Itt írja le a kutatás fő célkitűzéseit a témában jártas szakember számára.

A bonyolult összetételű biológiai és környezeti minták teljeskörű folyadékkromatográfiás analízise állandóan napirenden lévő probléma. A mintákban jelenlevő nagyszámú komponens miatt ezen analitikai feladatok megoldása új folyadékkromatográfiás módszerek kifejlesztését igényli. Az elmúlt évek során előtérbe kerültek a kétdimenziós folyadékkromatográfiás (2DLC) elválasztások köszönhetően ezen módszerek előnyös szelektivitásának valamint nagy hatékonyságának. A 2DLC elválasztásokkal elérhető hatékonyság több nagyságrenddel nagyobb, mint egydimenziós kromatográfia esetén.

Egy kétdimenziós elválasztás alkalmazhatósága függ az egyes dimenziókban alkalmazott elválasztások mechanizmusától és hatékonyságától, az egyes dimenziók közötti kapcsolat kialakításának módozatától, a kétdimenziós elválasztás robosztusságától, valamint a vizsgálatok során keletkező nagymennyiségű adat hatékony feldolgozásától. Ennek megfelelően a tervezett kutatás célja az alábbi három, logikusan összefüggő, egymásra épülő részre bontható:

(1) bioaktív multiprotikus ionok (karbonsavak, aminosavak, peptidek, proteinek) folyadékkromatográfiás elválasztásának fizikai-kémiai alapjainak vizsgálata és leírása különböző típusú állófázison (ioncserélő, hejszerkezetű fordított fázis, héjszerkezetű HILIC),
(2) kétdimenziós folyadékkromatográfiás elválasztások robosztusságának meghatározása,
(3) adatfeldolgozó és adatmegjelenítő algoritmusok kidolgozása 2DLC elválasztások kiértékeléséhez.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

A kutatás célja kétdimenziós folyadékkromatográfiás módszerfejlesztések és rutin analitakai felhasználásuk támogatása, elősegítése alapvető elméleti problémák vizsgálata és adatfeldolgozó algoritmusok fejlesztése révén.
A tervezett kutatás segítségével megválaszolandó kérdések, megoldandó problémák:
(1) kétdimenziós elválasztások robosztusságának meghatározása,
(2) multiprotikus ionok retenciós mechamizmusának, csökkent elválasztási hatékonyságának tisztázása,
(3) proteinek adszorpciós izotermájának meghatározása különböző típusú állófázisokon,
(4) kétdimenziós elválasztások gyors és hatékony adatfeldolgozása.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

A modern környezeti és biokémiai kutatások (pl. proteomika, metabolomika, lipodomika) során egyre bonyolultabb összetételű minták analízisére van szükség. A meghatározandó komponensek száma gyakran jelentősen meghaladja a jelenlegi kromatográfiás rendszerek csúcskapacitását. Ilyen esetekben egydimenziós módszerekkel nincs mód ezen mintaelegyek teljeskörű analízisére. Ezen analitikai feladatok megoldása új kétdimenziós folyadékkromatográfiás módszerek kifejlesztését igényli. A tervezett kutatás eredményei hasznos információval szolgálnak 2DLC elválasztások tervezéséhez, opimálásához. Az elvégzendő robosztusságvizsgálatok gyakorlati információt arról, hogy a kétdimenziós elválasztás eredményei mennyire érzékenyek a módszer paramétereinek megváltozására. A fejlesztett retenciós modellek további 2DLC módszerfejlesztések alapját képezhetik biológiai és környezeti minták analízisére. A projekt során fejlesztendő adatfeldolgozó algoritmus hozzájárul a 2DLC-s elválasztások széleskörű elterjedéséhez, rutinszerű felhasználásához biokémiai és környezeti kutatásokban.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

A bonyolult összetételű biológiai és környezeti minták kémiai összetételének meghatározása állandóan napirenden lévő probléma a modern kutatásokban. A mintákban jelenlevő nagyszámú komponens miatt ezen analitikai feladatok megoldása új kémiai vizsgálati módszerek kifejlesztését igényli. Az elmúlt évek során előtérbe kerültek az ún. kétdimenziós folyadékkromatográfiás módszerek, ugyanis ezen módszerek hatékonysága több nagyságrenddel nagyobb, mint a hagyományos, egydimenziós módszereké.

A kutatás célja három fő pontban foglalható össze:
(1) biológiailag aktív ionok (karbonsavak, aminosavak, peptidek, proteinek) folyadékkromatográfiás analitikai meghatározásának fizikai-kémiai alapjainak vizsgálata,
(2) kétdimenziós folyadékkromatográfiás analitikai módszerek megbízhatóságának meghatározása,
(3) adatfeldolgozó és adatmegjelenítő algoritmusok kidolgozása kétdimenziós folyadékkromatográfiás analitikai vizsgálatok eredményeinek kiértékeléséhez.

A három, logikusan összefüggő, egymásra épülő rész hasznos információkkal szolgál ezen új analitikai technika módszereinek fejlesztéséhez, hozzájárul széleskörű elterjedéséhez, rutinszerű felhasználásához a biológiai és környezeti kutatásokban.

Most of the important problems that analysts must face nowadays involve the analysis of samples consisting of hundreds of components present in a very wide range of concentrations. Such problems frequently arise in proteomic, lipodomic and metabolomic research. Separations by unidimensional chromatography are unable to provide acceptable analyses of these samples. For this reason, two-dimensional liquid chromatographic (2DLC) separations have attracted intense interest over the past years, due to the tremendous potential improvements in separation power that this method offers.

The efficient applicability of a two-dimensional separation depend onthe retention mechanism and efficiency of the separations used at the two dimensions, the scheme used for implementing the coupling between dimensions, the robustness of the 2D method used, and the method for managing the large amount of data produced during 2DLC separations. Accordingly, the goals of the proposed research project consists of three logically interconnected parts:

(1) investigation of retention mechanism of multiprotic bioactive ions (carboxylic acids, amino acids, peptides, proteins) on different kind of stationary phases (ion exchanger, reversed phase core-shell, silica core-shell) under isocratic and gradient conditions,
(2) determination of robustness of two-dimensional liquid chromatographic separations,
(3) development of data management algorithms for the evaluation of chromatographic data.

What is the major research question?

The aim of the work is the support of method development and routine analysis in two-dimensional liquid chromatography by investigating fundamental aspects and developing data management algorithms. The work of the proposed research project will:
- quantify the robustness of 2DLC separations,
- clarifiy the reason of smaller separation efficiency of multiprotic compounds,
- desribe the isotherm of peptides on different kind of stationary phases,
- produce an efficient data management algorithm for peak recognition and integration in 2DLC-DAD (DAD: diode array detection).

What is the significance of the research?

Modern environmental and biochemical research (e.g. proteomics, metabolomics, lipodomics) is inherently complicated by the tremendous complexity of the samples involved in these studies. Current HPLC technologies do not afford the opportunity to observe all the compounds simultaneously in these samples, since the number of components exceed the actual peak capacity of these separation systems. Development of two-dimensional liquid chromatographic methods that allows the complete separation of complex biological and environmental samples are indispensable for future biochemical, pharmaceutical and environmental research. The results of the proposed research project will support analysts in 2DLC method development and optimization. The investigation of robustness of these techniques can serve practical information about how the analytical results are affected by the variation of method parameters. The developed retention models can be used as a basis for 2DLC method development for analysis of complex biological and environmental sample. The data management algorithms to be developed in the work will help 2DLC become a widespread used routine analytical technique in biochemical and environmental research.

A projekt során nagyhatékonyságú kétdimenziós folyadékkromatográfiás (2DLC) elválasztások alapvető aspektusainak vizsgáltuk. Tanulmányoztuk 2DLC elválasztások zavartűrését. Megállapítottuk, hogy az elválasztások bizonytalansága nő a második dimenziónak köszönhetően. A nagy térfogatú frakció gyűjtés ezt a hatást ellensúlyozza. Off-line 2DLC módszert fejlesztettünk fehérje emésztvény analízisére. Általános algoritmust vezettünk le 2DLC elválasztások oldószer felhasználásának minimalizálására. Peptidek és proteinek retenciós viselkedését tanulmányoztuk. Meghatároztuk ezen komponensek adszorpciója során bekövetkező moláris térfogatváltozást és az adszorpciós izotermákat. Megmutattuk, hogy a mozgófázis hosszirányú hőmérséklet növekedése a kromatográfiás oszlopban a vizsgált komponensek retencióját igen, de elválasztásuk hatékonyságát nem befolyásolja. Folyadékkromatográfiás állófázisok szemcseméret eloszlásának (PSD) hatását vizsgáltuk a kromatográfiás elválasztás hatékonyságára. Bizonyítottuk, hogy a széles PSD jelentősen csökkentheti az analízis hatékonyságát. Megmutattuk, hogy a tömörmagvú állófázisok alkalmazása előnyös lehet preparatív folyadékkromatográfiában is. Numerikus módszert dolgoztunk ki folyadékkromatográfiás állófázisok felületi heterogenitásának meghatározására többrétegű adszorpciós izotermák esetén. A módszert sikeresen alkalmaztuk állófázisok jellemzésére.

In this project, fundamental aspects of high performance two-dimensional chromatographic (2DLC) separations were investigated. Robustness of 2DLC methods were studied. It was shown that the uncertainty of separations increased by adding one more dimension. The large volume of fraction collection, however, decreased this effect. Off-line 2DLC method was developed for the separation of protein digests. A general algorithm was developed for the minimization of consumption of organic solvent in 2DLC. Retention behavior of peptides and proteins were studied. The change of molar volumes and adsorption isotherms of these compounds were determined. It was shown that the axial temperature increase of mobile phase affected the retention but did not affect separation efficiency. The influence of width of particle size distribution (PSD) of HPLC stationary phases on chromatographic efficiency was studied. It was proven that wide PSDs have significant effect on the separation efficiency of molecules. It was shown that the use of core-shell phases is advantageous also in preparative liquid chromatography. A numerical method was developed for the estimation of the surface heterogeneity below an adsorbed multilayer of the analyte in liquid chromatography. The method was used succesfully for the caracterization of different stationary phases.
