A kutatás összefoglalója, célkitűzései szakemberek számára
Itt írja le a kutatás fő célkitűzéseit a témában jártas szakember számára.

A tervezett kutatómunka során fluid és szilárd határfelületeket, és e rendszerekben végbemenő biológiai vagy környezeti szempontból releváns folyamatokat tervezünk vizsgálni, elsősorban számítógépes szimulációs módszerrel. A tervezett kutatás három fő részre bontható: i) fluid (folaydék-gőz és folyadék-folyadék) határfelülettel kapcsolatos problémák, ii) lipid membránokkal kapcsolatos problémák, és iii) szilárd (szilárd-gáz és szilárd-folyadék) határfelülettel kapcsolatos problémák,. Az első problémacsoportba tartozik a folyadék felülettel kapcsolatos alapvető kérdések vizsgálata: termodinamikai mennyiségeknek (sűrűség, nyomás, felületi feszültség, energia, szabadenergia) a valódi (intrinsic) felülethez viszonyított profiljainak számítása, a felület termodinamikájának jellemzése e profilok illetve az egyes felület alatti molekuláris rétegek által adott járulékok segítségével. Ide tartozik a biomolekulák evolúciója szempontjából releváns kis molekulák szabad vízfelszíni adszorpciójának és adszorpciós rétegen belüli laterális asszociációjának vizsgálata, illetve a szobahőmérsékleten folyékony ionos rendszerek folyadékfelszínének részletes vizsgálata. A második kérdéscsoport központi kérdése az anesztetikumok hatása lipid membránok tulajdonságaira, célja hogy közelebb kerüljünk az anesztézia jelenségének molekuláris magyarázatához. Végül a harmadik kérdéskör atmoszférakémiai és asztrofizikai jelentőségű kérdésekkel (pl. halogénezett szénhidrogének megkötődése jég felületén magaslégköri körülmények között), valamint magnetit nanorészecskék felületi borításával és vizes közegben való stabilizálásával foglalkozik.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

A tervezett kutatómunka meglehetősen sokrétű, így több alapkérdés megválaszolását is tervezzük. A fluid határfelületeket érintő vizsgálatok alapkérdése az, hogy hogyan tudjuk a felületek termodinamikai tulajdonságait jellemezni valódi (intrinsic) profilok illetve rétegenkénti járulékok segítségével. Fontos alapkérdés az is, hogy a biomolekulák evolúciója során a korabeli óceánok felületén milyen mennyiségben tudtak feldúsulni az akkor jelen lévő kis molekulák, és mely ilyen molekulák milyen relatív orientációjú asszociátumai vezethettek el a mai biomolekulák kialakulásának további lépései felé. Lipid membránok esetén az alapkérdés az, hogy hogyan változtatják meg anesztetikumok lipid membránok tulajdonságait, mely membrántulajdonságok jöhetnek illetve nem jöhetnek szóba az anesztézia jelenségének molekuláris értelmezésekor. Szilárd felületek vizsgálata során alapkérdésünk, hogy mennyiben változnak meg az adszorpciós réteg tulajdonságai (orientációk, jégfelülettel való illetve laterális kölcsönhatások, elsősorban hidrogénkötések) ha egyszerre többféle molekula is adszorbeálódhat a jégfelületen, illetve mennyire szelektív ez az adszorpció egyes komponensekre. Hasonlóképpen, a szelektivitás az alapkérdése a klatrát hidrátokra vonatkozó tervezett kutatásoknak is. A tervezett kutatásokat módszerfejlesztési munkák teszik teljessé, itt értelemszerűen a kívánt célokra alkalmas, megvalósítható számításigényű módszerek kidolgozása a cél, amelyek majd a későbbiekben további kutatások alapjául szolgálhatnak.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

A tervezett projekt elsősorban alapkutatási jellegű, számos eredménye azonban reményeink szerint felhasználható lehet további, alkalmazott kutatások során is. A kutatómunka során egy módszerfejlesztést, nevezetesen a laterális nyomásprofil számítására alkalmas módszer kidolgozását és program elkészítését is tervezünk E munka már elkezdődött, befejezése azonban a projekt keretében várható, a projektben tervezett számos vizsgálat alapul e munka sikerén. Tisztán alapkutatási probléma az egyes termodinamikai mennyiségek, elsősorban a felületi feszültség molekuláris rétegenkénti járuléka számításának, a prebiotikus evolúcióval, valamint a klatrát hidrátokkal kapcsolatos vizsgálatainknak van. Előbbi révén mélyebben megérthetjük a felületi feszültség természetét és molekuláris okait, ezek függését a molekulák közötti kölcsönhatások típusától, prebiotikus evolúciós vizsgálataink segítségével közelebb kerülhetünk az élet keletkezésének megértéséhez, míg klatrát hidrátokkal kapcsolatos kutatásaink a Naprendszer keletkezésének molekuláris részleteibe adhatnak betekintést. Noha a többi tervezett kutatásunk szintén alapkutatási jellegű, hasznosulásukat inkább későbbi, elsősorban környezeti illetve orvosbiológiai jellegű alkalmazott kutatásoktól várjuk. A szobahőmérsékletű ionos folyadékok felületének szisztematikus vizsgálata segíthetne ezen ígéretes, „zöld” oldószer család adott célra megfelelő tagjának kiválasztásában, anesztetikumokat illetve ópiumszármazékokat tartalmazó lipid membránokra vonatkozó kutatásaink segíthetnek megérteni ezen élettanilag fontos, a gyógyászatban elterjedten használatos, ám visszaélésekre is alkalmat adó molekulák élő szervezetre gyakorolt hatásának molekuláris mechanizmusát, magnetit felületre vonatkozó vizsgálataink eredményeit pedig a későbbiekben a funkcionalizált magnetit nanorészecskék használatával kapcsolatos orvosbiológiai kutatások használhatják fel. A jég felületén történő egyszerű és kompetitív adszorpciós vizsgálataink későbbi légkörkémiai/környezetvédelmi kutatások alapjául szolgálhatnak.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

A tervezett kutatómunka elsősorban alapkutatási jellegű, így eredményeitől nem elsősorban egy mindennapi életben hasznosuló találmányok kifejlesztése, hanem a különböző fázisok közötti határfelületeken lezajló folyamatok mélyebb, molekuláris szintű megértése várható. Az ilyen folyamatok természetesen sokszor kapcsolódnak a gyakorlati élettel kapcsolatos problémákhoz is, kutatásaink eredményei remélhetőleg alapjául szolgálhatnak későbbi, már valóban gyakorlati alkalmazások fejlesztését célzó ún. alkalmazott kutatásoknak. Kis légköri szennyező molekulák jég felületén való megkötődése a magas légkörben lejátszódó folyamat, mely különböző környezeti károsodásokhoz (pl. ózonpajzs bomlása) vezet. E kutatásaink eredményei így később a környezetvédelemben hasznosulhatnak. A vizes közegben eloszlatott parányi, mágneses tulajdonságú magnetit részecskék nagyon ígéretes rendszerek orvosbiológiai szempontból: az ilyen részecskéket és velük a felületükre kapcsolt gyógyszermolekulákat a szervezetben akár külső mágnessel is a megfelelő helyre irányíthatjuk. Az alkalmazhatóság azonban megkívánja, hogy e részecskék felületét előzetesen olyan anyaggal borítsák be, ami megakadályozza összetapadásukat (az ugyanis, egyebek közt, trombózisveszéllyel járna). Ilyen borítások vizsgálata a célja a kutatás ezzel kapcsolatos részének. Alapkutatási jellegű munkáink közül a biológiai prekurzor molekulákra illetve klatrát hidrátokra vonatkozó kutatásaink segíthetnek közelebb jutni az élet illetve a Naprendszer keletkezésének molekuláris részleteibe.

Summary of the research and its aims for experts
Describe the major aims of the research for experts.

In the frame of this project we intend to investigate processes and properties occurring at various fluid and solid interfaces, in particular, those of biological or environmental relevance. The method of the planned investigations is computer simulation. The planned research project consists of three main tasks, namely the molecular level investigation of i) problems related to fluid (i.e., liquid-vapor and liquid-liquid) interfaces, (ii) lipid membrane related problems, and iii) problems related to solid (i.e., solid-vapor and solid-liquid) interfaces. The first bunch of problems includes investigation of fundamental problems related to the liquid surface: determination of the intrinsic profiles of various thermodynamic quantities (density, pressure, surface tension, energy, free energy, etc.), characterization of the surface thermodynamics by the intrinsic profiles and layer-by-layer contributions of these. This group of problems includes adsorption of small molecules of prebiotic relevance at the free water surface and their lateral association at the liquid surface, and investigation of the liquid surface of room temperature ionic liquids. The central question of the second group of problems is how anesthetics affect the properties of liquid membranes, aiming at a better understanding of the molecular mechanism of anesthesia. The third group includes atmospheric chemistry and astrophysics related issues (e.g., adsorption of halogenated hydrocarbons at the surface of ice under tropospheric conditions), and the problems of potential biophysical application (coating and stabilizing) magnetite nanoparticles in aqueous environment).

What is the major research question?
Describe here briefly the problem to be solved by the research, the starting hypothesis, and the questions addressed by the experiments.

The planned research covers the study of a number of problems, and hence it is intends to answer several basic questions. The main question behind the planned investigation of fluid interfaces is how can the surface thermodynamics be characterized through intrinsic profiles of thermodynamic quantities and through layer-by-layer contributions. Another important question is how small biological precursor molecules could be adsorbed at the surface of the primordial ocean during the evolution of biomolecules, and, at the water surface, which associates of them in which relative alignment could have possibly led to further steps of biomolecular evolution. Concerning lipid membranes the basic question is how anesthetics alter the properties of lipid membranes, which membrane properties could, and which of them could not be behind the molecular mechanism of anesthesia. In the planned investigation of solid interfaces the basic question is how much the properties (orientation adsorbate-adsorbent and lateral interactions, in particular, H-bonding) of the adsorption layer change in case of multicomponent adsorption, and how selective is the adsorption of given components at the ice surface. Selectivity is also the main question of the planned work concerning incorporation in clathrate hydrates. Finally, the planned project includes several methodological developments. The main aim here is evidently the development of computationally feasible methods for selected tasks; these methods would provide the basis of further research in the future.

What is the significance of the research?
Describe the new perspectives opened by the results achieved, including the scientific basics of potential societal applications. Please describe the unique strengths of your proposal in comparison to your domestic and international competitors in the given field.

The planned research project contains primarily pure scientific investigations; nevertheless, we believe that some of the results might be used later on in applied research, as well. We plan a methodological development and writing a corresponding computer code, namely that for the calculation of lateral pressure profiles at interfaces. This work is currently already in progress, but its completion is only expected in the frame of this project. Several investigations planned in the frame of this project are based on the success of this work. Problems of purely scientific nature are the calculation of the contributions of the subsequent subsurface layers to various thermodynamic quantities, in particular, to the surface tension at the liquid surface, the investigations concerning prebiotic evolution and clathrate hydrates. The former one could lead to a deeper understanding of the nature and molecular origin of surface tension, and its dependence on the intermolecular interactions, investigation of prebiotic evolution might provide a step towards the understanding of the origin of life, whilst the planned work on clathrate hydrates might shed some light to the molecular level details of the formation of the Solar System. The remaining tasks, although focusing also on pure scientific questions, might lead to results that can be used later on in applied research, in particular, in the field of environmental or biomedical science. The systematic investigation of the surface of room temperature ionic liquids might help in selecting the proper member of this promising, “green” family of solvents for specific purposes; our research on anesthetics and opioids in membranes might help in understanding the molecular mechanisms of the effect of these molecules, which are widely used in the medical practice but unfortunately also often abused, on the living cells; results concerning the magnetite surface might be used later on by biomedical investigations of the functionalized magnetite nanoparticles; whilst the planned studies on single and competitive adsorption at the ice surface might constitute the scientific basis of environmental/atmospheric chemistry oriented applied research in the future.

Summary and aims of the research for the public
Describe here the major aims of the research for an audience with average background information. This summary is especially important for NRDI Office in order to inform decision-makers, media, and others.

The character of the planned project is of pure scientific research, which means its results are expected to lead to a deeper understanding of processes occurring at the interface of different phases rather than to the development of new inventions to be used in everyday life. However, such processes are frequently related to everyday life problems, thus, we expect that the results of the present project might be later on the basis of further investigations of applied science, aiming now indeed the development of new applications. Adsorption of small air pollutant molecules at the surface of ice is a process occurring at the higher regions of the atmosphere, and leads to various environmental damages (e.g., destruction of the ozone layer). Results of our research in this direction are thus expected to have environmental applications in the future. Tiny magnetic particles of magnetite suspended in aqueous environment constitute a system with very promising biomedical applications: such particles, and also drug molecules attached to their surface can be directed to desired positions in the body even with an external magnet. However, to avoid, e.g., the risk of thrombosis, applicability of such particles requires sufficient coating of their surface to avoid aggregation. The corresponding part of this project aims at the investigation of such coatings. Among our investigations of purely scientific character the ones concerning biological precursors and clathrate hydrates might shed some light to the molecular level details of the origin of life and that of the formation of the Solar System.

Munkánk során elsősorban biológiai illetve környezeti szempontból érdekes jelenségek molekuláris hátterét próbáltuk felderíteni. Vizsgáltuk különböző folyadékok felületének molekuláris rétegeit, egy sor kis légköri szennyező molekula megkötődését jégszemcséken a magas légkörben uralkodó körülmények között, a csillagközi térben jelen lévő, az élet keletkezéséhez elengedhetetlen biomolekulák építőköveinek ottani keletkezésében lehetséges szerepet játszó kis molekulák megkötődését jégfelületen illetve csapdázódását klatrátokban a csillagközi térre jellemző körülmények között, altató hatású molekulák elrendeződését lipid membránokban, valamint lehetséges új, környezetbarát, „zöld” ipari oldószereket, elsősorban szobahőmérsékletű ionos folyadékokat. Munkánk során kifejlesztettünk egy új módszert amivel folyadékok felületén az oldalirányú nyomás változása, és ezáltal az egyes részecskék hozzájárulása a felületi feszültséghez számítható. Megmutattuk, hogy vannak olyan vízzel csak részlegesen elegyedő anyagok, melyek maguktól beoldódnának a vizes fázisba, a korlátozott elegyedés ilyenkor kizárólag a vízmolekuláknak tudható be, illetve hogy ionos felületaktív anyagok vizes oldatában az egyszerű ellenionok alapvetően fontos módon járulnak hozzá a felületi feszültség kialakításához. Megmutattuk, hogy a műtéti altatók hatásukat a sejtmembrán poláros és apoláros részének határán, az oldalirányú nyomás megváltoztatásával fejtik ki.

In the frame of this project we tried to shed light to the molecular reasons laying behind biologically or environmentally relevant processes. We investigated the molecular layers at various liquids surfaces, adsorption of a number of atmospheric pollutants at the surface of ice grains under tropospheric conditions, adsorption of small molecules, playing a potential role in the interstellar formation of building blocks of biomacromolecules, on ice surfaces and their inclusion in clathrates under interstellar conditions, the arrangement of anesthetic molecules in lipid membranes, and potential new, environmentally friendly, “green” industrial solvents, first of all, room temperature ionic liquids. We developed a new method for calculating the lateral pressure profile at liquid surfaces, enabling us to calculate the surface tension contribution of the individual particles. We showed that certain molecules, being only partially miscible with water exhibit a tendency for being dissolved in the aqueous phase, and partial miscibility can solely be attributed to the water molecules. We demonstrated the key role counterions play in determining the surface tension of aqueous solutions of ionic surfactants. We showed that the phenomenon of anesthesia is caused by the anesthetics accumulated at the boundary of the apolar and polar parts of the membrane, and the molecular mechanism of anesthesia is related to their ability of altering the lateral pressure profile across the membrane.
