A vanilloid receptor-1 (TRPV1) egy nem specifikus Ca2+ csatorna, amelyet magas hőmérséklet, alacsony pH, változatos endogén (pl. anandamid) és exogén (pl. kapszaicin) ligandok aktiválnak. Általános nézet szerint a TRPV1 a fájdalmas ingerek szenzora.
A szenzoros neuronokban expresszálódó TRPV1 aktivációja központi idegrendszeri (fájdalom) és helyi (neurotranszmitter felszabadulás az aktivált neuronális terminálisokból) hatásokban nyilvánul meg, Ezen lokális, úgynevezett „szenzoros-effektor” hatások közé tartozik pl. a substance-P vagy CGRP felszabadulása, amely fokozott endoteliális NO szintézis által vezet lokális vazodilatációhoz.
Bizonyos erekben (pl. arteria gracilis) azonban megfigyelések szerint a TRPV1 aktiváció vasokonstrikcióhoz vezet.

Hipotézis
A TRPV1 funkcionálisan expresszálódik az erek simaizomsejtjeiben. Ennek megfelelően a TRPV1 stimulációra bekövetkező érátmérő változásokat a szenzoros neuronokban (dilatatív) és a simaizomban (konstriktív) expresszálódó TRPV1 aktivitásának egyensúlya szabja meg.

Célok
1. A TRPV1 expresszió vizsgálata különböző szövetekben.
2. Az erek simaizmában expresszálódó TRPV1 élettani jellemzése.
3. A foszforiláció szerepének vizsgálata a TRPV1 válaszkészségének szabályozásában, szenzoros
neuronkban és simaizomsejtekben.
4. Agonisták és parciális agonisták struktúra-aktivitás összefüggéseinek meghatározása a
simaizomban expresszálódó receptorokon.
5. A TRPV1 agonisták esetleges terápiás hatásának ellenőrzése a hipotenzív sokk különböző
állatmodelljeiben.

The vanilloid receptor-1 (TRPV1) is a nonspecific Ca2+ channel which can be activated by heat, acids (low pH), various endogenous (e.g. anandamide) and exogenous (capsaicin) ligands and believed to be a polymodal integrator of various painful stimuli.
Activation of sensory neurons by stimulation of TRPV1 will result in central (pain) and local (release of neurotransmitters from the activated sensory neuronal terminals) effects. These local, so called ’sensory-effector’ effects includes vasodilation, mediated by the release of substance P or CGRP, which molecules activates the endothelial NO synthesis.
However, in some vascular beds (like in the arteria gracilis of the rat) TRPV1 activation leads to vasoconstriction.

Hypothesis
TRPV1 is expressed in the vascular smooth muscle cells and the vasoactive effect of TRPV1 stimulation is determined by the balance of the activity of the sensory neuronal (dilation) and the smooth muscle located (constriction) receptors.

Objectives
1. Immunohistochemical analysis of TRPV1 expression in various tissues.
2. Physiological characterization of TRPV1 located in the vascular smooth muscle cells.
3. Determination of the role of phosphorylation in the regulation of TRPV1 responsiveness in
smooth muscle and sensory neuronal cells.
4. Determination of the structure-activity of smooth muscle located TRPV1.
5. Investigation of the possible therapeutic effects of TRPV1 agonists in different models of
hypotensive shock.

Kutatásaink legfontosabb eredménye, hogy sikerült a transient receptor potential 1 (TRPV1) csatorna funkcionális arteriális expresszióját kimutatnunk. További kísérleteinkben felismertük, hogy a vaszkuláris TRPV1 farmakológiai tulajdonságai eltérnek a neuronális, fájdalomérzetért felelős receptorétól. Eredményeink arra utalnak, hogy a vaszkuláris TRPV1 egy független gyógyszergyári célpont lehet az erek összahúzására irányuló erőfeszítések esetében, továbbá, hogy a TRPV1 fájdalomcsillapítás érdekében történő gátlása nem kívánt vaszkuláris hatásokhoz vezethet.

The main finding of our research is that functional transient receptor potential 1 (TRPV1) channel expression is present in arterial smooth muscle cells. Further studies revealed that the pharmacological properties of the vascular TRPV1 are different from the TRPV1 expressed in sensory neuronal cells, responsible for sensory irritation. Our research pinpoints vascular TRPV1 as a potential pharmaceutical target to evoke arteriolar constriction in diseases where it is desired (e.g. shock) and suggests that inhibition of TRPV1 to antagonize pain may lead to unwanted vascular side effects.
