A kutatás összefoglalója, célkitűzései szakemberek számára
Itt írja le a kutatás fő célkitűzéseit a témában jártas szakember számára.

F--kötő Al(III)- és I--kötő Tl(III)- kétfunkciós ligandum (L) alkotta vegyeskomplexek előállítását, kémiai jellemzését tervezzük. Potenciálisan az AlLF- 18F PET, míg a TlLI-vegyeskomplex 123I- SPECT képalkotásra vagy 131I- izotóp beépítésével célzott β--radioterápiára alkalmas, azaz teragnosztikus készítmény. A gyors reakcióban képződő komplexekben a halogenid nem C-X kovalens kötéssel kapcsolódik és kinetikailag inert. A stabilitás és az inertség a ligandum méretének, oldalláncainak, a szubsztituens vagy a konjugációra alkalmas csoport változatásával befolyásolható.
A nagy stabilitású 68Ga(III) and 111In(III)-fémkomplexek jól ismertek a PET ill. SPECT képalkotásban. Új, célzottan szervezetbe juttatható vegyületként AAZTA és OBETA bifunkciós származékok komplexeit fogjuk vizsgálni 113In- és 69Ga-NMR segítségével mérve a szabad In(III) és Ga(III) vagy Ga(OH)4- részecskék koncentrációját, mert a pH-potenciometria nem alkalmas a nagy stabilitási állandók mérésére. Hasonló nehézségeket EPR-rel hidalunk át olyan Cu(II) komplexeknél, amelyek potenciális 64Cu PET ágensek.
A leginkább ígéretesnek tűnő komplexeket együttműködő partnereinkkel 18F és 123I illetve 68Ga-isotópokkal is előállítjuk és vizsgáljuk. Feltétlenül kísérletet teszünk a kiváló molekuláris sugárterápiás paraméterekkel rendelkező, α-sugárzó 212At-izotóp beépítésére is. A kiválasztásnál a “minél nagyobb stabilitás, annál jobb in vivo viselkedés” optimista elvét alkalmazzuk.
A kémiai és radiokémiai eredmények szolgálhatnak távlatilag a direkt orvosbiológiai kísérletek alapjául, ami majd egy újabb pályázat keretében sejt-biológiai és kisállat PET kutatást foglalhat magába.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

McBride ötlete, azaz hogy a fluoridot Al(III) centrumhoz kötik, több előnnyel jár. Szemben a kovalens C-F kötés képződésével pl. a 18FGD-ben (18-fluor-deoxiglükóz), a 18F izotóp ionos kötéssel, gyors reakcióban kötődik az Al-bifunkciós ligandum komplexhez. Az ilyen komplex célzottan használható radiodiagnosztikum lehet.
A munkahipotézisünk szerint a háromkomponensű rendszer egy vegyesligandumú komplexet alkot. A fluorid disszociációjának elmaradását a komplex inertségének tudhatjuk be, mivel a becsülhető termodinamikai stabilitás messze alatta marad annak, ami az igen híg oldatokban megakadályozhatná a halogenid ion kilépését. Az inert vegyeskomplex képződésen alapuló elképzelést kiterjesztjük a Tl(III)-I- rendszerre is. Az At-analóg előállítását is megkíséreljük, ami nagyon nehéz feladat, de a siker valóban kiugró eredmény lenne. Az inertség oka(i) nem könnyen magyarázható(ak), rendszeres egyensúlyi, kinetikai és szerkezeti vizsgálatok tisztázhatják ezeket és járulhatnak hozzá a ligandumtervezés sikeréhez, a készítmények optimális előállításának/formulázásának a megtalálásához.
A 68Ga (és feltehetően a 64Cu) szerephez juthat néhány éven belül mint rutinszerűen alkalmazott klinikai PET izotóp. A most még jórészt hiányzó megbízható kémiai stabilitási és kinetikai adatok segíthetik a legjobb bifunkciós ligandumok kiválasztását a célzottan alkalmazható radiodiagnosztikumok előállításához. A ciklikus AAZTA nagyon ígéretes ligandum, nagyon stabilis fémkomplexeket képes gyors reakciókban, és jól konjugálható bio-vektorokkal a szelektív célbajuttás érdekében.
A radiokémiai vizsgálatokat a legjobbnak vélt, kiválasztott komplexekkel tervezzük elvégezni.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

A Pozitron Emissziós Tomográfia érzékenysége, szelektivitása és pontossága jobb, mint a SPECT vagy CT módszereknek. Emiatt terjedt el a klinikai gyakorlatban tumorok kimutatására, a betegség stádiumának megállapítására és a terápia követésére egyaránt. Növekvő igény van célzott PET, teragnosztikus vagy bimodális készítményekre, emiatt aztán a kutatások a kémia és a képalkotási technikák terén egyaránt intenzívebbé váltak. A 68Ga izotóp (amit generátorból nyernek) lesz vélhetően a második széleskörűen elterjedt PET izotóp a jelenleg egyeduralkodó 18F mellett.
Az (18FAl)-alapú PET diagnosztikumok a 2009-es megjelenés óta jelentős tudományos érdeklődést keltettek, noha a fizikai-kémiai jellemzésük mindmáig várat magára. Az általunk tervezett módszeres kísérletek és DFT számolások segíthetnek tisztázni az alapokat, megtalálni a vegyeskomplex szerkezete és a funkció közötti kapcsolatot. A nagyon híg oldatokban mért kinetikai adatok remélhetően a radiokémikusok és a fiziko-kémikusok érdeklődését is felkeltik.
Ha munkahipotézisünk igazolást nyer, akkor az ötlet alkalmazható a Tl(III)-jodid rendszerre is, és kifejleszthető lesz egy új, különböző jód izotópokon alapuló teragnosztikus vegyületcsalád. A 123I/131I izotóp pár igen alkalmasnak látszik erre, mert egy 123I-al jelölt anyag érzékeny és olcsó SPECT diagnosztikum, míg ugyanaz a molekula (pontosabban annak izotopomerje) 131I jelöléssel alkalmas lehet célzott molekuláris radioterápiára. A remélt 211At-jelölt komplex pedig kiváló α-sugárzó molekulársi sugárterápiás készítmény lehet.
A DTPA analóg gyűrűs AAZTA sokféle célra alkalmas, könnyen konjugálható ligandum. Ga(III) és In(III) komplexeinek részletes vizsgálata vezethet el ezek preklinikai vizsgálataihoz.
A kompetíciós módszerrel mért, remélhetően megbízható Ga(III), In(III) és Cu(II) stabilitási állandók érdeklődésre tarthatnak számot a képalkotással foglalkozó kutatók és a koordinációs kémikusok között is.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

Az orvosi diagnosztika egyre fontosabb eszköze a képalkotás. Ehhez gyakran különböző vegyületek kellenek vagy magának a képnek a létrehozásához, vagy a kép minőségének, a kontrasztosságnak a javításához. A kémiai kutatások fontos szerepet játszanak az ilyen anyagok előállításában és jellemzésében, mivel az élőszervezetbe bejuttatott anyagoknak egy sor kívánalmat ki kell elégíteni: nem lehet mérgező, hatékony kell legyen kis dózisban is, teljesen ki kell ürüljön a testből, ne legyen drága stb. Ha a készítmény radioaktív, mint pl. a Pozitron Emissziós Tomográfiában (PET) alkalmazott szerek, akkor radiokémikus munkájára is szükség van. Molekuláris biológus szakember kell a sejt-kultúrák vizsgálatához, vagy az állatkísérletekhez pl. a PET vizsgálatoknál.
A mi kémikusokból álló kutatócsoportunk tumorok kimutatására alkalmas PET 18F- radiodiagnosztikumot fejleszt. Célunk olyan egyszerű és gyors szintézis út megtalálása, amiben a 18F izotóp alumínium tartalmú molekulához kötődik. Ebben a fluorid ionos kötéssel kapcsolódik. A molekula szerves anyagot, egy olyan kétfunkciós ligandumot is tartalmaz, ami egyrészt erősen megköti az Al18F egységet, másrészt a molekulát képes a vektor részével (egy csatolt biomolekulával, pl. peptiddel) odavinni a daganathoz. Tervezzük hasonló, de jodid-kötő molekulák kifejlesztését is. Ha alkalmasan kiválasztott különböző rádioaktív jód-izotópokat használunk, akkor un. teragnosztikus készítményhez jutunk. A teragnosztika a terápia és a diagnosztika összevonását jelenti, és fontos eszköz az individuális gyógymódok kidolgozásában.

Summary of the research and its aims for experts
Describe the major aims of the research for experts.

F--binder Al(III)- and I--binder Tl(III)- bifuntional organic ligand mixed complexes as potential 18F PET radiopharmaceuticals and theragnostic agents (SPECT imaging with 123I and targeted β--radiotherapy with 131I) will be prepared and chemically characterised. These compounds bind halide ions with non-covalent bond in fast reactions, and they can be kinetically very inert. The stability and inertness might be tuned by the size and/or by different pendant arms, anchors and substituents of the ligand.
68Ga(III) and 111In(III)-complexes play important role in PET and SPECT, respectively. As new compounds - suitable for targeting- complexes formed with bifunctional AAZTA and OBETA derivatives will be studied by 113In- and 69Ga-NMR measuring the free In(III) and Ga(III) or Ga(OH)4-, because pH potentiometry can not be used to determine the high stability constants. Similar difficulties occur for Cu(II) complexes (as potential 64Cu PET tracers) will be solved by EPR spectroscopy. The slow complexation reactions will be investigated by microwave activation.
Together with our partners radiochemical studies will be carried out by using 18F and 123I and 68Ga-isotopes to prepare selected complexes. We do want to make radioactive labeling with 212At-isotope having outstanding good parameters for molecular radiotheraphy. The higher stability (and/or the higher inertness) the better for in vivo test.
Results of chemical and radiochemical study may initiate further direct bio-medical project in a forthcoming application: cell biology and PET imaging studies will include receptor binding assay, small animal tumour model, MiniPET imaging and biodistribution experiments.

What is the major research question?
Describe here briefly the problem to be solved by the research, the starting hypothesis, and the questions addressed by the experiments.

The McBride-concept, i.e. binding fluoride to Al(III) provides several advantages. Instead of covalent C-F bond constructed for example in 18FDG (18-fluorine-deoxyglucose) 18F is bonded by ionic interaction in fast reaction to Al-bifunctional ligand conjugate. The complex can act as a targeted radiopharmaceutical agent.
Our hypothesis considers the three component adduct as a mixed ligand complex, and the luck of fluoride dissociation can be attributed to the kinetic inertness of the complex, because the estimated thermodynamic stability of the complex far not enough high to avoid dissociation in diluted solution. This concept can be extended to Tl(III)-I- system. We will try to make the At-analogue, it is surely ”high risk” project, but a positive result would be „ground breaking result”. There is no obvious explanation for the inertness, systematic research on equilibrium, kinetics and structure is needed to optimise the ligand design and the preparation/formulation methodology.
68Ga (and probable 64Cu) are among PET isotopes expected to enter the clinical practice in a few years. Reliable equilibrium and kinetic data can help to select the best bifunctional ligands for targeted imaging. The cyclic AAZTA seems to be very useful ligand, it forms very stable metal-complexes in fast reaction, and it can be conjugated with bio-vectors for targeting.
Radiochemistry studies are planned to control the stability of the most promising new complexes.

What is the significance of the research?
Describe the new perspectives opened by the results achieved, including the scientific basics of potential societal applications. Please describe the unique strengths of your proposal in comparison to your domestic and international competitors in the given field.

Pozitron Emission Thomography (PET) is found to be superior over SPECT and CT in sensitivity, specificity and accuracy, and it finds clinical application for detecting, staging and therapy of tumours. The demand for targeted PET and/or theragnostic and/or bimodal agents are growing. Therefore research activity is increasing both in chemistry and imaging. 68Ga isotope (produced by generator) is told to be the next widely used PET-isotopes following the only one registered 18F, so far.
Substantial scientific interest has been generated by the (18FAl)-entity-based PET tracers since 2009, although the physico-chemical characterisation of these new compounds has not been done. Our systematic experiments and DFT calculations can help to understand the new concept and to find relation between structure and properties of the mixed ligand complexes. Kinetic data measured in very diluted samples may have some importance both for radio- and physico-chemists.
If our hypothesis is being valid we can extend the concept to the Tl(III)-iodide system and we can develop a new family of theragnostic agents using different I-isotopes. The 123I/131I pair of isotopes can be considered as good candidate for theragnostic agents, i.e. a 123I-labeled molecule in SPECT could be a sensitive and economic diagnostic drug, whilst the same molecule labelled with 131I (a chemically identical isotopomer) might be used for targeted molecular radiotherapy. 211At-labelled complex could be a new α-radiating agent for targeted molecular radiotherapy.
The DTPA analogue cyclic AAZTA is a versatile ligand and it is suitable for conjugation. Detailed characterisation of Ga(III) and In(III) complexes with AAZTA is a prerequisite before preclinical experiments.
Expected results of measuring very large stability constants using competition methods in Ga(III), In(III) and Cu(II) systems have also meaning both for the imaging community and coordination chemists.

Summary and aims of the research for the public

Imaging is an important and growing field of medical diagnostic. Different molecules are needed either for generation of the image or improving the contrast. Chemical research is very important for synthesis and characterisation of these molecules, because chemicals used in living systems should fulfil several requirements like luck of toxicity, high effectiveness at low dose, good clearance from the body, moderate price etc. Active work of radio-chemist is also needed if the compound is a radiopharmaceutical drug used for example in Pozitron Emission Thomography (PET). Molecular biologists are responsible for tests with cell cultures and small animal experiment including PET imaging.
Our joined team is going to develop new PET radiopharmaceutical drugs based on 18F radioisotope to detect tumours. Our aim is to find simple and fast way of synthesis using aluminium-containing molecules. These molecules can bind fluoride by ionic interaction. The organic part of the molecule can be a bifunctional ligand, which is able to bind the Al18F entity with a chelating group, and can navigate the molecule to tumour with a vector (i.e. biomolecule like a peptide) conjugated to metal-binding part of the compound. We are going to develop analogous molecules for binding iodide. Using different iodine –isotopes one can make radiopharmaceuticals named theragnostic drug. Theragnostics (therapeutics and diagnostics) is a proposed process of diagnostic and therapy for individual patients.

Új mangán(II), réz(II), lantanoida(III), alumínium(III), gallium(IIII) és tallium(III) komplexeket állítottunk elő erre a célra tervezett és szintetizált merevített szerkezetű szerves ligandumokkal. A vegyületek részletes fizikai-kémiai jellemzése azt mutatta, hogy néhány új vegyületünk termodinamikai stabilitása, inertsége és hatékonysága jobb, mint az orvosi képalkotásban (pl. a mágneses rezonancia képalkotásban, MRI; a pozitron emissziós tomográfiában, PET) manapság alkalmazott fémkomplexeké. A ligandumok merevségének finomhangolását a nyílt láncú és a makrociklusos aminopolikarboxilátok szisztematikus módosításával oldottuk meg, ami érintette a vegyületek vázát és a koordinálódó oldalláncokat egyaránt. A komplexek rendelkezésre állnak a további in vivo vizsgálatokra, a Mn(II)- etilén-diamin-tetraacetát-bis(amid) vegyületcsalád szabadalmaztatása megtörtént, további vizsgálata ipari partnereinkkel folytatódik.

We synthesised new manganese(II), copper(II), lanthanide(III), aluminium(III), gallium(III) and thallium(III) complexes with rigidified organic ligands. Detailed physico-chemical characterisation shows that some of the compounds have better thermodynamic stability, inertness and effectiveness compared to metal complexes routinely used in different modalities of medical imaging (e.g. Magnetic Resonance Imaging, Positron Emission Tomography) or theragnostics (combination of diagnosis and therapy). The rigidity of the ligands was tuned by systematic substitution of open chain and macrocyclic aminocarboxylate ligands both in the backbone/macrocycle and at the pendant arms of the molecules. The complexes are ready for further studies in vivo, one family of Mn(II)-ethylene-diamine-tetraacetic-acid-bis(amide) derivatives was patented and earned industrial interest.
