A tervezett kutatás Drosophila melanogaster modellszervezetben azonosítja a sejt közvetítette immunitás szabályozó folyamatait. Laboratóriumunk a Drosophila immunrendszerének a vizsgálatára alkalmas molekuláris és genetikai eszközrendszert hozott létre, melynek alkalmazásával meghatározta a Drosophila funkcionális vérsejt-populációinak szöveti és differenciálódási vonal eredetét. Azonosítottunk továbbá egy eddig ismeretlen vérsejttípust a sokmagvú óriás hemocitát, mely a Drosophila melanogaster lamellocitáinnak funkcionális megfelelője és gerincesekben, a krónikus gyulladások során képződő óriássejtekkel mutat nagyfokú hasonlóságot. Ezeknek az eredményeknek az alapján a következő célokat tűztük ki:
i) A Drosophila melanogaster immunrendszerének a vizsgálatára alkalmas molekuláris és genetikai markereket hozunk létre és jellemezzük a markermolekulák funkcióit.
ii) Gén-expresszió mintázat analízis végzünk a hemocita aktiválódás és differenciálódás során D. melanogaster-ben megnyilvánuló gének sejttípus-specifikus azonosítására és jelölteket választunk további vizsgálatokra.
iii) Azonosítjuk az újonnan azonosított sejttípus-specifikusan megnyilvánuló gének gerinces homológjait
iv) Fenntartjuk és fejlesztjük a multidiszciplináris kutató és oktató egységünket.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Kísérleteink célja a sejt közvetítette immunitás szabályozásának a megismerése. Mivel a Drosophila immunrendszere a gerincesek veleszületett immunitásának prototípusa, vizsgálatainkban modellszervezetként a Drosophila melanogastert alkalmazzuk. Általánosan elfogadott, hogy a vérsejtek differenciálódását Drosophilában és gerinces szervezetekben egymáshoz hasonló hálózatok szabályozzák, azonban a komponenseknek egy átfogó leírása és a komponenseknek differenciálódó populációkhoz rendelése nem történt meg. A Drosophila sejt közvetítette immunitásának a sejtes komponensei, a hemociták, az emlősök vérsejtjeihez hasonló funkciókkal rendelkeznek, úgy, mint az antimikróbiális peptidek termelése, a mikrobák bekebelezése és a nagyobb részecskéknek pl. egyes parazitáknak a környezetüktől történő elhatárolása, a tokképzés és a granulómaképzés. A közelmúltban egy eddig ismeretlen sejttípust azonosítottunk, a sokmagvú óriás hemocitát, mely példátlan hatékonyságú parazitaölő képességgel rendelkezik. Ez a sejt funkcionális megfelelője a Drosophila melanogaster tokképző sejtjeinek, a lamellocitáknak, és sok tulajdonságában hasonlít a gerinces szervezetekben képződő granulómákra jellemző sokmagvú óriássejtekre. Mindezeknek alapján a szabályozó hálózatok kulcsszereplőit szeretnénk azonosítani a differenciálódó vérsejt alpopulációkban, differenciálódási vonalakban, Drosophila melanogaster modellszervezetben, a vérsejtek és a kompartmentumok gén-expresszió profil analízisével. A szabályozó elemek gerinces homológjait azonosítjuk.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

Jelen pályázat a vérsejt differenciálódás és a sejt közvetítette immunitás folyamatainak a megértését segíti az ecetmuslica és a laboratóriumi egér modellszervezetek vizsgálatán keresztül. A Drosophila melanogaster a vérsejtdifferenciálódás és a veleszületett immunitás vizsgálatára jól alkalmazható genetikai rendszerrel rendelkezik. Mivel több genetikai szabályozó út többek között a vérsejtdifferenciálódás és a veleszületett immunitás kulcselemei erősen konzerváltak, kísérleteink eredményei segítenek a vérsejtdifferenciálódás és a veleszületett immunitás általános érvényű szabályozási folyamatainak a megértésében. A Drosophila melanogasterben végzett kísérletek és a gerinces homológok azonosítása, összehasonlító analízise lehetővé teheti az eredményeink evolúciós vonatkozású értelmezését, amely által megismerhetünk olyan folyamatokat/faktorokat, amelyek a parazitákkal szembeni védekezésben kulcsszerepet játszanak. Továbbá, a gerinces szervezeten végzett kísérleteink nem csak támogathatják, és általános érvényűre terjeszthetik ki az eredményeinket, de lehetővé tehetik az alkalmazott orvostudományra történő hasznosulást. A törzsfejlődés során konzervált mintázatok és szabályozó útvonalak megismerése mindezeknek a biológiai és az orvostudományokban játszott jelentőségét hangsúlyozza. A kísérletek eredményeként generált ismeretek hosszútávon is alapvető fontosságúak, mivel a veleszületett immunitás szabályozó elemei specifikusan gátolhatók, amiért vonzó targetek lehetnek a gyógyításban, vagy a rovar-kártevők elleni védekezésben. A projekt új irányból enged betekintést a veleszületett immunitás folyamataiba, ezáltal megvalósulása esetén átfogó képet kapunk a mikrobákkal és a parazitákkal szemben kialakuló immunfolyamatokról. A projekt a reagensek széles körét eredményezi. Az adatbankokat és a reagenseket, a korábbiakhoz hasonlóan, a kutatói közösség rendelkezésére bocsájtjuk.

Az immunrendszer működésében létrejövő bármely hiba betegséget eredményez, ezért az immunrendszer fejlődésének és működésének a megismerése alapvető fontosságú. A rovarok és a gerinces szervezetek szabályozó folyamatainak a nagyfokú konzerváltsága és a Drosophila jól ismert genetikai rendszere ideális modellszervezetté teszi az ecetmuslicát az általános orvosbiológiai és immunológia folyamatok vizsgálatára. A szerzett immunitással nem rendelkező Drosophilán végzett kísérletek fontos szerepet játszanak az immunitás folyamatainak a megismerésében. Ezt mutatja a 2011-ben, Bruce Beutler és Jules Hoffman által kiérdemelt Nobel díj, melyet a Drosophila és az emlősök veleszületett immunitásának a vizsgálatában elért eredményeikért adományoztak. A tokkéző reakció és a granulóma képződés az állatvilágban a veleszületett immunitás elemeinek a krónikus ingerekre adott általánosan elterjedt válaszreakciója. Valójában, a Drosophila lárvának a parazita fertőzésre törtnő válaszreakciója, a tokképzés a granulómaképződés modelljeként szolgálhat. Kísérleteink célja az, hogy megismerjük a tokképző reakció során az immunsejtekben bekövetkező gén-expressziós változásokat Drosophilában és azonosítsuk a törzsfejlődés során konzervált elemeket a laboratóriumi egérben és az emberben. Summary of the research and its aims for experts

The proposed project will identify regulatory mechanisms of the cell mediated immunity in the Drosophila melanogaster model organism. Our laboratory has generated molecular and genetic tools for the analysis of the Drosophila immune system and characterized the hemocyte subsets with respect to their tissue and lineage origin. We recently discovered a so far unidentified cell type, dubbed the multinucleated giant hemocyte, which is the functional homolog of the lamellocyte in Drosophila melanogaster and is similar to multinucleated giant cells, a characteristic for granulomas, a manifestation of chronic inflammation in vertebrates. Based on these findings we propose a project, with the aims of:
i) identifying and constructing novel molecular and genetic markers for further analysis of the immune system in Drosophila melanogaster and characterizing the identified novel molecules with respect to their function;
ii) performing a comparative gene expression profile analysis of hemocyte activation and differentiation in Drosophila melanogaster and selecting gene candidates for further analysis;
iii) finding vertebrate homologs of the newly identified molecules
iv) maintaining and further developing the multidisciplinary research team and the undergraduate and postgraduate training unit with a broad interest in molecular immunology based on our research and teaching experience.

What is the major research question?
Describe here briefly the problem to be solved by the research, the starting hypothesis, and the questions addressed by the experiments.

The basic question which we would like to address is, how the cell mediated module of innate immunity is regulated. As the immune system of Drosophila is a prototype of innate immunity of vertebrates we use Drosophila melanogaster as a model organism. It is generally accepted that the differentiation of blood cells is regulated by similar regulatory networks in Drosophila and in vertebrates, however, a comprehensive description of the components, their assignment to definite stages of differentiation is still lacking. The cellular components of cell mediated immunity of Drosophila, the hemocytes, have several functions, orthologous with mammalians, i.e. production of antimicrobial peptides, phagocytosis of microbes and encapsulation of foreign bodies, such as parasites. Recently, a novel cell type, the multinucleated giant hemocyte was discovered in our laboratory with an unprecedented parasite killing capacity, which is the functional homolog of the lamellocyte in Drosophila melanogaster and with many features resembling the multinucleated cells developing in the course of the chronic inflammations in vertebrates. Taking these into account we suggest to pinpoint key components of the regulatory networks to defined subsets of differentiating hemocytes and hemocyte compartments, using Drosophila melanogaster as a model organism. We will find homologs of the newly identified Drosophila regulatory components in vertebrates.

What is the significance of the research?

The Proposal focuses on a better understanding of the cell-mediated immune response and blood cell differentiation using Drosophila melanogaster and the laboratory mouse as model organisms. Drosophila provides a genetic model system with a well established applicability to the genetic control of blood cell development and the first line immune defense, the innate immunity. As the core components of many genetic pathways, including those of blood cell development and innate immunity, are conserved from insects to humans our research will help to understand the regulation of blood cell development and innate immunity in general. A comparison of the results obtained from Drosophila melanogaster and the laboratory mouse has the potential to provide an evolutionary context to our conclusions, and may highlight specific conserved event processes/factors responsible for the anti-parasite immune response. Furthermore, the possibility of investigating candidates in a vertebrate system not only supports and extends the evolutionary aspect of the work, but may also allow the future translation of the results into applied medicine. The identification of phylogenetically conserved patterns and pathways will shed light to their biological significance in health and in disease. Furthermore, this knowledge is essential in the long term as the regulators and the mechanisms of innate immunity have chemical inhibitors making them attractive targets for therapeutic intervention or pest control. The project will lead to new insights into innate immunity and will provide a comprehensive overview of the innate immune response against microbes and parasites in general. The project will result in the generation of a broad range of immunological reagents and molecular probes. All databases and reagents will be provided for research purposes, similarly as in the case of the previously generated reagents, which are freely available for the research community.

Any malfunction of the immune system may be associated with diseases ranging from the common cold to cancer, therefore it is of great interest to understand the molecular basis of its development and functioning. The conserved genetic pathways between flies and vertebrates, combined with the powerful genetic resources provided by Drosophila, make this organism an ideal system to model immunological phenomena related to human biology and medicine. Work on Drosophila, which lack an acquired immune response, play an important role in our understanding of how innate immunity works. This was acknowledged by the award of the Nobel Prize in Physiology or Medicine in 2011, part of which went to Jules Hoffmann and Bruce Beutler for their work on innate immunity in Drosophila and mammals, respectively. The encapsulation reaction and granuloma formation in the animal kingdom are typical responses to chronic inflammatory stimuli of the innate immune system. In fact, the encapsulation reaction and the capsule formation in the Drosophila larva induced by parasitoid infection serve as a viable model through which to study granulomas. Our goal is to define the gene expression changes during the differentiation of immune cells by a global gene expression profile analysis, to identify key components of the regulatory networks in the course of the cell mediated immunity in Drosophila, and to recognize phylogenetically conserved elements of innate immunity in mammals, including the laboratory mouse and man. A projekt lehetőséget adott a Drosophila vérsejt-differenciálódás vizsgálatára alkalmas analitikai módszerek fejlesztésére, alkalmazására és ezáltal új ismeretek szerzésére. A projekt keretében a profenoloxidázon új immunológiai epitópokat azonosítottunk, amelyek egyaránt jelen vannak a Drosophila és a mézelő méh vérsejtjein, így lehetővé válik a kristálysejtek azonosítása a mézelő méhben is. Meghatároztuk a Nimród gén klaszter több tagjának a funkcióját. Az emberi homológgal rendelkező Drosophila headcase génről megállapítottuk, hogy a lamellocita-sors meghatározásában játszik szerepet. Az effektor vérsejtek két típusának eddig ismeretlen két differenciálódási irányát írtuk le. A molekuláris markerek segítségével egysejtes tömegspektrometriai analízist alkalmaztunk a Drosophila vérsejtek immun-profiljának jellemzésére. A módszer alkalmazásával lehetővé válik a vérsejtpopulációk és differenciálódási vonalak kapcsolatainak magas felbontású megismerése. Az előbbiekben említett genetikai és immunológiai rendszereket a gépi tanulás technológiával elemeztük és lefektettük az egysejtes transzkriptóm analízis gépi tanulásos módszerrel történő, matematikai algoritmusok segítségével történő megismerésének alapjait.

The aim of the studies was to identify and construct novel molecular markers with the aim to reveal the functional heterogeneity and regulatory networks of Drosophila blood cell populations. We found novel immunological epitopes on prophenoloxidases, which are present in Drosophila and in the honey bee (Apis mellifera), thus, enabling the identification of crystal cells in the honey bee too. The function of members of the Nimrod gene cluster were uncovered.The function of the Drosophila gene headcase, with human homolog (HECA) was revealed. We identified a so far unrecognized differentiation pathway of blood cells with the involvement of two types of encapsulating effector cells. Using the combination of genetic and immunological markers we invented single cell mass spectrometry too for immune profiling of Drosophila blood cells. This method permits the identification of hemocyte subsets and to reveal their connections and developmental relationships with an exceptional resolution. The developed immunological and genetic systems, the methodology and findings were combined with deep learning technology, and the basis for single cell isolation and transcriptome analysis at an analytical scale was established.
