Hazánkban a daganatos betegségek jelentik az egyik fő meghatározó halálokot. Napjainkban egyre nagyobb teret nyer a kutatásokban a célzott daganat terápiát illetve tumor diagnosztikát kitűző irányvonal, amely valamilyen receptor-specifikusan kötődő molekulához, pl. peptidhez kapcsolná hozzá a hatóanyagot vagy a jelzett csoportot oly módon, hogy a konjugátum az eredeti receptor kötődési tulajdonságait ne veszítse el. A rendelkezésre álló legújabb kutatási eredmények azt mutatják, hogy a különböző humán tumorsejtek jelentős hányada expresszál luteinizáló hormon-releasing hormon, növekedési hormon-releasing hormon, szomatosztatin vagy bombesin/ gasztrin releasing peptid (GRP) receptorok valamelyikét. A jelfogó receptorok révén a radioaktív jelzőmolekulát peptid hormon analógokhoz kapcsolva nagy specificitású, immáron PET diagnosztikai alkalmazásra felhasználható vegyületekhez juthatunk. Ebben a pályázatban célul tűztük ki három olyan (LH-RH, szomatosztatin és bombezin) analóg jelölését, amely az eddig farmakológiai eredmények ismeretében felhasználható lehet a célzott diagnosztikai alkalmazásra, mivel vagy dúsulnak bizonyos daganatfajtákban, vagy pedig csak bizonyos szövetekben fordulnak elő a tumorokon kívül. A peptideket alkalmas módosítás után az egy lépésben szintetizálható 4-fluorobenzaldehiddel, illetve 3-fluoronikotinaldehiddel szándékozzuk jelölni, majd receptor kötési tesztekkel, illetve tumorsejtek internalizációs vizsgálatával ellenőrizni a dúsulásukat. Amennyiben kötődési tulajdonságaikat elvesztenék, alternatív funkciós csoportok segítségével más pozícióban vinnénk be a radioaktív jelet.

Cancer is one of the most decisive death-causes in Hungary. Recently the targeted chemotherapy and diagnostics have a growing role in the research areas, where the agent or the labelled moiety are attached to some kind of molecules (for examples peptides) specific for receptors in such a way that this conjugated molecule should retain their original receptor binding features. The newest available scientific results reveal that the majority of different human tumour cell lines express either of the receptors for luteinising hormone-releasing hormone, grow hormone releasing hormone, somatostatin or bombesin/gastrin releasing peptide. Due to the relaying receptor the radiolabelled peptide hormone analogues with high specificity become useful diagnostic tools for PET. Now we have aimed the radiolabelling of three analogues (LH-RH, somatostatin and bombesin respectively) which are overexpressed in certain tumours based on the recent pharmacological results or exist in certain organs except the tumour cells. After the appropriate modification of the peptides they will be conjugated with the 4-fluorobenzaldehyde or 3-fluoronicotinaldehyde, which are available in a one step synthesis. The results will be assessed with radioreceptor binding and tumour cell uptake studies. If the binding features get spoiled, we plan to build in the signal in a different position by means of an alternative prosthetic group.

A projekt célkitűzése, hogy Magyarországon először, 18F-ral kemoszelektív módon jelölt peptid hormon receptorokon ható ligandumokat állítsunk elő. Ezek a vegyületek az egészséges sejtekben csak kis mennyiségben található proteinek megemelkedett expressziójának kimutatásával alkalmasak lehetnek célzott daganatdiagnosztika megvalósítására. Munkánk során e célokat sikeresen megvalósítottuk. A jelölések alapja a 18F-ral jelölt 4-fluorobenzaldehid volt, amely az aminooxi ecetsavval módosított peptid ligandumokkal oxim kötést hozott létre. Ezzel a metódussal a jelölő izotóp kovalens kötéssel történő bevitele kemoszelektív módon, védőcsoportokat nem tartalmazó peptidek esetében megvalósítható. Három receptor-szelektív szekvenciát készítettünk el, nevezetesen az LH-RH-, szomatosztatin- és a bombezin receptorhoz kötődő vegyületet. A receptorkötési vizsgálatok kimutatták, hogy a hipotézis, mely szerint a megfelelő pozícióban végzett konjugáció nem rontja le jelentősen a receptorkötési tulajdonságokat, helytálló. Az LH-RH receptor szelektív szekvencia esetében a jelölést elvégeztük 2-, illetve 6-fluoronikotinaldehiddel is. Megállapítottuk, hogy a hidrofil tulajdonság növekedése mellett a receptoraffinitás változatlan maradt, vagyis ezzel az alternatív jelölő ágenssel a lipofil sejtalkotó részekhez kevésbé kötődő radioligandum előállítható. Elkezdődtek a tumort nem hordozó kísérleti állatokkal a biológiai eloszlás vizsgálatok.

The scope of this project was to acclimatize in Hungary a modern, chemoselective 18F labelling method to prepare radioligans, acting on peptide hormone receptors. These ligands can detect the elevated expression of proteins existing in a low level in the normal cells, and they can be useful tools for the targeted tumour diagnostic purposes. During this project we successfully fulfil this goal. The key compound was the 18F-labelled 4-Fluorobenzaldehyde. This molecule tends to react with aminooxy acetic acid modified peptides to form oxime bond. With this method we were able to introduce in a chemoselective way the radioisotope into the protecting group free peptides. We have synthesized three different receptor selective ligands, for the luteinizing hormone-releasing hormone-, for the somatostatin- and for the bombesin receptors, respectively. The receptor binding assays revealed that our hypothesis that the conjugation at the appropriate position would not impair significantly the receptor binding properties, is correct. In the case of LH-RH selective sequence we performed the labelling with 2- and 6-Fluoronicotinic aldehyde also. We have concluded that we could improve the hydrophilic character without destroying the receptor affinity, and we were able to produce radioligand with a reduced ability to bind to the lipophilic cell constituents. The biodistribution studies with a non-tumour bearing animals are in progress.
