A kutatás összefoglalója, célkitűzései szakemberek számára

Az agykérgi idegsejteket az extracellulárisan regisztrált akciós potenciál (AP) hullámformájuk alapján jelenleg serkentő principális sejtekre és gátló interneuronokra tudjuk csoportosítani. A közelmúltban kifejlesztett sokcsatornás, nagy téri felbontású elektródokkal azonban lehetőség nyílt a neuronok AP hullámformáinak nagyobb térbeli mintavételezésére. A kutatás fő célkitűzése az agykéregben megtalálható idegsejt-típusok elektrofiziológiai azonosítása a kétdimenziós AP hullámformájuk alapján. Ehhez először nagy téri és idői felbontással regisztráljuk nagyszámú neuron AP hullámformáját altatott rágcsálókban, majd az AP hullámformák jellegzetes térbeli és időbeli tulajdonságait megvizsgálva kísérletet teszünk az idegsejtek osztályozására. Céljaink között szerepel szimultán juxtacelluláris és extracelluláris mérések végrehajtása, mely az osztályozás támogatásához járulna hozzá a megjelölt neuronok morfológiai tulajdonságainak és az extracellulárisan detektált AP hullámformáinak összevetésével. További célunk egy automatikus osztályozó algoritmus kifejlesztése, melyhez képfeldolgozási módszereket és a mesterséges intelligencia területén alkalmazott algoritmusokat kívánunk felhasználni. Az osztályozott neuronokat és az azokhoz tartozó kétdimenziós AP hullámformákat egy szabadon hozzáférhető adatbázisban tervezzük eltárolni és megosztani a nemzetközi idegtudományi közösséggel. Mivel nemzetközi viszonylatban is kevés nagy téri felbontási elektrofiziológiai mérési adat érhető el, így az adatok megosztásának nemzetközi jelentősége várhatóan nagy lehet. Továbbá eredményeink hozzájárulhatnak az agykérgi neuronhálózatokat alkotó idegsejtek szerepének jobb megértéséhez is.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Az agykérgi idegsejteket az extracellulárisan regisztrált akciós potenciál (AP) hullámformájuk alapján jelenleg serkentő principális sejtekre és gátló interneuronokra tudjuk csoportosítani, melyhez leggyakrabban az adott sejthez tartozó AP hullámforma egy csatornán számított szélességét használják. Az AP-ok sokcsatornás elektródokkal történő nagyobb térbeli mintavételezése révén azonban lehetőség nyílik a megnövekedett térbeli információ kiaknázására. Előzetes kísérleti eredményeink azt mutatják, hogy az interneuronokra jellemző keskeny AP-ok kétdimenziós hullámformája jelentősen eltér a piramissejtekre jellemző széles AP-ok kétdimenziós hullámformájától. Utóbbi esetben például megfigyelhető az AP visszaterjedése a sejttestből a dendritfába. Az infragranuláris kérgi rétegekben megfigyeltünk olyan idegsejteket is melyeknél az AP visszaterjedési iránya fordított volt. Az ezektől a neuronoktól eredő AP-ok feltehetőleg az infragranuláris rétegekben található fordított piramissejtektől származhatnak. A kutatás során a kétdimenziós AP hullámformák megvizsgálásával tehát arra a kérdésre keressük a választ, hogy a kiegészítő térbeli információ felhasználható-e a két fő sejttípuson belül további altípusok elektrofiziológiai azonosítására. Továbbá a kísérleteink során azt is meg kívánjuk vizsgálni, hogy az AP hullámformának melyek azok a téridőbeli tulajdonságai mellyel a legnagyobb megbízhatósággal azonosíthatók az egyes idegsejtcsoportok az agykéregben. A fenti kérdések megválaszolásához többféle kísérleti módszert (extracelluláris mérés nagy téri felbontású elektródokkal, szimultán juxta- és extacelluláris mérés, optogenetikai kísérletek) és elemző metódust fogunk alkalmazni.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

A kutatás során arra a kérdésre keressük a választ, hogy az agykérgi idegsejtek nagy téri felbontással regisztrált kétdimenziós akciós potenciál hullámformáiból ki tudunk-e nyerni olyan téridőbeli információkat, melyek felhasználásával nagy megbízhatósággal meghatározható az adott idegsejt típusa. Ezzel a módszerrel lehetőségünk nyílna a neuronok elektrofiziológiai jeleik alapján történő megfelelő osztályozására anélkül, hogy összetett intracelluláris vagy juxtacelluláris jelölő módszerek alkalmazásával, majd hosszadalmas szövettani vizsgálatokkal, utólagosan derülne ki azok pontos típusa. A neuronok elektrofiziológiai azonosítása például in vivo humán vizsgálatokban jelentős előnnyel járna, ugyanis ezekben az esetekben a sejtek megjelölése csupán az esetlegesen eltávolított agyszövet in vitro körülmények között történő vizsgálata során lehetséges. Az idegsejtek megbízható elektrofiziológia-alapú azonosításának megvalósulása esetén az egy kísérletben megfelelően csoportosított neuronok száma növekedne, így akár csökkenthető lenne a tanulmányokhoz szükséges kísérleti állatok száma. Továbbá jelentős költséget és időt lehet megtakarítani azzal is, hogy nem lenne szükséges az egyes neuronokat speciális jelölőanyaggal külön megjelölni, majd a rekonstruált morfológiájuk alapján a típusukat utólag meghatározni. A kutatás során kifejlesztésre kerülő automatikus osztályozó algoritmus egy továbbfejlesztett változata a későbbiekben akár alkalmas lehet az idegsejtek típusának már a kísérletek során, valós időben történő meghatározására, ami új távlatokat nyithatna az idegrendszeri kutatásokban. A kutatás eredményessége esetén jelentősen felgyorsulhatna az egyes idegsejt-típusok különböző fiziológiás és patológiás agyi folyamatokban betöltött szerepének részletesebb megértése is. A kutatás során mérési adatainkat és eredményeinket egy szabadon hozzáférhető adatbázisban tervezzük közzétenni, ugyanis jelenleg kevés hazai és nemzetközi kutatócsoport rendelkezik ilyen, nagy téri felbontású elektrofiziológiai méréseket lehetővé tevő eszközökkel és még kevesebb ilyen típusú adat érhető el szabadon. Az adatok megosztása hasznos, új kutatási eredményekhez és más kutatócsoportokkal való együttműködésekhez is vezethetne a jövőben.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

Az emlősök agykérge nagyszámú idegsejtből épül fel, melyek különböző alaktani, elektromos és kémiai tulajdonságaik alapján többféle, akár több tucat különböző típusba sorolhatók. Ezeket az idegsejteket elektromos impulzusaik vizsgálatával jelenleg két fő csoportba lehet besorolni: az idegi serkentést és gátlást megvalósító neuronok csoportjába. A technológiai fejlődésnek köszönhetően az elmúlt évek során olyan agyi érzékelőket fejlesztettek ki, melyek nagyszámú, egymáshoz közel elhelyezkedő, kisméretű mérőponttal rendelkeznek. Ezekkel az eszközökkel a kutatók a korábbiakhoz képest jelentősen nagyobb téri felbontással tudják rögzíteni az egyes idegsejtek elektromos tevékenységét. A kutatásunk célja megvizsgálni, hogy a nagyobb téri felbontással regisztrált elektromos impulzusok alkalmasak-e a két fő idegsejt-csoporton belül további altípusok elkülönítésére. Ennek a kérdésnek az eldöntésére altatott rágcsálókból nagyszámú idegsejt elektromos tevékenységét tervezzük rögzíteni, majd a mérési adatokat különböző, pl. a mesterséges intelligencia területén is alkalmazott, módszerekkel elemezni. Az idegsejteket egy speciális anyaggal megjelölve összevethetjük azok alaktani tulajdonságait a nagy téri felbontású elektromos impulzusaikkal, melyekből további hasznos információkat szűrhetünk le. További terveink között szerepel a mérési adatokból egy, az interneten elérhető, szabadon hozzáférhető adatbázis elkészítése is. A kutatás eredményei a jövőben hozzájárulhatnak majd az agyi hálózatokat felépítő egyes idegsejt-típusok különféle agyi folyamatokban betöltött szerepének részletesebb megismeréséhez, valamint az agyat érintő megbetegedések kialakulásának mélyebb megértéséhez is.

Summary of the research and its aims for experts

Currently, based on their extracellular action potential (AP) waveforms, cortical neurons can be grouped into excitatory principal cells and inhibitory interneurons. However, with multisite, high-density probes developed recently it became possible to sample the AP waveforms of neurons with higher spatial resolution. The main objective of this study is the electrophysiological identification of different types of cortical neurons based on their two-dimensional AP waveforms. To achieve this, we will first record the AP waveform of a large number of neurons with high spatial and temporal resolution in anesthetized rodents, then we will attempt to classify these neurons after examining characteristic spatial and temporal features of their AP waveforms. Furthermore, we plan to perform simultaneous juxtacellular and extracellular recordings, which would support the classification process by comparing the morphological attributes of labeled neurons with their extracellular AP waveforms. We also aim to develop an automatic classification algorithm in which we plan to incorporate different image processing methods and algorithms used in the field of artificial intelligence. We plan to store the two-dimensional AP waveforms of the identified neurons in a freely accessible database and to share this database with the international neuroscience community. The amount of electrophysiological data recorded with high-spatial density which can be accessed online is scarce, therefore sharing our data might have a significant international impact. Furthermore, our results could contribute to a better understanding of the role of different neuron types in cortical circuits.

What is the major research question?
Describe here briefly the problem to be solved by the research, the starting hypothesis, and the questions addressed by the experiments.

Currently, based on their extracellular action potential (AP) waveforms, cortical neurons can be grouped into excitatory principal cells and inhibitory interneurons, which is most often achieved by calculating the width of the AP waveform of the particular neuron on a single recording channel. However, by the higher spatial sampling of APs with multisite, high-density probes we can exploit the increased spatial information content. Our preliminary results show, that the two-dimensional waveforms of narrow spikes characteristic of interneurons differ significantly from the 2D waveforms of wide spikes characteristic of pyramidal cells. For example, in the latter case the backpropagation of the AP can be observed from the soma to the dendritic tree of the neuron. In the infragranular cortical layers we could also observe neurons which showed an AP backpropagation but with inversed direction. Spikes of these neurons could originate from inverted pyramidal cells found in infragranular layers. During the research we plan to answer the following question by examining the two-dimensional AP waveforms: can we use the higher spatial resolution to electrophysiologically identify different subtypes of neurons within the two main types? Furthermore, during the study, we plan to investigate which spatiotemporal properties of the AP waveforms are the most suited to identify different types of cortical neurons with the highest reliability. To answer these questions, we plan to apply various experimental techniques (extracellular recordings with high-density probes, simultaneous juxtacellular and extracellular recordings, optogenetic experiments) and data analysis methods.

What is the significance of the research?

During the study, we plan to answer the question whether it is possible to extract suitable spatiotemporal features from the two-dimensional action potential waveform of cortical neurons recorded with high spatial density which might be used to reliably determine the type of the particular neuron. Using these features it would be possible to classify neurons based solely on their electrophysiological signatures, without the need to use complex intracellular or juxtacellular labeling techniques and wearisome post hoc histological procedures to identify the type of neurons. Electrophysiological identification of neurons could have a significant advantage in human in vivo studies, because labeling of neurons in human experiments is only allowed in the pieces of brain tissue removed for subsequent in vitro examinations. In case it would be possible to reliably identify neurons based on their electrophysiological signals, then the number of properly classified neurons in a single experiment would increase. Therefore, the number of animals used in a research study could be decreased. Furthermore, a significant amount of cost and time could be saved if there would be no need to label individual neurons with a special neuroanatomic tracer and to determine their type subsequently based on their reconstructed morphology. In the future, an improved version of the developed automatic classification algorithm might be used to identify the type of neurons real-time, during experiments, which could open up new possibilities in neuroscience research. The deeper understanding of the role of different types of neurons during various physiological and pathological neural mechanisms could also be sped up in case the results of this study prove to be successful. We plan to share our recordings and findings through a freely accessible database because currently, only a few national and international research groups have neural probes which allow the recording of electrophysiological data with high spatial density. Furthermore, the amount of freely accessible high-density data is even lower. Sharing our data could also lead to novel and useful research findings and to collaborations with other research groups.

The cerebral cortex of mammals is made up of a large number of neurons which can be categorized into many, even dozens of different types based on their morphological, electrical and chemical properties. Currently, by the examination of their electrical impulses, these neurons can be classified into two main groups: the group of excitatory and inhibitory neurons. In recent years, thanks to the technological advancement, brain sensors containing a large number of closely spaced, small recording sites were developed. Using these devices researchers are able to record the electrical activity of neurons with a significantly higher spatial resolution. The objective of our study is to examine whether the electrical impulses recorded with higher spatial resolution are suitable to identify different subtypes of neurons within the two main types. To answer this question, we plan to record the electrical activity of a large number of neurons in anesthetized rodents, followed by the analysis of these recordings with various methods, including techniques used in the field of artificial intelligence. Furthermore, the morphological attributes of neurons labeled with a special substance will be compared with their electrical impulses recorded with high spatial resolution, which might provide additional useful information for the study. We also plan to build an online, freely accessible database from our high-density recordings. In the future, the results of the study could significantly contribute to the deeper understanding of the role of different types of cortical neurons in various neural mechanisms, including the development of brain diseases.

(2) Több ezer agykérgi idegsejt elektromos tevékenysége, illetve nagy téri felbontású akciós potenciál hullámformája került kinyerésre a rögzített adatokból. (3) A nagy téri felbontású adatok egy jelentős részét egy szabadon elérhető adatrepozitóriumban nyilvánosan megosztottuk. (4) A nagy téri felbontású agykérgi méréseket felhasználva mesterséges intelligencia alapú, illetve programozható logikai áramköröket tartalmazó eszközre (FPGA) sejtválogató algoritmusokat fejlesztettünk. (5) Kimutattuk, hogy a szilícium alapú elektródok lassú sebességgel történő beszúrása az agyszövetbe jelentősen jobb minőségű mért jeleket eredményez a gyorsabb beszúrási sebességekkel történt implantáláshoz képest. (6) Kimutattuk, hogy a szilícium alapú elektródok szélén található mérőpontokkal szignifikánsan jobb minőségű extracelluláris jelek mérhetők, mint az elektródok közepén található mérőpontokkal. (7) A kutatás eredményeiből eddig 11, lektorált tudományos folyóiratban megjelent publikáció, illetve több diplomamunka született.

(1) I recorded cortical activity in vivo from anesthetized rats and mice, as well as in vitro from brain slices using silicon-based probes able to record brain electrical activity with high spatial resolution. (2) The electrical activity as well as the high-spatial-resolution action potential waveform of thousands of cortical neurons were extracted from the collected dataset. (3) A large fraction of the high-resolution dataset was shared in a public data repository. (4) We developed artificial intelligence-based and Field Programmable Gate Array(FPGA)-based spike sorting algorithms with the significant involvement of the collected high-spatial-resolution cortical recordings in the development and testing process. (5) We have found that inserting silicon-based probes with a very slow speed into the brain tissue will provide neural signals with a significantly better quality than implantations with faster insertion speeds. (6) We have shown that recording sites located at the edge of the shaft of silicon-based probes provide extracellular signals with a significantly better quality compared to recording sites located in the center of the silicon shaft. (7) In total, eleven articles were published in peer-reviewed scientific journals based on the results of the research project, and several dipoma theses were finished as well.
