A projekt során megvizsgáltuk nyárfanövények felhasználhatóságát szennyezett talajok megtisztítására (fitoremediációjára). A vizsgálatokhoz egy szürkenyár hibridet, ennek két géntechnológiával módosított klónját, valamint a fekete nyár N-SL klónját alkalmaztuk. A génmódosított klónokat a baktériális eredetű gamma-glutamil-cisztein szintetáz génnel (gshI) transzformálták. Az enzim a növényi tripeptid glutation bioszintézisében játszik szerepet, így a transzformált növények magas glutation tartalommal rendelkeztek. A növények méregtelenítési reakciókban a glutation központi szerepet játszik. In vitro levélkorong kulturákban megvizsgáltuk a klón-stabilitást és a nyárfavonalak fitoremediációs kapacitását. A transzformált növényekbe beépített gshI gén jelenlétét PCR technikával ellenőriztük. Enzimológiai és klorofill fluoreszcencia mérésekkel összehasonlítottuk a transzgenikus és hagyományos nyárfa vonalak stressz-tűrését. A nyárfa növények jó stressz-ellenálló képességgel rendelkeztek acetoklór gyomírtószerrel és egyes nehézfémekkel szemben. A transzgenikus növények stressz-ellenállóképessége általában kis mértékben nagyobb volt, mint a nem-transzformált növényeké. Az erősen toxikus paraquat gyomírtószerrel szemben nem tért el jelentősen a transzgénikus és hagyományos nyárfa vonalak stressz-érzékenysége. A méregtelenítésben szerepet játszó egyes enzimek (aszkorbát-peroxidáz és glutation S-transzferáz) aktivitását jelentősen befolyásolta a nyárfa tápközegének szacharóz tartalma.

The suitability of different poplar varieties for the cleansing of polluted soils (phytoremediation) was studied. A grey poplar hybrid, its two transgenic clones, and the N-SL clone of Populus nigra were used during the investigations. The transgenic plants were transformed by a bacterial gene (gshI), which encodes the gamma-glutamyl-cysteine synthase enzyme. This enzyme participates in the biosynthesis of the endogenous plant tripeptide glutathione, and thus the transgenic plants contained elevated glutathione levels. Glutathione plays a principal role in plant detoxification reactions. The clone stability and the phytoremediation capacity of poplar lines was studied in vitro, in leaf disc cultures. The presence of the gshI transgene was verified by PCR technique. The stress tolerance of transgenic and normal poplar lines was compared by enzymatic analyses and chlorophyll fluorescence measurements. The investigations demonstrated the high stress tolerance of poplar plants against the herbicide acetochlor and heavy metals. Generally, the stress tolerance of transgenic poplars was slightly higher than that of normal poplar lines. The stongly phytotoxic herbicide paraquat caused similar toxic effects in both transgenic and normal poplar tissues. The activities of some enzymes participating in detoxification reactions (ascorbate peroxidase and glutathione S-transferase) were strongly influenced by the sucrose content of the nutrient medium of poplars.
