A reaktív oxigén származékoknak (ROS) számos jelentős élettani folyamatban van szerepük , mint az immunvédekezés, az értónus szabályozás, hormonszintézis, extracelluláris mátrix kialakítása és a megtermékenyítés. Bár különböző biokémiai folyamatok melléktermékeként is képződnek, szabályozott termelésüket jelenleg elsősorban a NADPH-oxidázok Nox/Duox családjának tulajdonítjuk. Ezek az enzimek a legkülönbözőbb szövetekben és szervekben is megtalálhatók, jelezve, hogy a ROS termelése élettani körülmények között sokkal szélesebb körű, mint korábban gondoltuk. Emberben az enzimcsaládnak hét tagja van: Nox1-5 és Duox1 ill. Duox2. Jelen pályázat témája a Nox5 enzim, mely nagymértékben expresszálódik a herében és a géntermék jelen van a spermiumokban is. Egy Nox enzim kifejeződése a spermiumban arra utal, hogy a ROS-nak fontos szerepe lehet az emberi megtermékenyítés során. Ugyanakkor megfelelő genetikai modell hiányában a ROS termelés jelentősége és a ROS hatásai nem vizsgálhatók. A spermiumokon kívül a Nox5 más sejttípusokban, mint az endotélsejtek és érfali simaizomsejtek, is megtalálható, ami a Nox5 által termelt ROS értónusszabályozásban játszott esetleges szerepére utal. A Nox5 gén nincs jelen rágcsálókban, de más emlősök termelik az enzimet, mint például a nyúl is, melyet kísérleti modellül választottunk. A Mezőgazdasági Biotechnológiai Kutatóközpont (MBK) kutatóival együttműködésben a nyúl Nox5 génben a nemrégen felfedezett CRISPR/Cas9 technológia segítségével hoztunk létre mutációt. Jelen munka célja a mutáns nyúl törzsek jellemzése és felhasználása a Nox5 spermiumokban és érfalban játszott szerepének tisztázására.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

A Nox/Duox enzimek részvételét több élettani folyamatban, mint az immunvédekezés, pajzsmirigy hormon termelés, a belsőfül otolith kristályainak képzése, genetikai bizonyítékok támasztják alá. A Nox5 expressziós mintázata, valamint irodalmi adatok és előzetes eredményeink alapján feltételezzük, hogy a Nox5 fontos szerepet játszik az emberi megtermékenyítésben. A ROS meghatározó szerepét már leírták alacsonyabb rendű élőlények, mint például a tengeri sün megtermékenyítési folyamatában, de egy specifikus ROS forrás jelenléte az emberi spermiumokban arra enged következtetni, hogy a megtermékenyítés ROS függése sokkal szélesebb körű, mint korábban hittük. A génhiányos egérmodellek alapvetőek voltak a Nox/Duox enzimek élettani és kórtani jelentőségének feltárásában, de a Nox5 hiánya az egér genomjában kizárja annak lehetőségét, hogy a Nox5 szerepét génhiányos egereken vizsgálhassuk. Meggyőződésünk, hogy egy Nox5 hiányos nyúl modell segítségével feltárhatjuk a Nox5 megtermékenyítésben játszott szerepét. Továbbá ez az állatkísérletes rendszer segíthet a Nox5 más sejtekben, mint az endotélsejtek és az érfal simaizom sejtjei, betöltött feladatának megértésében is. Az érrendszer sejtjei többféle Nox enzimet is kifejeznek, ezek között a Nox5-öt is. Mivel a Nox5 egy kálcium ionok által szabályozott enzim, aktivitása fontos kapocs lehet a sejten belüli kálcium szint és a ROS által szabáyozott jelátvitel között.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

A reaktív oxigénszármazékok (ROS) szabályozott termelését ma már a homeosztázis alapvető folyamatának tartjuk. Fagocitákban a ROS termelés hiánya súlyos immunhiány kialakulásához (krónikus granulomatózis, CGD) vezet, míg a pajzsmirigyben a ROS termelés hibája felelős a hipotireózis egyes formáiért. Mivel a Nox5 enzim a spermiumokban van jelen nagy mennyiségben, joggal feltételezhetjük, hogy az emberi fertilizációban lényeges feladatot lát el. Tekintve, hogy az emberi és nyúl Nox5 enzimek igen nagy mértékben hasonlóak, valószínűleg a spermiumokban játszott szerepük is hasonló. Ennek felderítése segíthet az emberi megtermékenyítés folyamatának jobb megértésében, amit egyébként – etikai okokból – nem lehet kísérletesen vizsgálni. Lehetséges, hogy ezen enzim működésének hibája felelős a férfi meddőség bizonyos formáiért. Ha feltevésünk helyes és a Nox5 termelte ROS alapvető a megtermékenyítés folyamatában, akkor érdemes vizsgálni, hogy az antioxidánsok korlátlan mértékű fogyasztása nem zavarhatja-e meg a termékenyítő képességet a Nox5 hatásának gátlásán keresztül.
A Nox5 szerepének megértése nagy jelentőséggel bírna az érrendszer vonatkozásában is, ugyanis a Nox enzimek által termelt ROS-nak mind előnyös, mind hátrányos hatásait is leírták már az érfalban, azonban az egér modellekből származó kísérleti adatok a Nox5-ről nem szolgáltathatnak információt.
A Nox/Duox kutatási terület igen kompetitív, a Nox4 enzim (amit 2000-ben azonosítottam) jelenleg a legintenzívebben tanulmányozott izoforma. A Nox5-ről közölt publikációk száma jóval csekélyebb, ami valószínűleg a génhiányos állatmodell hiányára vezethető vissza, ilyen modell minden más Nox/Duox enzimre elérhető ma már. Bősze Zsuzsanna munkacsoportja (MBK) régóta folytat tanulmányokat nyúl transzgenikus technikákkal, ezért a pályázatban leírt együttműködésünk révén egy egyedülállóan innovatív, újszerű megközelítésből tanulmányozhatjuk a Nox5 működését.

Szervezetünkben vannak enzimek, amelyek oxigéntartalmú, igen reakcióképes anyagokat állítanak elő oxigénből. Ezek a vegyületek, melyeket reaktív oxigén származékoknak (ROS) hívunk számos fontos élettani szereppel bírnak. Például a fehérvérsejtjeink ROS-t állítanak elő, amivel baktériumokat és gombákat képesek megölni, a pajzsmirigyünk pedig szintén reaktív oxigén származékokat használ a pjzsmirigy hormon előállításának folyamatában. Érdekes módon a spermiumok is képesek szabályozott ROS termelésre egy Nox5 nevű enzim révén, de ennek szerepe, jelentősége még nem világos. Lehetséges, hogy a ROS termelés valamilyen zavara az oka a férfi meddőség bizonyos formáinak. Kísérleteinkben ezt próbáljuk felderíteni egy Nox5 génhiányos nyúl állatmodell segítségével. A spermium nem az egyetlen sejttípus, ami tartalmaz Nox5-öt, hanem például az ereket bélelő endotélsejtekben is jelen van az enzim, ezért vizsgálni fogjuk a Nox5 erekben betöltött szerepét is.

Reactive oxygen species (ROS) have several important physiological functions, including a role in host defense, vasoregulation, hormone synthesis, extracellular matrix formation and fertilization. These compounds are produced in various biochemical pathways as side products, but the regulated production of ROS is now mainly attributed to members of the Nox/Duox family of NADPH oxidases. These enzymes are expressed in a wide variety of organs and tissues, indicating that the physiological production of ROS is more widespread than previously thought. In humans, there are seven members of the enzyme family: Nox1-5, Duox1, and Duox2. The subject of the current grant application is Nox5 which is highly expressed in testis and the gene product functions in sperm cells. The expression of a Nox enzyme in spermatozoa suggests that ROS play an important role in human fertilization, however in the absence of an appropriate genetic model the significance of ROS production and their mechanism of action can not be explored. Beside sperm cells, Nox5 is also expressed in other cell types including endothelial cells and vascular smooth muscle, implicating Nox5-derived ROS in vasoregulation. Nox5 is absent in rodents, several other mammals express the enzyme, including rabbits that we chose as the experimental model to be studied. In collaboration with researchers at the NARIC-Agricultural Biotechnology Institute we introduced mutations into the rabbit Nox5 gene by using the recently discovered CRISPR/Cas9 technique. The aim of the current study is to characterize these mutant rabbit lines and use them to delineate the function of Nox5 in spermatozoa and cells of the vasculature.

What is the major research question?

Involvement of Nox/Duox enzymes is now supported by genetic evidence in several physiological settings, including host defense, thyroid hormone synthesis and the biosynthesis of otoconia. Based on the expression pattern of Nox5, existing literature and preliminary data we hypothesize the Nox5 plays an important role in human fertilization. ROS have already been described to have a significant role in the fertilization of lower species such as sea urchin, but the presence of a dedicated ROS source in human sperm cells suggest that the ROS dependence of fertilization could be more widespread than previously thought. Knockout mouse models were instrumental to explore the role of Nox/Duox enzymes in health and disease, but the absence of Nox5 in the mouse genome excludes the possibility of studying the functions of Nox5 with the help of knockout mouse models. We believe that by using Nox5-deficient rabbits we can find out the role of Nox5 in fertilization. Furthermore, this novel animal model might help us to understand the enzyme's function in other cell types including endothelial cells and vascular smooth muscle. Cells of the vasculature express several different Nox enzymes, including Nox5. Since Nox5 is a calcium-regulated enzyme, its activity might represent an important link between calcium and ROS signaling.

What is the significance of the research?

Regulated production of reactive oxygen species (ROS) is now considered an essential part of homeostasis. The absence of ROS production in phagocytes leads to the development of a serious immunodeficiency, chronic granulomatous disease and deficient ROS production in the thyroid is responsible for some forms of hypothyroidism. Since Nox5 is highly expressed in human spermatozoa we can reasonably assume that the enzyme plays a significant role in human fertilization. Since the rabbit and human Nox5 enzymes are highly similar to each other, they likely have a similar role in sperm cells. Uncovering this role will help us to get a better understanding of human fertilization which - due to ethical reasons - can not be experimentally approached. It is possible that dysfunction of Nox5 enzyme is responsible for some forms of male infertility. If our hypothesis is correct, and Nox5-derived ROS do have a major role in the fertilization process, then it should be explored whether the uncontrolled consumption of antioxidants might disturb the fertilization process through the inhibition of Nox5 action. Understanding the role of Nox5 in vascular cells would also be of great significance because Nox-derived ROS were described to have both beneficial and harmful effects in blood vessels, however the data about the role of Noxes in the vessel wall was derived from mouse models, where Nox5 is not part of the vascular Nox repertoire.
The Nox/Duox research field is highly competitive and today Nox4 (which I have identified in 2000) is the most intensively studied Nox isoform. Publications about Nox5 are more scarce which is probably due to the lack of knockout models that now exist for all other Nox/Duox enzymes. The group of Zsuzsanna Bősze has a long-standing interest in rabbit transgenesis, thus the collaboration outlined in research proposal will put us in a unique position to study the function of Nox5 in an innovative, previously unprecedented way.

There are enzymes in our body which make highly reactive, oxygen-containing substances from oxygen. These compounds, called reactive oxygen species (ROS) have several important physiological functions. For example our white blood cells make ROS to kill bacteria and fungi, and the thyroid gland also uses these molecules in hormone synthesis. Interestingly, sperm cells also have the ability to make ROS in a regulated manner by an enzyme called Nox5, however the importance this process is unclear. It is possible that disturbed production of ROS plays a role in male infertility. In our experiments we intend to find out the role of ROS in sperm cells, by using a genetically modified rabbit model. Sperm cells are not the only cell types that contain Nox5, therefore we will also study the function of Nox5 in cells of the vessel wall.

Emberben a NOX/DUOX enzimek családjának összesen hét tagja van (NOX1-5, DUOX1,2). Ezek a fehérjék változatos biológiai funkciókkal rendelkeznek, fontos szerepet játszanak az immunvédekezésben, hormon bioszintézisben és vaszkuláris funkciók szabályozásában. A NOX/DUOX enzimcsalád legtöbb tagja egérben is megtalálható, így a különböző génhiányos egereknek fontos szerepük van a NOX/DUX funkciók azonosításában és az enzimek betegségek kialakulásában játszott szerepének vizsgálatában. A NOX5 enzim azonban hiányzik rágcsálókban, ezért az enzim funkciója nem vizsgálható KO egerek segítségével. Mivel nyulakban megtalálható a NOX5 enzim, ezért NOX5-hiányos nyúlmodellt hoztunk létre és annak segítségével vizsgáltuk a NOX5 funkcióját. Kimutattuk hogy a NOX5 nem nélkülözhetetlen a megtermékenyítés folyamatában és a NOX5 genetikai hiánya nem okoz súlyos fenotípusos változásokat. A NOX5 génhiányos nyulakon végzett kísérleteinkben felfedeztük, hogy a NOX5 véd az atherosclerosis kialakulásával szemben. Eredményeink arra utalnak, hogy a NOX/DUOX enzimek által termelt reaktív oxigén származékok korábban ismeretlen, kedvező élettani hatásokkal is rendelkezhetnek.

Members of the NOX/DUOX family of NADPH oxidases are primarily responsible for the regulated production of reactive oxygen species in living organisms. In humans, there are seven members of the family. These enzymes have a variety of biological functions, including an important role in immune defense, hormone biosynthesis, and regulation of vascular function. Most members of the NOX /DUOX family are also found in mice, so various gene-deficient mice models play an important role in identifying NOX/DUOX functions and investigating the role of enzymes in disease development. However, the NOX5 enzyme is absent in rodents, so the function of the enzyme cannot be tested in KO mice. Because the NOX5 enzyme is found in rabbits, a NOX5-deficient rabbit model was developed and used to study the function of NOX5. We have shown that NOX5 is not essential in the fertilization process and NOX5 deficiency does not cause severe phenotypic changes. In our experiments on NOX5 gene-deficient rabbits, we discovered that NOX5 protects against the development of atherosclerosis. Our results suggest that reactive oxygen species produced by NOX/DUOX enzymes may have previously unknown beneficial physiological effects.
