A kórokózó és a gyökérközegben élő, nem patogén mikroorganizmusok, a növényevő állatok valamint az abiotikus stresszorok jelátviteli utak hálózatát aktiválják, amelyek átfedhetnek, finoman szabályozottak, és amelynek következtében a növényi válaszreakciók aktiválódása egyaránt vezethet keresztrezisztenciához vagy indukálhatja a programozott sejthalált (PCD). A szalicilsav (SA) és a NaCl magas koncentrációi módosítják a reaktív oxigénformák (ROS) és NO homeosztázisát, a SA és NaCl együttes hatására koncentrációjuk csökkenhet is, ami hatással van a PCD kialakulására, az antiapoptotikus Bax inhibitor-1 protein és a PCD-vel kapcsolódó iniciátor vagy a fehérjebontást teljessé tevő proteázok (vakuoláris processzáló enzim (VPE), metakaszpázok, szerin proteázok) expressziójára és aktivitására. Az SA és a NaCl által indukált PCD morfológiai és biokémiai jellemzőit vizsgáljuk külön-külön és a két kezelést együttesen alkalmazva intakt paradicsom növényben és sejtszuszpenzióban abból a célból, hogy megvizsgáljuk az önálló és kombinációban alkalmazott kezelések hatását a ROS és az NO akkumulációjára, valamint a BI-1 és a PCD-vel kapcsolt proteázok expressziójára keresve a hatások specifikumát. Az SA és a NaCl hatását a ROS és az NO akkumulációjára, valamint a BI-1 és a PCD-vel kapcsolt proteázok expressziójára vizsgáljuk etilén receptor mutáns (Never ripe), ABS bioszintézis mutáns (flacca and sitiens) és jázmonsav bioszintézis mutáns (def1) paradicsomban a hormonális jelátviteli utak lehetséges kapcsolatainak felderítése céljából. Az SA- és a sóstressz-indukált PCD iniciációs eseményeinek, így a plazmamembrán depolarizációnak, az anion kieresztésnek, az NO és ROS produkciónak, a citokróm c mitokondriumból történő kieresztésének valamint a DNS fragmentációnak a vizsgálata a sejtszuszpenziós kultúrában történik.
Summary
The signalling networks that are activated in response to pathogenic, herbivorous and beneficial organisms or abiotic stressors may overlap and are finely balanced and the activation of plant responses may lead to cross-tolerance or cell death. ROS and NO homeostasis modified by high concentrations of salicylic acid (SA) or NaCl affects the initiation of the programmed cell death (PCD) and the gene expression of anti-apoptotic Bax inhibitor-1 (BI-1) protein and different types of plant cell death-associated proteases (VPE, metacaspases and serine proteases). ROS and NO production may significantly decrease when SA and NaCl are applied in combination. The morphological and biochemical features of PCD induced by SA or NaCl and their combined effect will be compared in tomato plants as well as in suspension cultures in order to reveal the role of ROS and NO fine-tuning in the induction of the expression of PCD associated proteins. The SA and NaCl-induced PCD and the expression of anti-apoptotic BI-1 protein and PCD-associated proteases will be monitored in parallel with ROS and NO accumulation in ethylene receptor mutant (Never ripe), ABA biosynthesis mutants (flacca and sitiens) and in jasmonic acid biosynthesis mutant (def1) tomato to find possible cross-talk between hormonal signalling pathways. The initiating events leading to SA- and salt stress-induced PCD, the plasma membrane depolarization, the efflux of anions from the cells, the NO and ROS production, the release of cytochrome c from mitochondria and the morphological features such as DNA fragmentation will be monitored in tomato cell suspension cultures.

Munkánk célja annak feltárása volt, hogyan szabályozzák a reaktív oxigén- (ROS) és nitrogénformák (RNF) homeosztázisában bekövetkező változások a NaCl és a szalicilsav szubletális és letális dózisaival kezelt paradicsom növényekben az akklimatizációt és a stressz-indukálta sejtelhalást (PCD). Megállapítottuk, hogy a poliaminok végoxidációja által indukált ROS és RNF a sótressz által indukált PCD iniciációban játszik specifikus szerepet. A hormon jelátviteli utak akklimatizációban és a PCD indukcióban betöltött szerepét az abszcizinsav bioszintézis mutáns sitiens, etilén receptor mutáns Never ripe és jázmonsav jelátviteli mutáns jai1-1 paradicsom növényekben vizsgáltuk. A vad típus számára szubletáis sóstressz minden mutánsban PCD-t indukált a gyökércsúcsokban akkor, ha a szuperoxid és peroxinitrit koncentráció szignifikánsan megemelkedett, míg a H2O2 és NO esetén ez elmaradt. Bár a levélben mindkét stresszor hatására gyorsan nőtt egyes cisztein proteázok expressziója, a gyökérben ezek elsősorban az SA hatására expresszálódtak. A cisztein proteáz inhibitorok expressziója a szubletális stressznél mindkét stresszornál magasabb volt, így a proteáz inhibitorok hiánya vezethet a proteázok erőteljes aktiválódásához letális stressz során. A gyors proteáz aktiválódás miatt a sóstressz nevezhető autolitikus PCD formának, míg a proteázok aktiválódását az SA letális koncentrációja mellett is megfigyelhető proteáz inhibitor és Bax Inhibitor-1 expresszió késleltetheti.
Results in English
Our aim was to reveal how the changes in the homeostasis of reactive oxigen (ROS) and nitrogen forms (RNF) regulate the acclimation and the cell death of tomato plants exposed to sublethal and lethal concentrations of NaCl or salicylic acid (SA). It was found that ROS and RNF generated by the oxidation of polyamines play a specific role in the initiation of PCD during salt stress. The role of various hormonal signaling pathways in the acclimation and in PCD induction were investigated in abscisic acid biosynthesis mutant sitiens, ethylene receptor mutant Never ripe and jasmonic acid signaling mutant jai1-1 tomato plants. The salt stress which was tolerated by the wild types, induced PCD in the root apex of mutants if the concentrations of superoxide anion and peroxinitrite significantly increased, while the level of H2O2 and NO declined. The expression of various genes encoding cysteine proteases increased rapidly by the two stressors, in the root they were expressed primarily under the effect of SA. The expression of cysteine protease inhibitors in the case of sublethal stress was higher under both stressors, so the absence of protease inbibitors may lead to significant activation of proteases during lethal stress. Due to the rapid increase in protease activity under salt stress, high salinity can be considered as autolytic PCD, while the expression of protease inhibitors and Bax Inhibitor-1 may delay the proteolysis the in tissues exposed to lethal SA concentration.
