A heparin poliszacharid antikoaguláns tulajdonságainak köszönhetően felbecsülhetetlen gyógyszer a thromboembóliás betegségek megelőzésére és kezelésére. Alkalmazása azonban számos korlátba ütközik polianionos és heterogén természete miatt. A kutatások során mérföldkő volt egy szintetikus heparin-pentaszacharid gyógyszer (Arixtra) kifejlesztése, amellyel sikeresen minimalizálták az antikoaguláns terápia mellékhatásait. Az Arixtrának azonban van néhány klinikai hiányossága (pl. rövid felezési idő), és szintézise is bonyolult. Az egyszerűbben előállítható és aktívabb Idraparinuxnak a túl hosszú felezési ideje miatt az orvosi alkalmazása kockázatos. A hasonló aktivitású és megfelelő felezési idejű, de egyszerűbben szintetizálható heparinoidok kifejlesztése fontos feladata a szénhidrátkémikusoknak. Az L-iduronsav a heparin egyik fontos eleme, de hatékony szintézise még nem megoldott, ami jelentősen megnöveli a szintézisek időigényét és költségeit. Megoldást jelenthet kutatásunk célja: az iduronsav helyettesítése egy egyszerűbb szerkezeti elemmel vagy új és hatékony reakcióút kidolgozása L-iduronsav előállítására. Célunk az L-iduronsav helyettesítése 6-dezoxi-L-talopiranózzal, ezáltal új típusú, olcsóbban előállítható véralvadásgátlók szintézise. Ismert, hogy az L-iduronsav konformációs flexibilitása elengedhetetlen a biológiailag aktív térszerkezet kialakulásához. Feltételezzük, hogy ezt az aktív konformációt a jóval egyszerűbben és olcsóbban előállítható 6-dezoxi-L-talopiranóz is biztosíthatja. Célunk parciálisan metilezett pentaszacharid analógok előállítása is, mivel a metilezettségi fok csökkentésével optimalizálhatók a molekulák farmakokinetikai tulajdonságai.

Mi a kutatás jelentősége?

A heparint az 1940-es évek óta alkalmazzák a gyógyászatban, mint véralvadásgátló gyógyszert. Az Enoxaparin (Lovenox, Xaparin, Clexane), a heparin frakcionálásával készült kis molekulatömegű oligoszacharidok keverékéből álló készítmény (LMWH), évek óta világviszonylatban a tíz legtöbbet forgalmazott gyógyszer között van. A szintetikus analógok (pl.: Arixtra, Idraparinux) ugyan hatékonyabbak, mint a heparin vagy az Enoxaparin készítmények, de szintézisük bonyolultsága miatt a gyógyszerpiacon nem képesek felzárkózni a természetes alapú származékokhoz.
Kutatási projektünk olyan új, véralvadásgátló hatású Idraparinux analógok szintézisére irányul, amelyek optimális felezési idővel rendelkeznek, és az általunk tervezett változtatásoknak köszönhetően gazdaságosabban, jobb hatékonysággal szintetizálhatók, mint a jelenleg ismert származékok. A heparinok szintézisének legproblematikusabb része az L-iduronsav egység, ez a különleges cukor szabad állapotban nem fordul elő, előállítására nincs jó eljárás, több mint 10 lépést igényel és nagyon kis hatásfokú. Ezért ennek a szerkezeti elemnek az egyszerűsítésével vagy gazdaságos előállításával igazi áttörést lehetne elérni a szintetikus véralvadásgátlás területén. Ebből kiindulva egyik célunk az L-iduronsav helyettesítése 6-dezoxi-L-talopiranoziddal, amely rendelkezik az iduronsavra jellemző, és a biológiai hatáshoz nélkülözhetetlen szerkezeti flexibilitással, és szintézise egyszerű és hatékony. Másik célunk a piranóz gyűrű konformációs inverzióján alapuló, új és hatékony L-iduronsav-szintézis kidolgozása. Ezek a stratégiák könnyebbé, gazdaságosabbá és hatékonyabbá tehetik az összes heparinoid molekula szintézisét, így jelentősen felgyorsíthatják az Alzheimer-kór, az ateroszklerózis vagy a daganatok terápiájának terén jelenleg folyó heparin-alapú gyógyszerkutatást is.
Mindezek mellett a tervezett vegyülettípusok előállítása hozzájárulhat a szénhidrátkémiai szintézismódszerek (glikozilezési reakciók, védőcsoport manipulációk) fejlesztéséhez. A leghatásosabb molekulák gyógyszerkémiai fejlesztés alapját képezhetik.

A thromboembólia korunk veszedelmes járványának mondható, különösen az iparosodott országokban megdöbbentő a gyakorisága. A verőereket érintő problémák, mint a szélütés, szívizomelhalás és érszűkület közismertek, de mellettük az érrendszer „másik”, vénás oldalának a megbetegedései is gyakoriak, és következményeikben ugyanolyan félelmetesek. A vénás trombózis, illetve a tüdőembólia az Európai Unió évente mintegy félmillió polgárának a halálához vezet, és a megbetegedés gyakorisága egyre nő, amelynek több oka is van. Ezek közt megemlítendő a mozgáshiányos életmód, a hosszú repülőutak, az egyébként hatásos gyógyszerek trombózisra hajlamosító mellékhatása, illetve a fogamzásgátlók és egyéb hormonkészítmények fokozott elterjedése. Hasonlóan a nyugati országokhoz, hazánkban is komoly népegészségügyi terhet jelent a vénás thromboembóliás betegek egyre növekvő száma. Mindezen betegségek kezelésére és megelőzésére véralvadásgátló szereket alkalmaznak. A modern szemléletű alvadás-gátló megelőzés és kezelés döntő jelentőségű a mai társadalomban, kulcsfontosságú a leghatékonyabb és elegendő ideig alkalmazott terápia megválasztása. A téma jelentőségét jól mutatja, hogy jelenleg is számos kutatás foglalkozik az újabb, kedvezőbb tulajdonságokkal bíró véralvadásgátlók előállításával. A probléma jelentősége miatt célul tűztük ki olyan szénhidrátszármazékok előállítását, amelyek egyszerűbben és gazdaságosabban előállíthatók, mint az eddigi készítmények, hatékonyabbá téve így a vérrögképződés elleni védekezést. Jelentős előrelépés lehet a heparin-típusú molekulák szintézisében az általunk választott szerkezeti elem kiváltása egy egyszerűbb szerkezettel vagy előállításának optimalizálása.

The anticoagulant properties of the heparin polysaccharide made it an invaluable drug for prevention and treatment of thromboembolic diseases. However, application of heparin suffers from a number of limitations due to its polyanionic and heterogenous nature. The development of a synthetic pentasaccharide into a novel antithrombotic drug (Arixtra) was a milestone, minimizing the side-effects in anticoagulant therapy. Arixtra, however, also has some clinical shortcomings and requires a multistep, challenging synthesis. Thus, the search for additional heparinoid derivatives with specific activity and feasible synthesis is a major challenge to oligosaccharide chemists. The L-iduronic acid is an important element of heparin, but its efficient synthesis has not been solved, which significantly increases the time and cost of the preparation of heparinoids. In the frame of this project our aim is to resolve this issue both by substitution of iduronic acid with a simple structural replacement and development of a new and efficient pathway for the L-iduronic acid. Our goal is to substitute the L-iduronic acid with 6-deoxy-L-talopyranoside, in this way new types of anticoagulants can be produced at significantly lower cost. It is known that the conformational flexibility of the L-iduronic acid is essential for the formation of the bioactive structure. We assume that the 6-deoxy-L-talopyranoside, synthesis of which is much more simple and less expensive, may provide the active conformation. As the pharmacokinetic properties of the molecules can be optimized by reducing the degree of methylation, we also aim at the preparation of partially methylated pentasaccharide analogues.

Can the reduction of the degree of the methylation be beneficial for the biological half-life and activity of the planned molecules?
Can a new and efficient pathway based on the conformational inversion of the carbohydrate ring be developed for the synthesis of L-idose?

Heparin has been employed in medical practice since the 1940’s as a blood-anticoagulant. Enoxaparin (Lovenox, Xaparin, Clexane), a mixture of low molecular weight oligosaccharide fragments of heparin (LMWH) has been among the world's ten most traded drugs for years. The synthetic analogues (eg.: Arixtra, Idraparinux) are more effective than heparin or Enoxaparin preparations, however, because of their clinical shortcomings and the complexity of their synthesis, there is a constant need for the development of new synthetic anticoagulants of both a better pharmacological profile and a feasible synthesis.
The aim of our research project is to develop new Idraparinux-analogue anticoagulants possessing an optimal half-life, and due to the planned structural simplification, synthesis of which can be carried out in a more economical and efficient way than the currently known derivatives. The most problematic part of the synthesis of heparin derivatives is the L-iduronic acid unit. This unique sugar does not occur as a free monosaccharide, and is not available commercially. Its synthesis requiring more than 10 steps is very laborious and inefficient. Consequently, either by replacement of idose unit with a substantially simpler structural element or by developing an improved, cost- and time-saving method for the iduronic acid building block may be a real breakthrough in the field of anticoagulant therapy. One of our goals is the substitution of L-iduronic acid with 6-deoxy-L-talopyranoside which can ensure the adoption of the bioactive sterical structure of the pentasaccharide due to its crucial conformational flexibility. Moreover, its efficient and simple synthesis is well-documented. Another aim of our research is to develop a fundamentally new synthesis of the L-iduronic acid based on the conformational inversion of the pyranose ring. These strategies could be utilized in the synthesis of other heparin-based drug candidates. Thus, the pace of drug-development in the therapy of Alzheimer's disease, atherosclerosis or cancer metastasis could be significantly improved.
Our work may also contribute to the development of synthetic methods in the field of oligosaccharide chemistry.

The thromboembolism is a dangerous epidemic of our time. The frequency is especially startling in the industrialized countries. Problems affecting the arteries, such as stroke, myocardial infarction and atherosclerosis are well known, but venous diseases are also common, and the consequences are similarly dreadful. Venous thrombosis and pulmonary embolism lead to the death of about half a million people each year in the European Union, and the frequency of these diseases is increasing. The sedentary lifestyle, long flights, side effects predisposing to thrombosis of some drugs and the increased consumption of contraceptives and other hormones are among the reasons. Similarly to the Western countries, an increasing number of patients with venous thromboembolism is a burden on the public healthcare system of our country. For the therapy of such diseases anticoagulant agents are typically used. Modern approaches to the anti-clotting prophylaxis and treatment are crucial in today's society, and the selection of the most effective therapy is highly important. The significance of this issue is well demonstrated by the substantial research efforts directing at the synthesis of novel anticoagulants of improved properties. Because of the relevance of the problem, our aim is to develop anticoagulant agents which are easier and more economical to synthesize compared to the current drugs, thus making the fight against blood clotting more effective. Our goal is to achieve substantial progress in the synthesis of heparin-like molecules by changing their most complicated unit for a simple structural substituent or by developing an efficient method for its synthesis.

A heparin poliszacharid antikoaguláns tulajdonságainak köszönhetően felbecsülhetetlen gyógyszer a thromboembóliás betegségek megelőzésére és kezelésére. Alkalmazása azonban számos korlátba ütközik. A hasonló aktivitású és megfelelő felezési idejű, de egyszerűbben szintetizálható heparinoidok kifejlesztése fontos feladata a szénhidrátkémikusoknak. A posztdoktori pályázatom futamideje alatt idraparinux analóg pentaszacharidok szintézisével és az L-idóz rész gazdaságosabb előállításnak lehetőségével foglalkoztunk. Ezen belül egyrészt az iduronsav helyett 6-dezoxi-L-talopiranóz egységet tartalmazó heparin-analóg pentaszacharidokat állítottunk elő és vizsgáltuk ezen származékok véralvadásgátló hatását humán vérplazmán. Másrészt egy irodalmi eljárás módosításával kifejlesztettünk egy hatékony, új módszert L-idopiranozil donorok előállítására. Az általunk fejlesztett módszert alkalmazva elsőként állítottunk elő megfelelően védett, aktiválható anomer csoporttal rendelkező, 5,6-telítetlen D-glükóz származékokból jó hozammal, és az eddigieknél gazdaságosabban L-idóz donorokat. Eredményeinket referált nemzetközi folyóiratokban publikáltuk, valamint poszter és előadás formájában hazai és külföldi konferenciákon mutattuk be.

Because of the anticoagulant properties of heparin polysaccharide, it is an invaluable drug for preventing and treating thromboembolic diseases. However, its use has many limitations. The development of heparinoids with high activity and proper half-lives, which are easier to prepare is an important task for carbohydrate chemists. During my post-doctoral application period we are dealing with synthesis of idraparinux analogue pentasaccharides and the development of a more economical production of the L-idose unit. First we prepared heparin-analogue pentasaccharides containing 6-deoxy-L-talopyranose unit instead of the iduronic acid and the anticoagulant activity of these derivatives was studied on human plasma. On the other hand, by modifying a literature process, we have developed an effective new method for the preparation of L-idopyranosyl donors. Using our new method, we have produced L-idose donors from 5,6-unsaturated alpha-1-thio-D-glucose derivatives with good yields and more economically than before. The results were published in high ranking international journals and presented at national and international conferences in the form of posters and presentations.
