Az endoplamás retikulum (ER) a sejten belüli reaktív oxigénszármazékok (ROS) termelésének fő helyszíne. Az oxidatív fehérje folding során az oxigén hidrogén-peroxiddá (H2O2) redukálódik. A folyamat a ROS termelés negyedéért felelős egy szekréciós sejtben. A ROS képződése károsíthatja az ER lumen redox és antioxidáns homeosztázisát, valamint visszahathat a foldingra is. A hatások együttesen ER stresszhez vezethetnek, mely aktiválja az UPR (unfolded protein response) adaptációs mechanizmusát.
A nagyobb H2O2 termeléssel járó fokozott oxidatív folding modellezése azonban kísérleti nehézségekbe ütközik. Ennek megfelelően sem a luminális ROS felhalmozódást, sem a következményes redox változásokat a tiol/diszulfid és a piridin nukleotid rendszerekben nem lehetett bizonyítani in vivo vagy sejtes rendszerben. A kérdés tehát megoldatlan: hogyan tud egy szekréciós sejt alkalmazkodni a diszulfidhíd képződés melléktermékének felhalmozódásához.
A pályázat fő céljai a következők:
- modell rendszer létrehozása az excesszív protein folding imitálására, melyben a luminális H2O2/ROS felhalmozódás validálható;
- a tiol/diszulfid és a piridin nukleotid rendszerek redox státuszának vizsgálata fokozott H2O2 termelés esetén az ER lumenben;
- az ER csatornák/pórusok esetleges szerepének vizsgálata a H2O2 kiáramlásban;
- a tiol/diszulfid és a piridin nukleotid rendszerek redox státusza manipulálásának hatása a H2O2 eliminációjára, illetve az ER stressz kialakulására fokozott H2O2 termelés esetén.
Az eredmények hozzájárulhatnak a számos betegség patomechanizmusában kulcsszerepet játszó ER stressz jobb megértéséhez és terápiás befolyásolásához.

Endoplasmic reticulum (ER) seems to be a major place of reactive oxygen species (ROS) production. Oxidative protein folding results in the incomplete reduction of oxygen to H2O2 in the ER. The process can be responsible for one fourth of ROS produced in a professional secretory cell. ROS generated during oxidative folding can compromise the redox and antioxidant homeostasis of ER lumen, which in turn could affect the protein folding itself. These alterations in the ER homeostasis have been postulated as a causative factor of ER stress and the consequent activation of an intracellular signaling pathway (UPR, unfolded protein response), a surveillance mechanism to adapt ER capacity to the varied conditions.
However, the construction of a model system, in which the increased H2O2 generation is due to the stimulated oxidative folding, raises difficulties. Hence elevation of luminal ROS levels and the changes in the redox state of thiols and/or pyridine nucleotides have not been proved under these conditions. The question remains how secretory cells with heavy loads of protein thiols in the luminal compartment of the ER can accommodate to or eliminate the byproducts of disulfide bond formation.
The major aims of the present project:
- to design a model system, which can imitate the situation of excessive oxidative folding, and to confirm its validation by detecting luminal H2O2/ROS accumulation upon increased production in the ER;
- to detect the redox state of luminal thiols and pyridine nucleotides in the ER lumen upon increased H2O2 formation;
- to clarify the role of ER channels in the eventual H2O2 efflux;
- to investigate the effect of manipulations of the redox state of thiols and/or pyridine nucleotides and of ER channels on ER stress in conditions with high H2O2 generation.
The results can contribute to the better understanding of the participation of reactive oxygen species in the pathomechanism of ER stress during excessive oxidative protein folding.

Az endoplazmás retikulum (ER) oxidatív protein foldingjának luminális H2O2 termelése hozzájárul az organellum oxidatív környezetének kialakulásához. Kimutattuk, hogy az Ero1α, az oxidatív hajtogatás és ER redox homeosztázis egyik legfontosabb szabályozója, feldúsul a MAM frakcióban, és szabályozza a Ca2+ áramokat. Az Ero1α szintjének mind növelésével mind csökkentésével módosítani lehetett az ER Ca2+ fluxusokat, amely feltárja a fehérje kulcsfontosságú szerepét a korai szekréciós kompartimentumban. Azt is megfigyeltük, hogy a máj ER luminális H2O2 szint emelkedése in vivo egerekben a mikroszómális GSH és fehérje tiol tartalom csökkenését, valamint luminális oxidoreduktázok redox állapotának eltolódását eredményezte. Az oxidatív hatás kiváltotta ar ER tágulását, mely redukálószerekkel kivédhető volt. ER-be célzott katalázt overexpresszáló, antitest termelő sejtekben az érett antitest polimerek csökkent szekrécióját, míg az antitest prekurzor monomerek/dimerek intracelluláris felhalmozódását észleltük. Az eredmények szerint a helyi H2O2 termelés elősegíti, míg a H2O2 eltávolítása rontja a diszulfidok kialakulását. Három review-t közöltünk az ER redox viszonyairól. Egy tanulmányban az oxidatív protein foldingra új paradigmát javasoltunk: a több oxidáns hipotézist. Két átfogó review-ban a jelenlegi ismereteket foglaltuk össze az ER legfontosabb redox rendszereiről. Másik két cikkben pedig a kompartimentáció jelentőségét alátámasztó eredményeket tárgyaltuk.

Oxidative protein folding in the endoplasmic reticulum (ER) results in luminal H2O2 production, contributing to the formation of the oxidative environment of the organelle. We showed that Ero1α, a key controller of oxidative folding and ER redox homeostasis, is enriched in mitochondrial-associated ER membranes (MAM) and regulates Ca2+ fluxes. Either increasing or decreasing the levels of Ero1α affected Ca2+ fluxes, which reveals a pivotal role for this oxidase in the early secretory compartment. We also observed that the elevation of hepatic ER luminal H2O2 levels of mice in vivo resulted in a decrease in microsomal GSH and protein-thiol contents and in a redox shift of certain luminal oxidoreductases. The oxidative wave was accompanied by reversible dilation of ER, prevented by concomitant reducing treatment. ER targeted catalase overexpressing antibody producing cells showed diminished secretion of mature antibody polymers, while incomplete antibody monomers/dimers were accumulated and/or secreted. The results indicate that local H2O2 production promotes, while quenching of H2O2 impairs disulfide formation. We published three reviews on the redox conditions in the ER. In a Hypothesis paper we proposed a new paradigm for the oxidative folding: the multiple oxidant hypothesis. In two comprehensive reviews we summarized the present knowledge on the major redox systems in the ER. We summarized the facts showing the importance of compartmentation in two other reviews.
