A tervezett kutatás kémiailag módosított szintetikus nanoporúsokban végbemenő biomolekuláris kölcsönhatások megértését és szelektív molekuláris felismerésre való alkalmazását tűzi ki célul. A nagy affinitású szintetikus bioreceptorokkal módosított nanopórusok alkalmazása analitikai célokra megoldást nyújthat a jelenlegi bioszenzorok stabilitási problémáira és mindemellett egy jelölés mentes és rendkívül széles körben alkalmazható bioérzékelési elvet biztosít. A megfelelően kis átmérőjű nanopórusok esetében a célvegyület kötödése a nanopórusokban immobilizált receptorokhoz megváltoztathatja különböző mechanizmusok alapján, mint például sztérikus gátlás vagy/és elektrosztatikus kölcsönhatás, a nanopórusok átjárhatóságát megfelelően megválasztott marker-ionok számára. Mivel a marker-ionok koncentrációja sok nagyságrenddel nagyobb a nanopórusok átjárhatóságát moduláló célvegyületek koncentrációjánál, ez a detektálási elv lehetővé teszi a biomolekuláris kölcsönhatások nagyérzékenységű nyomonkövetését. Tekintettel arra, hogy egy nanopórus esetében akár egy molekula bekötődése is jelentős változást okozhat a nanopóruson keresztüli ionáramban, a módszer elviekben akár egy molekula detektálására is alkalmas lehet. A pályázat alapkövei a kontrollált méretű és alakú nanopórusok előállítása (arany nanocsövek, nanopipetták, illetve fókuszált ionsugár maratással nanofabrikált nanopórusok) és ezek új szintetikus specifikus bioreceptorokkal való módosítása molekuláris önrendeződés alkalmazásával. Ennek megfelelően tervezzük nanopórusok peptidnukleinsavakkal (PNS), illetve DNS aptamerekkel történő kémiai módosítását amely a molekuláris felismerés széles tárházát nyitja meg. Míg a PNS szálak alkalmazása nukleinsavak specifikus felismerését szolgálják, addig DNS aptamerek in-vitro szelekcióval rendkívül nagyszámú komponens (toxinok, proteinek, gyógyszermolekulák) szelektív felismerésére állíthatók elő, amelyre a jelen pályázatban sor kerül. A tervezett kutatás a nanopórusok gyártásának, valamint a velük történő analitikai meghatározás módszertanának kidolgozását és optimalizálását is magában foglalja. A kifejlesztett módszerek alkalmazhatóságát orvosdiagnosztikai ill. élelmiszeranalitikai szempontból kiemelkedő fontossággal bíró nukleinsavak, toxin ill. protein meghatározásával kívánjuk igazolni. A pályázat a stratégiai fontosságú interdiszciplináris nano-biotechnológia területen egy nemzetközi pályázatokon versenyképes hazai kutatócsoport kialakulásához vezethet.

The project focuses on the fundamental understanding of the mechanisms of biomolecular interactions in chemically modified synthetic nanopores with the aim to design novel biosensing interfaces at molecular level. The essential requisites of the proposed study are the development and implementation of novel bioreceptors with tailored chemical and binding properties as well as technologies for the controlled fabrication of single and multi nanopore devices based on nuclear track-etch membranes, nanopipetes and focused ion beam milled microchips having various dimensions and shapes. Nanopores are going to be chemically modified with peptide nucleic acids (PNA) and DNA aptamers by using molecular self assembly. PNAs provide the opportunity for the specific recognition nucleic acids while DNA aptamers prepared in the frame of the present project by in-vitro selection provide high affinity recognition of a wide range of compounds ranging form small molecules to proteins. The biomolecular interactions (binding events) are going to be detected based on their modulating effect on the ion transport through the nanopores. In principle, the use of single nanopore membranes modified with proper bioreceptors provides the prospect of label free detection of single molecules. The analytical approach of the present study aims to design the receptor modified nanopores and assay methodology to achieve optimal performance of the biosensing process. Representative targets such as nucleic acids, toxins and proteins with practical relevance for medical diagnostics and food safety have been chosen to delineate the applicability and potential of the nanopore sensing method. The project has the potential to generate the core of a competitive research group with a valuable knowledge base in an interdisciplinary research area of internationally recognized strategic importance. This can lead to further attraction of international funds and collaborative international research.

A pályázat elsődleges célkitűzése a nanopórusok analitikai alkalmazásainak vizsgálata volt. Három platformot állítottunk elő és alkalmaztunk érzékelésre: arany nanopórusos membránokat, nanopipettákat, illetve 3D MEMS technológia és fókuszált ionnyaláb marással készült membránokat. Megoldottuk a nanopórusok kémiai módosítását szintetikus receptorokkal: peptid-nukleinsavakkal és originális aptamerekkel. Érzékelőket fejlesztettünk ki fehérjék, DNS szálak meghatározására, és nanorészecskék számlálására. Új aptamereket fejlesztettünk ki az alma törzsgöndörödés vírus meghatározására, amellyel sikerült a vírust közvetlenül növényi extraktumokban kimutatni. Elsőként sikerült szelektív szilárdtest ioncsatornákat előállítani, amelyek a biológiai ioncsatornákat meghaladó több mint hat nagyságrendnyi szelektivitással rendelkeztek. Szilárdtest ioncsatornákon alapuló nanomólos kimutatási határú potenciometriás ezüstion érzékelőket fejlesztettünk ki. Fehérje módosított pórusokat reaktorként alkalmazva polimer rudakat állítottunk elő, amelyek a felületükön szelektív kötőhelyekkel rendelkeznek a pórust módosító fehérjékre. Numerikus módszerekkel és durva szemcsézettségű molekuladinamikai számításokkal meghatároztuk a célkomponens átlagos pórushoz jutási idejének koncentráció függését, a nanopórusok felületi töltéssűrűségét, illetve a nanopórusosban fellépő nanofluidikai effektusokat. A pályázat támogatásával 1 könyvfejezet és 28 közlemény született, amelyek összesített impakt faktorja 134,3.

The primary objective of the project was to explore new aspects of nanopore sensing. Three different nanopore platforms were prepared applied for sensing: gold nanoporous membranes, nanopipepettes and nanoporous membranes made by combining 3D MEMS technology and focused ion beam milling. Methods for chemical modification of nanopores with synthetic receptors, i.e., peptide nucleic acids and original aptamers, were developed. Based on this we have introduced sensors for proteins, DNA strands, and for nanoparticle counting. New aptamers were selected for the determination of apple stem pitting virus, which could detect the virus directly in plant extracts. Selective solid-state ion channels with selectivities exceeding that of the biological ion channels, i.e., more than six orders of magnitude, were introduced for the first time. They formed the base for potentiometric silver sensors with nanomolar detection limit. Reactors using protein modified pores were implemented to generate surface imprinted polymer rods able to selectively recognize the template protein. Numerical methods and coarse grain molecular dynamics were implemented to determine the encounter time of the analyte and nanopore, the surface charge density of nanopores and to interprete nanofluidic effects in the nanopores. One book chapter and 28 scientific papers having a cumulated impact factor of 134.3 were published with the support of the project.
