A kutatási támogatás első hét hónapjában a kutatási terv alábbi pontjaiban értünk el eredményeket: 1. Biobankunk bővítése, a mitokondriális betegek fenotípusos és biokémiai karakterizálása
2. Teljes exom szekvenálás: a 3 évre kitűzött 20 ismeretlen mitokondriális betegségben szenvedő esetből a 7 hónap alatt a NeurOmics FP7 program keretében a müncheni Helmholtz Zentrumban eddig 5 genetikailag nem azonosított magyar mitokondriális beteg teljes exom szekvenálását végeztük el. Az 5 esetből eddig 3 esetben jutottunk egyértelmű diagnózishoz. mindegyik esetben egy nagyon ritka kórképet azonosítottunk.
A familiáris ismeretlen eredetű myopathiás fenotípust mutató magyar családjainknál a kanadai Care4Rare projekt révén 7 neuromuskularis esetünkből egy esetben WES analízissel egy nagyon ritka myopathiára derült fény, az eset publikációja folyamatban van.
3. A kapott WES adatok kiértékelését, a kapott eltérések konfirmálását és szegregáció-analízisét, az ismert génmutációknál a fenotípus-genotípus korreláció elemzését, hasonló fenotípusú betegek gyűjtését és célzott vizsgálatát, valamint az új mutációknál predikciós szoftveranalízisek alkalmazását intézetünkben végeztük el.
Sajnos a funkcionális (proteomikai és iPSC) vizsgálatokra nem került sor.

In our project we aimed to investigate the complex molecular background of a selected group of Hungarian patients with genetically yet unidentified mitochondrial disease with the approach of next generation methods, whole exome sequencing and proteomic studies.
In the first 7 months of the research support we could reach the following goals of our project:
1. Extending our neuromuscular biobank, phenotypic and biochemical characterization of our mitochondrial cohort
2. WES or targeted NGS sequencing: out of the 20 patients with undiagnosed mitochondrial disease aimed to be characterized in 3 years we were able to perform WES in 5 cases with the help of the NeurOmics FP7 project in the Helmholtz Institute in Munich. Out of the 5 cases we were able to come to a probable molecular genetic diagnosis identifying a rare disease in 3 cases.
In our cohort of Hungarian patients with yet unidentified myopathies a cooperation with the Care4Rare Canadian project yielded the diagnosis of a rare mitochondrial myopathy in one case so far.
3. The analyis of the WES data as well as the cosegregation studies were all performed at our institute. We also analyzed phenotype-genotpye correlations and started to collect patients with similar phenotypes. For new mutations prediction softwares were used successfully.
Unfortunately, due to the termination of the project no functional analyses (e.g. proteomics or iPSC generation) have been performed.
