A projekt keretében látogatni tervezett nemzetközi tudományos/innovációs rendezvény bemutatása és a tervezett részvétel ismertetése
Mutassa be a nemzetközi rendezvény jellegét, célját, szervezőit és jelentőségét a nemzetközi tudományos életben. Ismertesse a részvétel célját (pl. előadás, poszter, egyéb aktivitás), a rendezvényen való részvétel várható hatását és jelentőségét a saját kutatói karrier építésében.

Szakterületünk a vér-agy gát patológia és farmakológia európai szinten legfontosabb rendezvénye a 24th International Symposium on Signal Transduction at the Blood-Brain Barriers konferencia. Ez egy rendszeresen, évente megrendezésre kerülő konferenciasorozat. Minden országból a témával foglalkozó legfontosabb kutatócsoportok beszélnek a legfrissebb kutatási eredményeikről. Az elmúlt években a pandémia miatt több alkalommal elmaradt a konferencia. Idén egy hasonló meeting online került megrendezésre, melyen poszter prezentációval vettem részt. 2022-ben Olaszországban már az élőben megtartandó konferenciára készülünk. A legfrissebb tudományos eredmények megosztásában és megtárgyalásában rendkívül fontos ez a nemzetközi esemény a tudományterületünk számára. Lehetőségem van a kutatási területünk legkiemelkedőbb kutatóival találkozni.

Poszter prezentációval tervezek részt venni ezen a nemzetközi konferencián.

A konferencián való részvétel jó kapcsolatépítési lehetőséget biztosít számomra a kortárs PhD hallgatókkal, postdocokkal és a szakterület legkiválóbb kutatóival. A rendezvény új együttműködések kialakítására ad lehetőséget a vér-agy gát témakörében jártas szakemberekkel. A konferenciákon előadott szóbeli és poszter prezentációk segítik megismerni a legújabb tudományos eredményeket és megismerni a jelenleg a kutatókat foglalkoztató tématerületeket. A saját poszter prezentáció elősegíti a tudományos előmenetelemet és fejleszti az előadó készségemet. A pandémiás időszak után jó lehetőség, hogy személyesen is megismerkedjünk a témán dolgozó kutatókkal és a legfrissebb kutatási eredményeinket megoszthassuk egymás között.

A projekt összefoglalója, célkitűzései laikusok számára
Adjon egy rövid, közérthető ismertetést a látogatni tervezett nemzetközi rendezvényről, illetve a részvétel céljáról, várható eredményeiről.

A vér-agy gát funkcionális alapját az agyi endotélsejtek alkotják, ez az agy egyik legfontosabb védelmezője, és létfontosságú az agyszövet oxigén- és tápanyagellátásának fenntartásában is. Azonban az agyi endotélsejtek számos módon képesek megakadályozni a hatóanyagok agyba való bejutását: speciális (efflux) transzporterekkel, szoros sejtközötti kapcsolatokkal és gátolt nem specifikus endocitotikus útvonallal. A modern gyógyszerfejlesztés során a gyógyszerhatóanyagok agyba való bejutásának és toxicitásának tesztelése elengedhetetlen sejttenyészetek segítségével (Veszelka és mtsai, 2011). A gyógyszerkutatásban a legújabb trend a hatóanyagok agyi szferoidokon és organoidokon történő tesztelése, amelyet az indukált pluripotens őssejt-alapú technikák elterjedése tesz lehetővé, amely mára már egy új kutatási irányt jelent (Choudhury és mtsai, 2020). Jelenleg az agyi gyógyszerbejuttatás legelterjedtebb vizsgálata 2D ko-kultúrákkal történik, amely során a sejttenyészeteket egy permeábilis membránra tenyésztik monokultúrában, vagy ko-kultúrában több sejttel (Nakagawa és mtsai, 2009; Veszelka és mtsai, 2011; Helms és mtsai, 2016). A gyógyszerkutatásban egyre fontosabbá válnak a humán sejtes modellek. Csoportunk széleskörű tapasztalattal rendelkezik sejttenyésztő inzerten létrehozott, egy vagy több sejttípusból álló epitél és endotél-alapú barrier-ko-kultúrák létrehozásában és használatában. Emellett nagy tapasztalattal rendelkezünk mikrofluidikai biochip modellek létrehozásában, fejlesztésében és fenntartásában (Walter és mtsai, 2016; Kincses és mtsai, 2020; Santa-Maria és mtsai, 2021). Ezt a két területet agyi organoid (Nickels és mtsai, 2020) tenyésztéssel tervezzük összevonni, amely eredményeinket a konferencián tervezzük bemutatni. Ez a modell a legmodernebb kutatási irányokhoz illeszkedik.

The 24th International Symposium on Signal Transduction at the Blood-Brain Barriers Conference is the most important event of our research field at the European level. This conference series is held annually in different European countries. Research groups from each country present their latest research findings in the blood-brain and other brain barrier study fields. The conference has been cancelled several times in recent years due to the pandemic. A similar meeting was held online this year, where I attended with a successful poster presentation. In 2022 we would like to participate at the in-person conference in Italy. This international event is extremely important to our field of research to share and discuss the latest scientific findings. I would have the opportunity to meet the most outstanding researchers in our field of research.

I plan to attend this international conference with a poster presentation.

The conference provides me a good opportunity to meet PhD students, postdocs, and many of the best researchers in the field. The event provides an opportunity to establish new collaborations with experts in the field of blood-brain barrier research. Oral and poster presentations at the conference will help me to learn about the latest scientific findings. My own poster presentation promotes my academic progress and the development of my presentation skills.

Summary and aims of the project for a general audience

The functional basis of the blood-brain barrier is formed by the brain endothelial cells, which are one of the most important defenders of the brain. Brain endothelial cells can prevent the entry of drugs into the brain with special (efflux) transporters and close intercellular connections. In the current modern drug development, testing the entry and toxicity of drugs to the brain using cell cultures is essential (Veszelka et al., 2011). The latest trend in drug discovery is the testing of drugs on brain spheroids and organoids (Choudhury et al., 2020). Currently, the most common investigation of cerebral drug delivery is performed with 2D co-cultures, which are grown on a permeable membrane in monoculture or co-culture with multiple cell types (Nakagawa et al., 2009; Veszelka et al., 2011; Helms et al., 2016). Human cell culture models are becoming increasingly important in drug discovery. Our group has extensive experience in establishing epithelial and endothelial-based barrier co-cultures on a cell culture insert. In addition, we have wide knowledge in creating, developing, and maintaining microfluidic biochip models (Walter et al., 2016; Kincses et al., 2020; Santa-Maria et al., 2021). We plan to combine these two models of study with brain organoid culture (Nickels et al., 2020). We would like to present our latest results using this cutting edge model at the proposed conference.

A rendezvény új együttműködések kialakítására adott lehetőséget a vér-agy gát témakörében jártas szakemberekkel. A mások által előadott előadások és poszter prezentációk segítették megismerni a legújabb tudományos eredményeket. A saját poszter prezentációm elősegítette a tudományos előmenetelemet és fejlesztette az előadó készségemet. Ez a konferencia a pandémiás időszak után jó lehetőséget biztosított, hogy személyesen is megismerjük a témán dolgozó kutatókat és a legfrissebb kutatási eredményeinket megoszthassuk egymás között.

I presented my results in poster format on the conference. The title was " Morphological and functional characterization of a human co-culture blood-brain barrier model and brain organoids in a microfluidic biochip ". This conference was a good opportunity to build relationships with other PhD students, postdoctoral fellows and the most outstanding researchers in our field. The event provided an opportunity to form new collaborations with the experts of the blood-brain barrier. The lectures and poster presentations by others helped to learn about the newest scientific results. My own poster presentation helped my academic carrier and developed my presentation skills. After the pandemic situation, this conference provided a good opportunity to get to know the researchers who working on the topic personally and to share our latest research results with each other.
