A szintetikus cathinonok csoportjába tartozó szerek 'party drogként' váltak ismertté. Elterjedtségük folytán egyre időszerűbb e drogok központi idegrendszerre gyakorolt hatásainak tudományos igényű vizsgálata. Miután a droghasználók jelentős része fogamzóképes fiatal nő, indokolt annak vizsgálata is, hogyan hatnak e drogok a fejlődő embrionális agyszövetre. Kísérleteink első csoportját fejlődő csirke embriókon végezzük, melynek során a striatum fejlődésének kritikus stádiumában szintetikus cathinonokat (mefedron, MBDB, MDPV) juttatunk tyúktojásokba, majd a posztembrionálisan proliferáló neuronokat a striatum subventricularis zónájában kvantitatívan elemezzük, jelzett nukleozid (BrdU) beépülése alapján. A változásokat mutató neuroncsoportok pontosabb azonosítása céljából vizsgáljuk e sejtek dopamin-transzporter, DARPP-32 (dopamin és cAMP regulált foszfoprotein) tartalmát, valamint a rajtuk végződő afferensek kémiai természetét (tirozin-hidroxiláz, kolin-acetil-transzferáz). További kérdésként vetődik fel, hogy e cathinonok által befolyásolt fejlődő neuronok melyik subpallialis telep származékai, ill. várhatóan milyen struktúrákba épülnek be (pallidalis, striatalis, septalis, olfactoros, stb.). Ennek vizsgálatát telep-specifikus géntranszkripciós faktorok immuncitokémiai ill. in situ hibridizációs kimutatásával tervezzük. A vizsgálatok második csoportja arra keres választ, hogy az embrionálisan adott (krónikus hatású) cathinonok hogyan befolyásolják a tanulás képességét, összehasonlítva a posztembrionálisan adott droghatással. Az alkalmazott kísérleti tesztek fiatal csirkék passzív elhárító tanulása, kiegészítve a motilitás és sztereotíp viselkedések vizsgálatával.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

A pályázatban megfogalmazott kérdések a következők: Milyen rövid és hosszú távú viselkedési hatásokat fejtenek ki a cathinonok családjába tartozó designer drogok? Hogyan befolyásolják ezek az ágensek a striatummal kapcsolatos tanulási képességet? Milyen hatásuk van ezeknek a drogoknak a fejlődő embrióra, különös tekintettel a neurogenezisre? Hogyan hatnak e szerek prenatalis alkalmazásban a korai tanulásra, összehasonlítva a posztnatalis alkalmazás következményeivel?

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

Az eredmények várhatóan elvezetnek az új party drogok rövid és hosszú távú hatásmechanizmusának jobb megértéséhez. Mivel ezen a területen ezidáig csak nagyon kevés állatkísérletes eredmény ill. humán vizsgálati adat áll rendelkezésre, így a pályázatban megfogalmazott kutatásból származó eredmények nagy jelentőséggel bírnának.

Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

A ”designer drogok" vagy „party drogok” alatt a szintetikus pszichoaktív szereknek egy olyan csoportját értjük, melyek szerkezetileg és farmakológiailag nagyon hasonlóak valamely kábítószerként nyilvántartott vegyülethez. Ezen szerek használata, az Internet adta lehetőségeknek köszönhetően, rohamosan terjed. 2010-ben a legnépszerűbb, újonnan elterjedt party drog a mefedron volt, és a közelmúltban számos, új rokon vegyület is megjelent az online piacon. A szintetikus party drogok rohamos léptékű terjedése miatt egyre sürgetőbbek azon vizsgálatok, melyek ezen szerek rövid és hosszú távú hatásait kutatják. Mivel nagy számban használnak fogamzóképes korban lévő nők is ilyen szereket, a magzatra való esetleges negatív hatás feltárása is nagy jelentőséggel bír. A pályázat fő célja, hogy adatokat nyerjünk a party drogok neurotoxikus, hosszú távú hatásairól, valamint az agyfejlődésre és a tanulási képességre gyakorolt hatásaikról.

Synthetic cathinones are known as 'designer drugs'. Given their widespread use, scientific investigation of the effect of these drugs on the CNS is getting evermore topical. Since drug users often are young women of generative age, investigation of the effects of these drugs on developing embryonic brain tissue is well justified. We will perform the first group of experiments on chicken embryos by injecting synthetic cathinones (mephedrone, MBDB, MDPV) into incubated chicken eggs in critical stages of striatal development. This will be followed by quantitative analysis of post-embryonic proliferating neurons in the subventricular zone of striatum, based on incorporation of the labeled nucleozide BrdU. For a more precise characterization of the neuronal groups showing alterations, we will investigate the dopamine transporter and DARPP-32 (dopamine and cAMP regulated phosphoprotein) contents, as well as the chemical nature of afferents terminating on them (tyrozine-hydroxylase, choline-acetyl-transferase). Further question to be studied is: which subpallial primordium gives rise to those developing neurons affected by the cathinones, and, which neuronal structures (pallidal, striatal, septal, olfactory, etc.) are they expected to be incorporated into. This will be investigated by the demonstration of anlage-specific gene transcription factors, using immunocytochemistry and in situ hybridization. A second group of experiments attempts to answer how cathinones administered in embryonic age can influence learning ability as compared to post-embryonic drug effects. Here we plan to use passive avoidance learning of young chicks and tests of motility and stereotypic behavior.

The questions of the proposed research are as follows: What are the short-term and long-term effects of the synthetic cathinones belongig to the group of designer drugs? Does the exposure to these drugs affect the learning ability in tasks mainly focused on the striatal brain regions? Do they have an effect on the developing embryo, especially on the neurogenesis? Does prenatal exposure to these drugs have any effect on early learning abilities?

What is the significance of the research?
Describe the new perspectives opened by the results achieved, including the scientific basics of potential societal applications. Please describe the unique strengths of your proposal in comparison to your domestic and international competitors in the given field.

This research project may lead to a better understanding of the short-term and long-term effects of the widely abused, novel designer drugs. Since there are only few experimental data on these drugs available yet, all the information obtained from the proposed research would be highly valuable.

The term of ’designer drugs’ or ’party drugs’ refers to a group of synthetic substances that are structurally and pharmacologically very similar to an illegal substance. The issues related to their toxicity and potential long term effects are a rapidly increasing problem, due to the ease of accessibility and the extensive marketing via the Internet. Although widely abused by teenagers and young adults, there is an important lack of epidemological, animal and clinical data concerning designer drugs. Taken to account, that these drugs are used mostly by young people being in generative age, special attention should be focused on the potential effect of prenatal exposure to these compounds.
The main objective of the present proposal is to gain information of neurotoxicity, long-term effects and developmental effects of these novel designer drugs.

Kutatásunkban ún. 'dizájner drogok' csoportjához tartozó, tudatmódosító szerként széles körben használt cathinonok fejlődő agyra kifejtett hatását vizsgáltuk különféle állatmodellek segítségével. Kimutattuk, hogy egérben a humán terhesség harmadik trimeszterének megfeleltethető posztnatális 7. napon egyszeri injekcióként adva az egyik széles körben használt cathinon, az MDPV (metilén-dioxi-pirovaleron) sejtpusztulást okoz a fejlődő agy bizonyos, a motivációval és addikcióval kapcsolatos területein. Ugyanez a drog vemhes nőstény egereken történő krónikus (több napon át napi egyszeri) alkalmazást követően az újszülött egerekben számos agyi és viselkedésbeli változást okoz. Több szerkezetileg rokon cathinon fiatal madarakon történt viselkedésvizsgálata során megállapítottuk, hogy bár ezen vegyületek szerkezeti hasonlósága nagy, mégis teljesen eltérő és sokszor kiszámíthatatlan viselkedési reakciót válthatnak ki. Eredményeink felhívják a figyelmet a manapság igen elterjedt dizájner drogok (más néven "partidrogok") fejlődő agyra gyakorolt veszélyeire, valamint ezen szerek hatásának kiszámíthatatlan természetére.
Results in English
We investigated the effects of the 'designer drogs' belonging to the cathinone family on the developing brain, using various animal models. Our results showed that acute injection of the frequently abused cathinone, MDPV (methylene-dioxy-pyrovalerone) causes cell death (apopotis) in 7-day-old mouse pups in several brain centers responsible for motivation and addiction. After chronic systemic administration (once a day for several days) to pregnant mice, MDPV caused visible alterations in the brain and behavior of the offspring. When we compared the effect of several cathinone agents on the behavior of young precocial birds, our results led to the conclusion that while those compounds have a similar chemical nature, they might have very different, and often unexpected effects on the behavior. Our results draw the attention to the potential harmful effects of the frequently abused designer cathinones on the developing brain.
