A C3 komplementfehérje eredetű fragmentumok kapcsolódása az antigénhez lehetővé teszi annak kötődését a komplementreceptorokat (CR) hordozó sejtekhez. A β2-integrinek családjába tartozó CR3 és CR4 komplementreceptorok kifejeződnek dendritikus sejteken (DC), makrofágokon és neutrofil granulocitákon. Ezen receptorok extracelluláris domén hasonlósága igen nagy; 87 %. A CR3 (CD11b/CD18) és CR4 (CD11c/CD18) receptorok ligandumai hasonlóak, fő ligandumuk az iC3b fragmentum. A receptorok eltérő szerkezetű intracelluláris doménjei lényeges funkcionális különbözőségre utalnak. A CR3 legfontosabb szerepe az iC3b által opszonizált patogének megkötése és fagocitózisa. A CR3 deficienciája a veleszületett immunválasz komoly zavarát okozza.
A vizsgálatok túlnyomórészt a CD11b-vel foglakoznak, ezekben a természetes ligandumot, az iC3b-t használják, ami a CR3-hoz és a CR4-hez egyaránt kötődik, ennélfogva nem különíthető el a kétféle receptor hatása. Előkísérleteink eredményei szerint a monocita eredetű emberi DC-k C3 komplementfehérje-függő fagocitózisa elsősorban a CR3 közvetítésével valósul meg.
A CD11c molekula a mieloid sejtek markereként ismert. A CR4 szerepe kevéssé jellemzett, DC-k funkciójára gyakorolt hatásáról keveset tudunk. Munkánk célja az, hogy meghatározzuk a CD11b és CD11c expresszióját és specifikus szerepét egér és emberi DC-k esetében.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

A legtöbb CD11b/CD18 (CR3) és CD11c/CD18 (CR4) funkcióra vonatkozó ismeretünk egér eredetű sejtek vizsgálatából százmazik. Ezeknek a molekuláknak a pontos működése dendritikus sejtek (DC) esetében nem ismert.
A CR3 és CR4 kifejeződésének és funkciójának különbségeit emberi eredetű sejteken elsőként szeretnénk feltárni.
Szeretnénk megválaszolni a kérdést: mi a specifikus szerepe a CD11b és a CD11c molekulának dendritikus sejtek esetében? Vizsgálni fogjuk ezeknek a molekuláknak a jelentőségét CD11b illetve CD11c géncsendesített monocita eredetű emberi DC-kben. Tervezzük meghatározni a CD11c specifikus szerepét egér rendszerben is, CD11b KO egerek csontvelői eredetű DC-iben.
Tisztázni szeretnénk, hogy melyek azok a sejtfunkciók amelyeket a CR3 vagy CR4 receptorok befolyásolnak. Ennek érdekében vizsgáljuk az aktivált sejtek adherencia képességét, endicitótikus aktivitását és citokin szekrécióját. A CD11b illetve CD11c közvetített módon aktivált sejtek citokin termelésének típusát – Th1, Th2 - szintén vizsgáljuk.
Meghatározzuk a CR3 és CR4 szerepét a CD11b vagy CD11c közvetítésével aktivált humán és egér DC-k T-sejt aktiváló képességének mérésével. Ezen vizsgálatok elvégzése után képesek leszünk arra, hogy megválaszoljuk a kérdést; mi a pontos, egymástól eltérő funkciója a CD11b és CD11c molekuláknak DC-ken.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

A visszatérő fertőzésekkel járó leukocyte adhesion deficiency (LAD) betegség fefedezésével a β2-integrinek az érdeklődés középponjába kerültek. A leukocyte adhesion deficiency type I (LADI) betegek esetében a CD18 génjében mutatciót azonosítottak, ami a sejtfelszíni CD11/CD18 integrinek súlyos vagy teljes funkció károsodásához vezet. Az álláb kialakulása során szintén fontos szerepet kap a β2-integrinek átrendeződése, ami a Wiskott Aldrich Syndrome protein (WASp) hiányában a WAS betegek esetében nem megy végbe.
Számos tanulmány igazolja, hogy a (CD11b/CD18) és a CR4 (CD11c/CD18) receptorok nem csak az effektor funkciókban, hanem az adaptív immunválasz stimulálásában is fontos szerepet. Ebben a folyamatban a DC-n kifejeződő receptorok pontos szerepét nem jellemezték.
A CD11b funkciót vizsgáló tanulmányokban általában az iC3b természetes ligandummal aktiválják a sejteket, ami CR3-hoz és CR4-hez egyaránt kötődik így a receptor specifikus hatások átfednek. Ezidáig a CD11c humán mieloid marker molekula specifikus műkődését nem sikerült meghatározni. Tecnikai nehézséget jelent az is, hogy a CD11b és CD11c molekula egyidőben fejeződik ki, ugyanazon a sejten. Ígéretesnek tünne specifikus ellenanyaggal keresztkötve vizsgálni a CR4 közvetített funkciókat. Ennek az az akadálya, hogy nincs megfelelő, a ligandum-kötőhellyel reagáló ellenanyag. Mindezen problémák ellenére egyre nagyobb az érdeklődés a CD11c funkciójának feltárására iránt. Célunk ez említett folyamatok minél részletesebb megértése, hogy eredményeinkkel megvilágítsuk a CR4 és CR3 egyedi funkcióit.
A CD11b KO egerek tanulmányozása lehetőséget kínál a CD11c pontos szerepének meghatározásához. Továbbá CD11b illetve CD11c géncsendesített humán DC-k használatával tisztázhatjuk a másik receptor pontos funkcióját. Az egér és humán rendszerben kapott eredmények összehasonlításával lehetőségünk lesz tisztáni az esetleges különbségeket a két faj között. A CR3 és a CR4 funkcióinak minél jobb megértése segíthet a CD11b és CD11c receptorokat célzó új DC-alapú terápia kifejlesztésében, amely autoimmun betegségek kezelésében és vaccina tervezésében nyújt lehetőséget.

A dendritikus sejtek az immunválasz fontos sejtes résztvevőit képezik. Ezek a sejtek karmesterei számos kulcsfontosságú veleszületett immunfolyamatnak. A dendritikus sejteket Ralph Steinman fedezte fel, akit ezért élettudományi Nobel díjjal jutalmaztak 2011-ben.
A komplementrendszer szintén fontos eleme az immunrendszernek. A komplementrendszer központi eleme a C3 fehérje, ami a különböző immunsejtek – pl. dendritikus sejtek - felszínén jelenlévő komplementreceptorokhoz tud kötődni. Ezeknek a receptoroknak a deficienciája súlyos zavart okoz az immunológia védekezésben. Ezen receptorok bizonyos funkciói dendritikus sejteken nem ismertek.
Célunk, ezeknek a receptoroknak a szerepét minél részletesebben feltárni. Előzetes kísérleti eredményeink szerint, ezek a receptorok a C3 fragmentumokat megkötve aktiválódnak, ami fokozza a dendritikus sejtek védekező működését. A CR3 és a CR4 funkcióinak minél jobb megértése segíthet a CD11b és CD11c receptorokat célzó új DC-alapú terápia kifejlesztésében, amely autoimmun betegségek kezelésében és vaccina tervezésében nyújt lehetőséget.

The deposition of activated fragments of complement component C3 allows the binding of antigens to cells expressing different complement receptors (CR). Dendritic cells, macrophages and neutrophil granulocytes express CR3 and CR4 complement receptors, that are members of the family of β2-integrins. Their similarity is very high regarding their extracellular domain; 87% respectively. The ligand specificity of CR3(CD11b/CD18) and CR4(CD11c/CD18) is highly overlapping, and their main ligand is identical: the iC3b fragment. Moreover the CD11c molecule is accepted as a marker of myeloid cells. However they differ in their intracellular domain and this suggests fundamental differences between the function of these receptors. The most important role of CR3 is the binding and phagocytosis of pathogens opsonized by iC3, and CR3 deficiency seriously compromises the innate immune defence of the host. Most of the studies focus on the role of CD11b. However, for the experiments the natural ligand, iC3b is used that stimulate both CR3 and CR4 in the same time, hence the effects mediated by the two receptors can not be separated. Our preliminary results show that complement C3 dependent phagocytosis of MDCs is mediated mainly by CR3. The role of CR4 is not well characterized and little is known about its function in DCs. Our aim is to determine the expression pattern and the exact role and function of CD11b and CD11c of mouse and human dendritic cells.

What is the major research question?

Most of the data available for CD11b/CD18 (CR3) and CD11c/CD18 (CR4) functions come from studies carried out using cells of mouse origin. The exact function of this molecules on dendritic cells is not determined. We want to reveal the differencies in the expression and function of CR3 and CR4 for the first time in human system.
We would like to answer the question what is the specific function of CD11b and CD11c molecule on dendritic cells? We will assess the role of these structures in CD11b or CD11c gene silenced monocyte derived human dendritic cells. Our goal is to determine the specific role of CD11c also in mouse models, using CD11b KO bone marrow derived dendritic cells.
We want to clarify what kind of functions are medieted by CR3 and CR4? This question will be answered by determining the endocytotic activity of the cells specifically activated via these complement receptors. Furthermore the array of cytokines secreted by CD11b and CD11c activated mouse and human dendritic cells will also be investigated. The role of CR3 and CR4 will be analyzed by assessing T cell activating capacity of human and mouse DCs after activation via CD11b and CD11c. Hence we will able to answer the question what are the distinct functions of CD11b and CD11c on dendritic cells.

What is the significance of the research?

The β2 integrins came into prominence when the clinical syndrome leukocyte adhesion deficiency (LAD), characterized by recurrent infection was discovered. Patients with type I deficiency (LADI) have mutations in CD18 that lead to severe or total lack of the CD11/CD18 integrins on the cell surface. Podosome assembly is also known to be associated with the specific recruitment of β2 integrins, which in the absence of the Wiskott-Aldrich Syndrome protein (WASp) did not occur.
Several studies demonstrate that that CR3 (CD11b/CD18) and CR4 (CD11c/CD18) play important role not only in the effector mechanisms but also in the initiation of adaptive immunity. The distinct function of these receptors on dendritic cells are not well characterised.
Most of the publications focus on CD11b and use natural ligands that stimulate both CR3 and CR4 in the same time, hence the effects mediated by the two receptors can not be separated. Up to now very little is known about the specific function of CD11c, the human myeloid marker. Another technical difficulty is that CD11b and CD11c molecules are expressed simultanously by the same cells. Using specific antibodies for receptor crosslinking would be a promising tool for analyzing CR4-independent functions, but there is no ligand binding site specific antibody available for this receptor. Despite these problems, there is increasing interest in the function of CD11c expressed by cells of the immune system.
Our goal is to understand the underlying mechanisms of CR4 functions in details, and our results will shed light on the unique and distinct funcions of CR4 and CR3. Investigation of mouse CD11b KO dendritic cells offers an opportunity to determine the exact role of CD11c. Using CD11b or CD11c silenced human dendritic cells we will clarify the proper function of these receptors separately. Comparison of mouse and human data will clarify the potential differences between the appearance and function of these receptors in these two species. Better understanding of the function of CR3 and CR4 will help to clarify how targeting of CD11b or CD11c can lead to the identification of new targets in DC-based therapies in autoimmune disorders as well as in immunization.

Dendritic cells are known to be key cellular participant of the immune response. These cells were discovered by Ralph Steinman, who has been awarded the Nobel Prize in Life Sciences in 2011. The complement system is also an important element of the immune system. The central component, protein C3 can bind to different complement receptors present on the surface of several immune cells, including dendritic cells. Deficiency of these receptors seriously compromises our immune defence. Certain functions of these complement receptors on dendritic cells are not well characterized yet, thus our aim is to reveal the role of these receptors in more detail. Our preliminary data show, that dendritic cells can be activated via complement receptors and this enhances the protective effect of these cells. Better understanding of the function of complement receptors on dendritic cells will help to find new possibilities in DC-based therapies in autoimmune disorders as well as in vaccination.

Munkánk célja a CR3 (CD11b/CD18)és CR4 (CD11c/CD18) komplementreceptorok specifikus funkcióinak felderítése. • Meghatároztuk a CR3 és CR4 mennyiségét és konformációját emberi fagociták esetében. • Megállapítottuk, hogy az emberi fagociták C3-függő fagocitózisában a CR3 meghatározó szerepet játszik. • Az emberi fagociták fibrinogénhez történő adherenciájában a CD11c szerepe döntő. • Az sejtek adherenciájának jellemzésére a klasszikus módszer mellett state-of-the-art biofizikai módszereket is bevezettünk.

Our goal was to dissect and determine the individual functional properties of human CR3 (CD11b/CD18) and CR4 (CD11c/CD18). • The absolute numbers and the conformational state of CR3 and CR4 expressed by the human phagocytes were assessed. • The complement C3-dependent phagocytosis of human phagocytes is mediated mainly by CR3. • The cellular adherence to fibrinogen by human phagocytes is dominated by CD11c. • The strength and the kinetics of adherence were estimated using classical and state-of-the-art biophysical methods.
