A Huntington kór (HD) egy domináns módon öröklődő, halálos kimenetelű progresszív neurodegeneratív megbetegedés. Kiváltó oka az IT15 génben bekövetkező CAG ismétlődés expanziója, amely meghosszabbodott poliglutamin doménnel rendelkező toxikus huntingtin fehérje termelését eredményezi. A mutáns huntingtin aggregátumokat képez és számos fehérjével lép kóros kölcsönhatásba, amely többrétű pathomechanizmushoz vezet. A génkifejeződésben beálló zavar a HD pathomechanizmusának egyik fő komponense. A mutáns huntingtin gátolja a hiszton acetiltranszferáz enzimeket, és az acetiláció egyensúlyának genetikai vagy farmokológiai eszközökkel történő helyreállítása enyhíti a mutáns huntingtin hatásait. A fehérje acetilációt általánosan befolyásoló terápiás megoldások azonban széleskörű mellékhatásokkal járhatnak. A betegség biológiai hátterének jobb megismerése és specifikusabb terápiák kifejlesztésének előmozdítása céljából tervezzük a hiszton fehérjék azon lizin oldalláncainak azonosítását, amelyek módosítása releváns a HD szempontjából. A projekt fő célja, hogy megállapítsa, kritikus szereppel bírnak-e meghatározott hiszton módosítások a HD kór szempontjából, és hogy ezek hogyan befolyásolják a mutáns huntingtin kiváltotta pathogenezist. Célunk meghatározni, hogy az azonosított lizin oldalláncoknak szerepe van-e a hiszton acetiltranszferáz és deacetiláz enzimek, ill. deacetiláz gátló vegyületeknek a HD-re gyakorolt hatásának közvetítésében. Emellett meg kívánjuk határozni az azonosított hiszton módosításoknak a vad-típusú ill. mutáns huntingtint expresszáló állatok génexpressziós profiljára gyakorolt hatását.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

A kutatási projektben a különböző specifikus hiszton lizin módosításoknak a mutáns huntingtin fehérje által kiáltott pathogenezisben betöltött szerepét fogjuk vizsgálni. Munkánk kiinduló hipotézise az, hogy a Huntington kór modelljeiben megfigyelhető, a sejtek acetilációs egyensúlyában bekövetkező eltolódás elsősorban a hiszton fehérjék megváltozott acetilációs állapotain és az ennek következményeként beálló génexpressziós változásokon keresztül fejti ki hatását. Ezzel kapcsolatban a következő kérdésekre keresünk válaszokat: A hisztonok mely lizin oldalláncainak módosítása bír kiemelt jelentőséggel a Huntinton kórral kapcsolatban? Hogyan befolyásolja a mutáns huntingtin által kiváltott fenotípusokat olyan hiszton mutációk, amelyek mimikálják ezeknek a specifikus lizinek módosított állapotait? Mely hiszton lizin oldalláncok módosítása felelős a hiszton acetiltranszferáz illetve deacetiláz enzimeknek, illetve deacetiláz gátló vegyületeknek a mutáns huntingtin kiváltotta fenotípusokra gyakorolt hatásának közvetítéséért? Milyen hatást gyakorolnak a specifikus hiszton mutánsok a sejtek transzkripciós aktivitására; helyreállítják-e a mutáns huntingtin által kiváltott génexpressziós változásokat?

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

Számos tanulmány támasztja alá, hogy a sejt acetilációs egyensúlyának megváltozása fontos szereppel bír a mutáns Huntingtin fehérje által kiváltott pathomechanizmusok között, és hogy a hiszton deacetiláz inhibitorok ígéretes terápiás szerek lehetnek Huntington kórban. Egy jelentős probléma azonban ezzel a megközelítéssel, hogy az epigenetikai jeleket módosító kezelések széles körűen befolyásolhatják a genom expresszióját, és így nemkívánatos mellékhatásaik lehetnek. E kutatás várhatóan olyan eredményeket szolgáltat, amelyek azonosítják azokat a specifikus lizin oldalláncokat a hisztonokon, amelyek módosítása döntő a Huntington kórban; és így hozzájárulhat specifikusabb, kevesebb mellékhatással járó terápiás megoldások kifejlesztéséhez.

A Huntington kór egy jelenleg gyógyíthatatlan, halálos kimenetelű, öröklődő idegrendszeri megbetegedés, melynek tünetei középkorú felnőttekben jelentkeznek először, és több mint 10 éves leépülés után vezetnek a beteg halálához. A betegséget egy meghatározott fehérjét, a huntingtint kódoló gén mutációja okozza. A betegség biológiai háttere azonban ennek ellenére sokrétű, mivel a mutáns fehérje számos sejtbeli folyamatot megzavar. Ezeknek a folyamatoknak egyike a gének átírása, kifejeződése. Ennek a zavarnak egyik oka a DNS-t, így a géneket is becsomagoló és átírásukat, aktivitásukat alapvetően befolyásoló hiszton fehérjék szabályozó szerepű módosításainak megváltozása. E módosítások egyikéről, az acetilációról már korábban kimutatták/-uk, hogy szerepe fontos a Huntington kórban, és befolyásolásával a betegség tünetei állat modellekben enyhíthetőek. E pályázatban célunk ennek a módosításnak pontosabb megismerése, a betegségben kritikus szerepet játszó szabályozási pontok azonosítása. Ehhez olyan specifikus mutánsokat használunk, melyekben a hiszton célfehérjéknek mindig csak egy – egy pozícióját változtatjuk meg, így azonosíthatjuk azokat a helyeket a hiszton fehérjéken, amelyek módosítása a betegség szempontjából fontos. Reményeink szerint a kutatás során szerzett ismeretek hatékonyabb és specifikusabb terápiák kifejlesztéséhez vezethet.

Huntington's disease (HD) is a dominantly inherited, progressive neurological disorder with fatal consequences. It is caused by an expansion of a CAG trinucleotide repeat in the IT15 gene that results in the production of toxic huntingtin proteins with an expanded polyglutamine domain. Mutant huntingtin forms aggregates and mediates aberrant protein interactions that lead to a multifaceted molecular pathogenesis. Transcriptional dysregulation is one of the major pathological mechanisms of HD. Mutant huntingtin inhibits histone acetyltransferase enzymes and restoration of the acetylation balance by genetic or pharmacological means ameliorates phenotypes caused by mutant huntingtin. Unfortunately, the potential side effects of therapeutic interventions aimed at protein acetylation in general can be widespread. Therefore, we plan the identification of relevant acetylation targets on histone proteins that affect gene expression changes in HD to better understand the biological background of the disease and for aiding the development of more specific therapies. The main goal of this project is to determine whether specific histone modifications have a fundamental influence on HD pathogenesis. We aim to determine whether modification of specific histone lysine residues have a critical role in the disease, and how they affect mutant huntingtin induced pathogenesis. We aim to determine whether these lysine residues are the main targets of histone acetyltransferase and deacetylase enzymes and histone deacetylase compounds. We also aim to determine the effect of these modifications on gene expression patterns in wild-type an huntingtin expressing animals.

What is the major research question?

In this research project we plan to asses the contribution of different specific histone lysine modification to mutant huntingtin induced pathogenesis. Our starting hypothesis is that altered acetylation state of histone proteins and consequent changes in gene expression is the major outcome of the disturbed cellular acetylation balance that can be observed in Huntington's disease models. We are looking for answers for the following questions: Which acetylated histone residues are critical in respect of HD pathogenesis? How do specific modification mimicking mutations alter HD phenotypes? Which lysine residues and modifications are important in the mediation of the effects of histone acetyltransferase and deacetylase mutants and deacetylase inhibitor compounds exerted on HD phenotypes? What are the transcriptional consequences of these modifications, and do they restore the dysregulated transcriptional activities due to mutant huntingtin?

What is the significance of the research?

Several previous studies support that upsetting the cellular acetylation balance bears an important role among pathomechanisms induced by mutant huntingtin protein, and that histone deacetylase inhibitors are promising therapeutic drugs in the treatment of Huntington's disease. A significant problem with this therapeutic approach is, however, that treatments affecting epigenetic marks might have a widespread effect on gene expression patterns and therefore could lead to undesirable side effects. This research project aims to identify those specific lysine residues of histones whose modifications have a critical role in Huntington's disease. By this way it may contribute to the development of more specific epigenetic therapeutic solutions with less side effects.

Huntington's disease is a currently untreatable, devastating inherited degenerative disorder of the nervous system. Its symptoms first appear in middle-age adults and leads ultimately to death after more than 10 years of disease progression. Huntington's disease is caused by the mutation of a single gene encoding the huntingtin protein. However, the biological background of the disease is multilayered, as the mutant huntingtin protein disturbs several cellular processes. One of the cellular mechanism altered in Huntington's disease is the expression and activity of genes. Dysregulated gene activities are partly due to changes in regulatory modification of histones, proteins that package DNA and fundamentally influence gene expression. We and others have previously shown, that acetylation, one form of histone modifications, plays an important role in the pathogenesis of Huntington's disease and by increasing acetylation the toxic effects of mutant huntingtin can be reduced in animal models. In this project, we aim to collect detailed information about this histone modification in a Huntington's disease model to better understand the role it plays in the pathogenesis of the disease. For this, we will generate and use such specific mutants of histones that are altered at a single position of the protein. This way we can identify and characterize those positions on the protein, whose modification is critical in regard of the disease. We hope that the results that will derive from this project will lead to the development of more potent and specific therapeutics.

Projektünk a Huntington kór (HK) nevű, dominánsan öröklődő, végzetes kimenetelű neurodegeneratív betegség epigenetikai hátterének jobb megértését tűzte ki céljául. A HK-t a huntingtin gén funkciónyeréses mutációja váltja ki, és többrétű patogenezis jellemzi. Célunk az volt, hogy azonosítsuk és jellemezzük a kromatint alkotó hiszton fehérjék azon poszt-transzlációs módosításait (PTM), amelyek szerepet játszanak a betegség patomechanizmusában. Ehhez a H3.3A és His4r hiszton fehérjék PTM-ait mimikáló mutánsait expresszáló transzgenikus Drosophilákat hoztunk létre, és azoknak a mutáns Huntingtin (mHtt) által kiváltott fenotípusokra gyakorolt hatását genetikai interakciós tesztekben vizsgáltuk. E módszert használva kimutattuk, hogy a H3.3A lizin (K) 14 aminosavának acetilált állapotát mimikáló mutáns menekítette a mHtt kiváltotta fenotípusokat. Transzkriptomikai elemzéssel arra jutottunk, hogy ezért a jelenségért azonban nem a génexpresszióban beálló változás felelős. Kimutattuk, hogy a His3.3A K27 aminosavának metilációs állapota jelentősen befolyásolja a HK modell fenotípusait, és a UTX K27 specifikus demetilázt terápiás célpontként azonosítottuk. Előállítottuk a His4r hiszton és a Hat1 H4-K5-K12 specifikus citoplazmatikus hiszton acetiltranszferáz null mutációit, és kimutattuk, hogy mindkettő enyhe pozitív hatással van a mHtt kiváltotta patogenezisre. Továbbá, kimutattuk, hogy a polifenolokat tartalmazó zöld tea enyhíti a HK Drosophila modelljében megfigyelhető fenotípusokat.

The goal of this project was to elucidate the epigenetic background of Huntington’s disease (HD), a dominantly inherited, fatal neurodegenerative disorder. HD is caused by a gain of function mutation of the huntingtin gene, and characterized by a multifaceted pathogenesis. We aimed to identify and characterize post-translational modifications (PTM) of histone proteins (the main constituents of chromatin) that are involved in HD pathogenesis. We generated transgenic flies expressing PTM mimic mutations of histones His3.3A and His4r, and used genetic interaction tests to analyze their effect on mutant Huntingtin (mHtt) induced phenotypes. Using this method, we found that acetylation mimic of lysine (K) 14 of H3.3A ameliorated HD phenotypes. Transcriptomic analysis, however, showed that this modification affects mHtt toxicity by other mechanisms than transcriptional regulation. We also showed that the methylation state of K27 of His3.3A influences HD pathogenesis, and identified UTX, a K27 specific demethylase enzyme as a drugable therapeutic target in HD. We generated null mutants of histone His4r and the cytoplasmic histone H4-K5-K12 specific acetyltransferase, Hat1, and showed that reduced His4r and Hat1 levels had a modest positive effect on mHtt induced pathology. Furthermore, we showed that polyphenol containing green tea infusion has ameliorating effects in a Drosophila model of HD.
