Az ivarrendszer működését a hypothalamus luteinizáló hormon-releasing hormon (LHRH) neuronrendszere szabályozza. Az LHRH neuronhálózat különféle idegi neuromodulátor anyagok és hormonok befolyása alatt áll. Az idegszövetben a preszinaptikus bemenetek működését lipid hírvivők, endokannabinoidok módosítják. A pályázat célja az endokannabinoid szignalizáció szerepének megismerése az LHRH neuronok élettevékenységének szabályozásában. Korszerű molekuláris biológiai, elektrofiziológiai, elektronmikroszkopós módszerek és transzgenikus állatkísérleti modellek alkalmazásával tervezzük kimutatni 1. az LHRH neuronokban termelődő endokannabinoidok szintézisében és lebontásban résztvevő enzimeket és vizsgálni az endokannabinoid metabolizmus ösztrogén-függését; 2. jellemezni az LHRH neuronok gátló és serkentő afferens idegi bemeneteinek kannabinoid receptor 1 (CB1) tartalmát; 3. megismerni az LHRH idegsejtek CB1 tartalmú glutamáterg és gamma-aminovajsav (GABA)-erg idegi bemeneteinek eredő sejtjeit az agyban; továbbá, 4. whole cell patch-clamp módszer alkalmazásával jellemezni az LHRH neuronrendszerre és specifikus idegi afferenseire irányuló retrográd endokannabinoid jelátvitel főbb elektrofiziológiai jellemzőit és élettani szerepét. A vizsgálatok az LHRH sejtműködés új szabályozó mechanizmusait hivatottak feltárni. A várható eredmények hasznosíthatók lesznek gyógyszeripari fejlesztésekben és kábítószer függőségben szenvedők szaporodásbiológiai kórképeinek megértésében.

The luteinizing hormone-releasing hormone (LHRH) neuronal network of the hypothalamus centrally regulates the gonadal system. The LHRH system is under the regulatory control of different neuromodulators and hormones. In the central nervous system, the presynaptic neuronal inputs are also regulated by the lipid neuromessengers, endocannabinoids. The major goal of the project is to explore the role of endocannabinoid retrograde signaling in the regulation of the LHRH neuronal system. By means of molecular biological, electrophysiological and ultrastructural techniques and novel transgenic mice strains, we plan 1. to detect the enzymes that participate in the synthesis and degradation of endocannabinoids in LHRH neurons with a special emphasis on the estrogen dependency of the regulatory processes; 2. to characterize the cannabinoid receptor 1 (CB1) content of excitatory and inhibitory neuronal inputs of LHRH neurons; 3. to map and localize CB1 expressing neuron populations of the brain that give rise to glutamatergic and gamma-aminobutyric acid (GABA)-ergic innervation of LHRH cells; furthermore, 4. to elucidate - by the use of whole cell patch-clamp technique - the major characteristics and physiological significance of retrograde endocannabinoid signaling acting upon the LHRH system and its neuronal afferents. The project is aimed at the exploration of novel regulatory mechanisms of the LHRH neuronal network. The obtained results will be utilized in pharmaceutical drug development strategies and understanding the pathogenesis of gonadal malfunctions of drug-dependent patients.

A szaporodás idegrendszeri szabályozásában kulcs szerepet tölt be a gonadotropin-releasing hormone (GnRH) neuronrendszer. A rendszer működését perifériás hormonhatások és más agyi neuronhálózatok szabályozzák. Multidiszciplináris megközelítés alkalmazásával tanulmányoztuk a GnRH neuronrendszer kapcsolatait és szignál transzdukciós mechanizmusait, különös tekintettel a retrográd endokannabinoid szignalizáció szabályozó szerepére. Kísérleti eredményeinkről 24 tudományos közleményben adtunk számot, 96 összesített impakt értékkel. Feltártuk a hypothalamus kannabinoid receptor 1 (CB1) hírvivő RNS-t termelő idegsejtjeinek lokalizációját, valamint azok glutamáterg és GABA-erg fenotípusait. Igazoltuk, hogy a GnRH sejteken végződő GABA tartalmú idegvégződések CB1-t tartalmaznak, valamint bebizonyítottuk, hogy a GnRH idegsejtekből felszabaduló endokannabinoidok befolyásolják a GABA közvetítette információ átadást a GnRH neuronok felé. Feltártuk a ghrelin és endokannabinoid szignalizációs útvonalak kapcsolt jellegét a hypothalamusban. Igazoltuk a humán GnRH idegsejtek glutamát- és GABA-erg beidegzését. A GnRH neuronrendszer kisspeptinerg afferensei vonatkozásában új regulációs adatokat szolgáltattunk. Vizsgáltuk az ösztrogén szignalizáció szerepét a GnRH idegsejtek működésében, valamint az agykéregben. A GnRH idegsejtek működésének elmélyültebb tanulmányozására matematikai modellt alkottunk. Összegezve, eredményeink a reprodukció szabályozásának új mechanizmusait tárták fel.

Gonadotropin-releasing hormone (GnRH)-synthesizing neurons play a pivotal role in the central regulation of reproduction. Their operation depends on signaling by peripheral hormones and interactions with other neuronal circuits. By means of a multidisciplinary approach, the networking and signal transduction mechanisms of GnRH neurons were studied, with special reference to a putative retrograde endogenous cannabinoid signaling mechanism. The research results were published in 24 original papers representing a cumulative impact value of 96. Specifically, we mapped the hypothalamic distribution of cannabinoid receptor 1 (CB1) mRNA-expressing neurons and their GABA- and glutamatergic phenotypes, proved the presence of CB1 in neuronal afferents of GnRH neurons and characterized the impact of endocannabinoids liberated from GnRH neurons on the GABA-ergic signal transduction to GnRH cells. We provided evidence for the coupled nature of the ghrelin and the endocannabinoid signaling mechanisms. New GABA- and glutamatergic afferents of human GnRH neurons were also identified. In addition, novel regulatory mechanisms executed by kisspeptinergic circuits upon GnRH cells were revealed. We elucidated further characteristics of estradiol feedback effects to GnRH and cortical neurons. We established a mathematical model for the better understanding of GnRH cell performance. Collectively, our results shed light on novel mechanisms regulating reproduction at the hypothalamic level.
