TPPP/p25 are localized at the N- and C-terminals straddling a flexible region; more importantly, TPPP/p25 can bind and specifically hydrolyses the GTP. Since the assignation of the major part of this protein was unsuccessful, we plan to use site-directed mutants, N- and/or C-terminal free truncated forms and homologues of TPPP/p25 for mapping the binding motifs of GTP and other nucleotides, of bivalent cations such as the physiologically relevant Zn2+, as well as that of PROTEINS involved in physiological OR pathological interactions. Some of the binding motifs have been predicted by sequence alignment such as those of GTP, NAD+, Zn2+ and tubulin. In addition, our preliminary or recent data suggested that the tubulin acetylation, which plays an important role in the modulation of the dynamics and stability of the microtubule system, can be effectively enhanced by TPPP/p25 likely via its inhibition of the activities of deacetylases. TPPP/p25 has been found to be interacts with alpha-synuclein and enriched in inclusions characteristic for synucleinopathies so far, however, its interactions with beta-amyloid has been suggested as well. Thus our objectives are i) to identify the binding motifs of TPPP/p25 involved in associations with ligands and macromolecules; ii) to characterize the interactions of TPPP/p25 with potential physiological partners involved in tubulin deacetylation (HDAC6 and SIRT2); iii) to characterize the interactions of TPPP/p25 with potential partners involved in the formations of pathological inclusions (alpha-synuclein and beta-amyloid). To achieve these objectives in vitro and in vivo structural and functional studies will be performed with isolated proteins/peptides and in cell lines expressing endogenously TPPP/p25, tubulin deacetylases, alpha-synuclein or amyloid precursor protein such as oligodendrocyte CG-4, HeLa, neuroblastoma and inducible CHO cell lines.
The heteroassociations will be characterized by circular dichroism, fluorescence spectroscopies, surface plasmon resonance, ELISA and isothermal calorimetry; in some cases theses studies will be complemented by multinuclear NMR analysis. The functional consequences of these interactions will be tested by TPPP/p25-induced tubulin polymerization and GTPase activity assays. At cell level the interactions will be studied by “single cell experiments” monitoring the effect of co-expression of the physiological and pathological partners on the level of tubulin acetylation, the dynamics and stability of microtubule system, cell morphology, viability, polarization of mitochondrial membrane, differentiation/myelinization of oligodendocytes, protein aggregations/aggresome formation by fluorescent/confocal microscopy.
The results of these studies will provide new, valuable information on the role of TPPP/p25-derived interactions in the formation of microtubular structures/ultrastructures coupled with specific functions, and assist to identify the contact surfaces of the complexes relevant at physiological and pathological circumstances, thus open a new way to design drugs with high specificity for treatment of neurological disorders. Therefore, the successful evaluation of this project will provide valuable data not only for basic research, but it has innovative aspects/relevance as well.

A 2016-os év végén lezárult projekt tudományos és költségvetési tételei a vállaltaknak megfelelően teljesültek, melynek eredménye 19 tudományos publikáció és 17 nemzetközi konferencián való részvétel. Egy új innovatív stratégiát dolgoztunk ki olyan többfunkciós fehérjék (Neomorphic Moonlighting Protein) patológiás kölcsönhatásainak szelektív befolyásolására, melyek fiziológiás és patológiás funkcióval rendelkeznek, amit a kölcsönható partnerek határoznak meg. Specifikus gyógyszercélpont abban az esetben azonosítható, amennyiben az adott célfehérje (hallmark) kötőszegmense és/vagy konformációja eltérő a két különböző partnerrel alkotott komplexben. A stratégia molekuláris és sejtszintű alkalmazhatóságát, specifikus gyógyszercélpont azonosítását a Parkinson-kórra (PD) jellemző eset-tanulmányban bizonyítottuk. Korábbi munkáinkból ismert volt, hogy a TPPP/p25 marker fehérjéje a PD-nek és más szinukleinopátiáknak; az alfa-szinukleinnel (SYN) együtt halmozódik fel és ko-lokalizál agyi zárványtestekben. A PD kialakulásában a két rendezetlen fehérje kölcsönhatása fatális; a patológiás aggregátumuk felhalmozódása a betegség későbbi szakaszában jellemző kórképként jelenik meg. Megmutattuk, hogy a patológiás TPPP/p25-SYN komplex kialakulásában a TPPP/p25 147-156, míg a tubulinnal alkotott komplexében a 178-187 szegmensek vesznek részt. A patológiás TPPP/p25 komplex kötőszegmensének validálása, mint potenciális gyógyszercélpont, molekuláris és sejtszinten megtörtént.

Scientific achievement and the budget of the project terminated at the end of 2016 correspond to that undertook resulting in 19 scientific publications and invited and/or supported speaker at 17 international conferences. A new innovative strategy has been developed for selective destruction of pathological interactions of multifunctional proteins (Neomorphic Moonlighting Protein) that are involved in both physiological and pathological functions determined by the interacting partners. Consequently specific drug target can be exclusively validated when the binding domains/conformations of the target protein are distinct in the physiological and pathological complexes. The applicability of the strategy at molecular and cellular levels for validation of drug target was established in a case-study characteristic for Parkinson's disease (PD). Previously we had identified TPPP/p25 as a hallmark of PD and other synucleinopathies; it is enriched and co-localized with alpha-synuclein (SYN) in brain inclusions. In the etiology of PD, the heteroassociation of the two disordered proteins is fatal; the accumulation of protein aggregates is proceeded at the later state of the disease as characteristic clinical symptom. We established that the 147-156 and 178-187 aa segments of TPPP/p25 are involved in complex formation with SYN and tubulin, respectively. The interface of the pathological complex, that is distinct from the physiological one, was validated as a specific drug target.
