Az agyi véráramlás (CBF) állandó szinten tartását a vérnyomás nagyobb kilengései ellenére autóregulációnak nevezzük. Eddig ezt a nyomás-érzékeny miogén mechanizmussal magyarázták. Kísérletes kutatások azonban kimutatták, hogy az agyi erek nem húzódnak össze jelentősen az intraluminális nyomásfokozódás hatására. Jelen pályázatunkban kimutattuk egy másik mechanoszenzitive mechanizmus jelelétét ami intraluminális áramlásnövekedést érzékeli és hatására jelentős összehúzódást hoz létre patkányból izolált arteria cerebri mediá-n (200 µm) és annak mellékágain (50 – 70 µm), valamint humán agyi ereken. Kiderítettük a molekuláris jelátviteli mechanizmusokat és a kórfolyamatok lépéseit, éspedig az arachidonsav (AS) citokróm (CYP) P 450 metabolitjainak, az intracelluláris Ca2+ szintnek és a ROS-nak a szerepét az agyi erek áramlás-indukálta válaszainak a mediálásában. Szintén mind az élettani kórélettani és molekuláris mechanizmusokat leírtuk hiperóniában, strokeban és idődödéssel ill azok hatását a mentalis funkciókra. Ezen állatkísérletek eredményei nem csak fontos elméleti szempontból fontosak, de klinikai problémák megoldásához is hozzásegítenek. Az összegyűlt ismeretek birtokában új terápiás eljárásokat lehet kifejleszteni az agyi vaszkuláris betegségek kezelése és megelőzése céljából.

Keeping the cerebral blood flow (CBF) at a constant level despite the higher fluctuations in blood pressure is called auto-regulation. So far, this has been explained by the pressure-sensitive myogenic mechanism. However, experimental studies have shown that cerebral vessels are not significantly constricted by intraluminal pressure increase. In our present application, we have demonstrated the presence of another mechanosensitive mechanism that detects intraluminal flow increase and causes a significant constriction of the rat cerebral artery (200 µm) and its adrenal glands (50-70 µm) and human brain vessels. We have discovered molecular signaling mechanisms and stheir role in disease processes, namely the role of the arachidonic acid (AS) cytochrome (CYP) P450 metabolites, intracellular Ca2+ levels, and ROS in mediating cerebral vascular flow-induced responses. We have also described physiological pathophysiology and molecular mechanisms in hypertension, stroke and aging, and their effects on mental function. The results of these animal experiments are not only important from theoretical point of view, but also they help to solve clinical problems. With the knowledge gained, new therapeutic methods can be developed for the treatment and prevention of cerebral vascular diseases.
