Az agresszió szabályozásának új mechanizmusát szeretnénk leírni optogenetikai technikák felhasználásával. A kutatás célpontjai azok a projekciók, amelyeket egyes prefrontális neuronok küldenek az agresszív viselkedést szabályozó kéreg alatti központokhoz. Korábban leírtuk, hogy a prelimbikus és infralimbikus kéreg neuronjainak egy populációja efferenseket küld a hipothalamus-nak azon régióiba, amelyek az agresszív támadások kiváltásában kulcsszerepet játszanak. Folyamatban levő kutatásaink során e projekciók lokális (célterületen érvényesülő) optogenetikai ingerlése fokozta a szociális ellenfelek agresszív támadásának gyakoriságát, és drámaian megnövelte a fejre, torokra, és hasra irányuló támadások arányát. Hasonló támadások agresszióval járó pszichikai zavarok laboratóriumi modelljeiben figyelhetők meg. E meglepő eredmények azt mutatják, hogy a prefrontális kéreg idegsejtjeinek egy specifikus populációja működésbe hozza az agresszió kiváltásának hypothalamikus központjait, sőt, abnormális jellegű agressziót vált ki. A vizsgálatokat ki szeretnénk terjeszteni olyan más hypothalamikus és amygdaláris központokra, amelyek afferenseket kapnak a prelimbikus és infralimbikus kéregből, és amelyek kulcsszerepet játszanak mind a fajra jellemző, mind az abnormális agresszióformák szabályozásában. Az optogenetikai ingerlés hatásaival kapott eredményeket a kapcsolatok optogenetikai gátlásának vizsgálatával ellenőrizzük. Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Bár a prefrontális kéreg funkcionális deficitje fokozott agresszivitással jár, a terület aktiválódik az agresszív cselekmények végrehajtása során, ami felveti a kérdést, hogy hogyan kapcsolódhat agresszióhoz ugyanannak a kérgi területnek a gátolt működése és aktiválódása. Korábbi hipotézisek szerint a prefrontális kéreg a kognitív és emocionális működések révén, közvetetten befolyásolja az agresszivitást. Előző, és folyó kutatásaink eredményeiből kiindulva feltételezzük, hogy a prefrontális kéregben olyan idegsejt-populációk találhatók, amelyek közvetlen szerepet játszanak az agressziónak, sőt, az agresszió abnormális formáinak kiváltásában. Hipotézisünk szerint tehát a prefrontális kéreg kettős szerepet játszik az agresszió szabályozásában: egy általános funkcionális szinten a szociális konfliktus kognitív és emocionális komponenseit értékeli, és amennyiben a döntés az agresszió végrehajtása felé mozdul el, ugyanezen kérgi terület idegsejtjeinek egy specifikus populációja működésbe hozza az agresszív cselekmények végrehajtását kiváltó kéreg alatti hálózatokat. Fő célkitűzéseink a következők: (1) azonosítani szeretnénk azokat a prefrontális idegsejt populációkat, amelyek az agresszió szabályozásában kulcsszerepet játszó kéreg alatti területekbe vetítenek; (2) karakterizálni szeretnénk e projekciók szerepét az agresszivitás szabályozásában; (3) azonosítani szeretnénk a projekciós és cél-neuronok neurokémiai sajátosságait, és (4) össze szeretnénk hasonlítani e projekciók specifikus hatásait a prefrontális kéreg általános, kognitív és emocionális működésekben játszott szerepével.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

Az agresszió szabályozásának idegrendszeri hátterére vonatkozó eddigi elméletek három fő módszerrel nyert eredményeken alapulnak: állatokon végzett elektromos ingerléses kísérleteken, állati és emberi alanyokon végzet, vagy általuk elszenvedett agyi léziók következményeinek feltárásán, illetve az agresszió által kiváltott idegrendszeri aktivációs mintázatok felderítésén. E technikák anatomiai felbontása elég alacsony, ami elsősorban az olyan nagy kiterjedésű, és bonyolult szerveződésű struktúrák esetében problematikus, mint amilyen a prefrontális kéreg. Az optogenetikai technika közelmúltbeli fejlődése új perspektívákat nyitott az agresszió idegrendszeri hátterének megértése szempontjából. E technikák lehetővé teszik specifikus idegsejt populációk, és azok projekcióinak elkülönített vizsgálatát. Optogenetikai technikákat mindeddig egyetlen agresszivitással kapcsolatos tanulmányban használtak fel; ez a hypothalamus szerepét vizsgálta (Lin et al.; Nature. 2011; 470: 221-226). A hypothalamus "végrehajtó" mechanizmusainak feltárása természetesen fontos lépés az agresszió idegrendszeri hátterének megértése szempontjából, de legalább ilyen fontos azoknak az idegrendszeri folyamatoknak a megismerése, amelyek e végrehajtó mechanizmust működésbe hozzák. Az optogenetikai technika alkalmazásának fő célja az lesz, hogy különválasszuk a prefrontális kéreg kognitív/emocionális funkcióit azoknak a prefrontális idegsejteknek a szerepétől, amelyek az agresszió kiváltásában közvetlenül szerepet játszanak. Úgy véljük, hogy e technikák bevetésével az agresszió szabályozó mechanizmusai kiemelkedő részletességgel írhatók le. Az általános kognitív/emocionális és a specifikus, agresszió-szabályozó mechanizmusok elkülönített és összehasonlító vizsgálata ugyanakkor olyan információkat szolgáltathat, amelyek alapján feltárhatóvá válik a döntéshozó és magatartás-szabályozó funkciók komplex kapcsolata az agresszív viselkedés vonatkozásában. Végezetül úgy gondoljuk, hogy a prefrontális és kéreg alatti kapcsolatok neurokémiájának feltárásával hozzájárulunk majd az agresszióval járó pszichiátriai zavarok új terápiás célpontjainak felderítéséhez.

A kutatás összefoglalója, célkitűzései laikusok számára

Az Egészségügyi Világszervezet 2002 óta követi nyomon az agresszió és egészség kapcsolatát (lásd "http://who.int/violence_injury_prevention/violence/world_report/en/index.html"), és 2014 januárjában - miután megállapította, hogy az agresszív cselekmények súlyosan megterhelik a tagállamok költségvetését, amelyek dollármilliárdokat költenek az agresszióból származó egészségügyi, jogi, és emberi erőforrás problémák kezelésére - felhívta a tagállamokat olyan akciók kezdeményezésére, amelyek ezt a terhet csökkentik. Pályázatunk e technikák kiaknázását javasolja az agressziót szabályozó mechanizmusok megismerése céljából, és előrevetíti a döntéshozó és magatartás-végrehajtó mechanizmusok kapcsolatának felderítését a prefrontális kéregben, amely az agy legmagasabb döntéshozó régiója.

Summary of the research and its aims for experts
Describe the major aims of the research for experts.

We aim at revealing a novel mechanism of aggression control by using optogenetic techniques. The targets of research are the projections of specific prefrontal neurons to subcortical regions that are involved in the execution of aggressive attacks. We revealed earlier that subpopulations of neurons located in the prelimbic and infralimbic cortex project to hypothalamic regions directly involved in aggression control. In an ongoing study, the optogenetic stimulation of prelimbic/infralimbic efferents in one of the target areas (e.g. the lateral hypothalamus) increased the number of attacks delivered to opponents, and markedly increased the share of attacks aimed at the head, throat and belly, a common feature of attacks performed by rats submitted to etiological models of aggression-related psychopathologies. These findings surprisingly suggest that subpopulations of prefrontal neurons activate subcortical mechanisms involved in the execution of attacks; moreover, they trigger the delivery of abnormal and violent attacks. We plan to extend these studies to other target areas, particularly the mediobasal hypothalamus, as well as the medial and central amygdala, which all receive projection from the prelimbic and infralimbic cortex, and are tightly involved in the control of species-typical and escalated aggression. The effects of optogenetic stimulations will be crosschecked by those of the optogenetic silencing of projections. We also plan to characterize the neurochemical nature of the neurons implicated in this mechanism, and to compare the specific effects of these projections with the more general effects of the prefrontal cortex in cognitive and emotional processing.

What is the major research question?
Describe here briefly the problem to be solved by the research, the starting hypothesis, and the questions addressed by the experiments.

While deficits in the prefrontal cortex (PFC) exacerbate aggressiveness, the PFC is activated when aggressive behaviors are performed, which opens the question of how deficits and activations in the very same brain region can be associated with aggressiveness. It was postulated earlier that the PFC inhibits aggressiveness indirectly by affecting cognitive operations and emotional responses during conflict. Based on earlier findings and the results of an ongoing study we hypothesize that the prefrontal cortex also hosts mechanisms that are directly involved in the elicitation of species-typical aggression; moreover, it can trigger the execution of abnormal and violent forms of attack. We posit that higher-order decisions that result from cognitive and emotional processing at the level of the PFC are mediated by specific subgroups of neurons located in the same area, which target subcortical structures involved in the organization and execution of aggressive behavior. Thus we propose that the prefrontal cortex has a dual role in aggression control: at a general level it evaluates the conflict situation by cognitive and emotional processing, while a subpopulation of its neurons directly activates executive processes embedded in lower brain areas when cognitive and emotional processing results in the decision to attack. The experiments will (1) identify the PFC neuron populations that target subcortical structures; (2) characterization their roles in aggression, (3) identify their neurochemical properties and (4) compare their specific roles in aggressive behavior with more general roles of the PFC in aggression-related cognitive and emotional processing.

What is the significance of the research?

Current theories on the circuits that control aggressive behavior in general and on the role of the prefrontal cortex in particular are based on three main lines of evidence: electrophysiological studies performed in animals as well as brain lesion and brain imaging studies performed in animals and humans. The anatomical resolution of these techniques is relatively low, which is problematic especially in the case of large brain areas that have a complex organization like the prefrontal cortex. Advancements in the field of optogenetics open new perspectives for understanding the neuronal underpinnings of aggressive behavior. These techniques allow the dissection of mechanisms at the level of neuron populations, and at the level of their specific projections to identifiable target neurons. The only available study using optogenetic techniques in the field of aggression research focused on the hypothalamus (Lin et al.; Nature. 2011; 470: 221-226). While understanding the executive mechanism that resides in this brain area is unquestionably important, the understanding of higher order, decision-related mechanisms that control its activation appears at least similarly essential, especially from the point of view of aggression-related psychopathologies. By using optogenetic techniques we aim at dissecting general cognitive/emotional and specific executive roles played by the prefrontal cortex in aggression control. These studies forecast the possibility of describing the circuitries underlying aggressive behavior in great detail. At the same time, by comparatively studying general and specific effects, we hope to provide the first data on the complex relationship between decision-making mechanisms that reside in the PFC, and lower order executive mechanisms that are controlled by neuron population located in the same area. Finally, we hope that by characterizing the neurochemical characteristics of prefrontal-subcortical projections we will contribute to the development of novel treatment strategies for aggression-related psychopathologies.

Summary and aims of the research for the public

The World Health Organization monitors issues relating violence and health since 2002 and launched a global campaign for violence prevention in January 2014 (see "http://who.int/violence_injury_prevention/violence/world_report/en/index.html"). This organization concluded that violence places a massive burden on national economies, costing countries billions of US dollars each year in health care, law enforcement and lost productivity, and urged Member States to implement programs to limit this burden. While alleviating violence requires concerted action in many fields from education to legal actions, one important field of such actions involves scientific research into the factors that elicit and maintain high rates of violent and aggressive behavior. Neurobiological factors are under close scrutiny since 1926 when the Swiss physiologist Hess discovered that the electrical stimulation of certain brain regions immediately releases aggressive attacks on social partners. These findings were followed by a large number of animal and human studies that described the main brain mechanisms underlying aggressive behavior. Understanding was hampered, however, by technical limitations, which were recently greatly reduced by the introduction of optogenetic techniques, which allow the study of behavioral control at neuronal level. Our proposal exploits the great potentials of this technique in the field of aggression research, and promises the understanding of the processes that translate decisions taken at the level of the prefrontal cortex (the highest-order decision-making area of the brain) into neuronal "commands" that result in the expression of violence.

Ezt a hipotézist sikerült kísérletileg igazolni. A projekt második, származtatott célja az volt, hogy azonosítsuk a prefrontális kortex specifikusan agressziót szabályozó idegsejt csoportjait, és leírjuk szerepüket az az agresszió szabályozásában. Végül azonosítani szerettük volna azokat a projekciós területeket, amelyek e hatások közvetítésében szerepet játszanak. Végül le szerettük volna írni a prefrontális kortex érintett neuron populációinak neurokémiai jellegzetességeit. A javasolt fő kutatási módszer az optogenetika lett volna. Ezeket a célokat maradéktalanul két egymástól nagyrészt elkülönült neuronpopuláció esetében sikerült elérni. Ezek a prefrontális neuron populációk agresszió szabályozásában kulcsszerepet játszó hipotalamusz különböző régióit idegzik be, és az agresszív viselkedés különböző megnyilvánulásainak szabályozásában játszanak szerepet. Mindkét neuron csoport glutamáterg volt. Igazoltuk továbbá a cannabinoid jelátvitel, valamit a két fő szertonerg központ egymástól különböző szerepét az agresszióban. A kutatási eredményeket néhány vezető review folyóiratban is összegeztük (pl. Nat Rev Neurosci).

The basic hypothesis of the project was that the prefrontal cortex – in addition to its widely accepted negative inhibitory role - has an active role in the positive control of aggression. This hypothesis was confirmed experimentally. The second goal of the project was to identify the prefrontal neuron populations and their subcortical targets that control aggression. We also aimed at the neurochemical characterization of prefrontal neurons. The main experimental methodology was to be optogenetics. These aims were achieved fully in the case of two rather distinct prefrontal neuron populations which target two different hypothalamic regions – both involved in aggression control .- and which control different aspect of aggressive behavior. Both populations were glutamatergic. In addition we described the role of cannabinoid signaling, and the differential role of the two main serotonergic nuclei in the control of aggressive and social behavior. Our findings were summarized in leading review journals, including Nat Rev. Neurosci.
