A kromatográfiás és elektroforetikus elválasztási folyamatok részleteinek megértéséhez nagy fontosságú az oszlopban lejátszódó fizikai-kémiai folyamatok feltárása, megértése, hiszen azok alapvetően befolyásolják a retenciót, a szelektivitást, illetve a csúcsok aszimmetriáját és szélesedését, és így az elválasztás hatékonyságát.

A biológiailag aktív molekulák viselkedésének jellemzése az elválasztási folyamatban, a nemlineáris adszorpciós és megoszlási izotermák meghatározása alapján felderíthető az egyes fázisokat jellemző felismerési mechanizmus. Az izotermák ismerete ahhoz is szükséges, hogy a biológiailag aktív molekulák preparatív tisztítását optimalizáljuk. Ennek segítségével dolgozhatók ki gazdaságos biotechnológiai tisztítások.

A kromatográfiás állófázis és az oszlopra juttatott minta molekulái közötti kölcsönhatást a leggyakrabban az oszlop túlterhelésével, a nemlineáris izotermák meghatározásával jellemzik. A kromatográfiás állófázisok sok esetben nagyon kis mennyiségben tartalmaznak erős kötőhelyeket. Ezek jellemzése célszerűbb lineáris (analitikai) kromatográfiával, megfelelő retenciós modellt felhasználva.

Kutatásunk célja az, hogy a nemlineáris és a lineáris kromatográfia, illetve a kapilláris elektroforézis területén jellemezzük a molekuláris kölcsönhatásokat.

To understand the fundamentals of the chromatographic and electrophoretic processes, it is of utmost importance to study the physical-chemical phenomena taking place in a separation capillary or chromatographic column. They control the retention, the selectivity, as well as the asymmetry, the broadening of the peaks, hence the separation efficiency, too.

The characterization of the behavior of biomolecules during their migration, and the determination of their nonlinear adsorption or partition isotherm will provide fundamental information to understand the nature of the recognition mechanisms. The knowledge of the isotherms is necessary to scale-up and optimize the industrial separation and purification of biologically active molecules. This allows an economical increase of productivity for the related biotechnological purifications.

In chromatography, the overloading of the column is most often used to investigate the chromatographic process. However, when the stationary phase contains high energy adsorption sites in low abundance, linear chromatography can offer a better characterization of the underlying phenomena.

The objective of our study is to combine the tools of nonlinear and linear chromatography as well as capillary electrophoresis to characterize the molecular interactions in liquid phase separations.

Fordított fázisú töltetet tartalmazó HPLC oszlopokon szisztematikusan jellemeztük az oldószer és oldott anyag adszorpciós jellemzőit. A vizsgálatok lehetővé tették a szilanol aktivitás és a hidrofób kölcsönhatások nyomon követését. Többletizotermák meghatározásával megállapítottuk a fordított fázisú tölteteken az adszorbeált szerves módosító rétegvastagságát. A metanol adszorpciója során legfeljebb monomolekuláris borítottság alakul ki, míg acetonitril vagy tetrahidrofurán megkötődése során 3-4 adszorbeált molekuláris réteg is kialakulhat. A hidrofil kölcsönhatású kromatográfia adszorpciós tulajdonságait tanulmányozva megállapítottuk, hogy szoros a kapcsolat a szilikagél felületén adszorbeált víz mennyisége és a prolin adszorpciós kapacitása között. Ez az eredmény megerősíti azt a feltételezést, amely szerint HILIC elválasztásoknál a retenció mértékében fontos a felületen adszorbeált víz szerepe. Tanulmányoztuk a tömörmagvú töltetek anyagátadási tulajdonságait, és összehasonlítottuk azokat a teljesen porózus részecskék jellemzőivel. A számításaink megmutatták, hogyan határozható meg egy ekvivalens részecskeméret, amellyel a két állófázis diffúziós jellemzői közvetlenül összehasonlíthatók.

We have systematically characterized solvent and solute adsorption on a number of reversed phase columns. The study allowed the tracing of silanol activity and hydrophobic interactions. We have determined – via excess isotherm measurement – the thickness of the layers of adsorbed organic modifier on the surface of reversed phase stationary phases. The adsorption of methanol barely leads to a monolayer, while in the case of acetonitrile of tetrahydrofuran 3-4 molecular layers are adsorbed. When studying the adsorption properties in hydrophilic interaction chromatography, we established a strong correlation between the amount of water adsorbed on the silica surface and the adsorption capacity for proline. This confirms the hypothesis that retention in HILIC is due to the water adsorbed on the silica surface. We have investigated the mass transfer properties of core-shell particles and compared those to fully porous particles. Our calculations showed how an equivalent fully porous particle size can be determined when the two types of phase are compared.
