A mikrotubuláris rendszer (MT) érzékelő, integráló és koordináló jellege jelentős szerepet játszik sokrétű funkciónak ellátásában, melyeket komplex ultrastruktúrája tesz lehetővé. Ez a projekt a TPPP/p25 fehérje „neomorphic moonlighting” jellegére összpontosít, mely eltérő fiziológiás (MT/tubulin deacetilázok) és patológiás (alfa-szinuklein, SYN) kölcsönhatásaiból ered. A TPPP/p25 a MT stabilitását és dinamikáját befolyásolja a tubulin acetilációt elősegítő és a MT kötegelő aktivitása révén. A MT acetilációja szoros kapcsolatban áll mind az autofágiával, mind az aggreszóma képződéssel, melyek a sejtek védekező mechanizmusai. Korábban igazoltuk, hogy a TPPP/p25 over-expressziója aggreszóma képződéshez vezet, SYN-nel való asszociációja fehérje aggregációt indukál, továbbá a proteaszóma rendszer gátlása növeli az intracelluláris TPPP/p25 szintet. Azt feltételezzük, hogy a TPPP/p25 fiziológiás körülmények között az autofagoszómák érésében is szerepet játszik, így koordinálhatja a sejtvédő rendszerek funkcióit. A következő célokat tűzzük ki: i) jellemezzük a TPPP/p25 különböző hatásait az autofágia kialakulására SYN távol- és jelenlétében ii) jellemezzük a TPPP/p25 az autofágia és a proteoszóma rendszer közti kapcsolatra gyakorolt hatását; iii) feltárjuk a TPPP/p25 és a tubulin deacetilázok kölcsönös kapcsolatát az autofágiában és az aggreszóma képződésben. Ha a sejtet védő mechanizmusok nem működnek, a TPPP/p25-SYN komplex kialakulásának gátlása, illetve annak disszociációja szükségeltetik, így célunk iv) foldamer szerkezetek feltárása potenciális szinukleinopátiás gyógyszer tervezése, fejlesztése céljából.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

A projekt célja jellemezni a mikrotubuláris rendszer (MT) sokrétű funkcióinak és az ultrastrukturális szerveződésének kapcsolatát fiziológiás és patológiás körülmények között, a rendezetlen, agyspecifikus Tubulin Polymerization Promoting Protein (TPPP/p25) szerepére koncentrálva. Ezidáig sok információnk gyűlt össze a fehérje i) rendezetlen szerkezetéről, eltérő szegmenseinek kötő motívumairól ii) homológ és heterológ kölcsönhatásairól molekuláris és sejtszinten iii) jelenlétéről Parkinson-kórra és más szinukleinopátiákra jellemző patológiás zárványtestekben. A kérdés, amire ezen projekt keretein belül választ kívánunk adni, szorosan kapcsolódik a TPPP/p25 fehérjéhez, mely egy egyedülálló rendezetlen fehérje, elősegíti a tubulin acetilációt, a mikrotubulusokat pedig kötegekbe rendezi fiziológiás körülmények között. Célunk i) jellemezni a TPPP/p25 hatását az autofágia kialakulására HeLa, CHO és CG4 sejtekben, melyekben a TPPP/p25 expressziója tranziensen transzfekcióval, indukcióval vagy endogénen valósul meg; ii) jellemezni az autofágia és a proteoszóma rendszer közti kapcsolatot TPPP/p25 és/vagy SYN távol- és jelenlétében a MT rendszer különböző mértékű acetilációja esetén; iii) meghatározni a TPPP/p25 mikrotubulus kötegelő és acetilálót befolyásoló aktivitása által módosított MT ultrastuktura szerepét az autofágiában és aggreszóma képződésben. Patológiás körülmények között a TPPP/p25 az SYN-nel kolokalizál és együtt halmozódnak fel agyi zárványtestekben, ezért egy további kérdésre is választ tervezünk adni: hogyan akadályozható meg /indukálható a szinukleinopátiákra jellemző TPPP/p25-SYN komplex kialakulása/disszociációja.

A mikrotubuláris rendszer (MT), a citoszkeleton egyik fő alkotója, szorosan részt vesz mind fiziológiás, mind patológiás folyamatokban. Sokrétű funkcióit a makro- és mikromolekuláris kölcsönhatások, valamint a poszttranszlációs módosítások szabályozzák. A nemrég felfedezett, TPPP/p25-nek elnevezett mikrotubulus asszociált fehérje „neomorphic moonlighting” funkcióit fiziológiás és patológiás körülmények között különböző partnerekkel kialakuló kölcsönhatásai biztosítják. Jelen projektünkben megvizsgáljuk a TPPP/p25 szerepét a sejtvédő mechanizmusokban, melyek a felesleges fehérjéket távolítják el, amilyen például a patológiás TPPP/p25-SYN komplex. Ezt a feltételezést megerősítik előző adataink, miszerint a TPPP/p25 indukált tubulin acetiláció kulcs szerepet játszik az aggreszóma képződésben, a patológiás zárványtest képződéshez hasonló sejtvédő mechanizmusban. Az aggreszóma autofágiával lebontható. Az autofágia egy még nem kutatott terület, legalábbis a TPPP/p25 szerepét tekintve, ami egy új szereplő lehet ebben a komplex folyamatban. Továbbá a TPPP/p25 kapcsolatban áll a proteaszóma rendszerrel, melynek gátlása növeli az intracelluláris TPPP/p25 szintet, ami a patológiás TPPP/p25-SYN komplex kialakulásához vezet. Ezért a TPPP/p25 autofágiával kapcsolatos aktivitását tervezzük vizsgálni, ami koordinálhatja a fiziológiás fehérjeeltávolító/sejtet védő funkciókat; patológiás körülmények között a toxikus TPPP/p25-SYN komplex kialakulását peptidomimetikus foldamer(ek)kel tervezzük megelőzni, mely szinukleinopátiás betegségek elleni gyógyszertervezéshez fontos támpont lehet.

A kutatás összefoglalója, célkitűzései laikusok számára

A citoszkeleton fő alkotója, a mikrotubulus számos funkcióval rendelkezik, amit sokféle kölcsönhatás és poszttranszlációs módosítás révén kialakuló komplex szerkezete biztosít. A mikrotubulus asszociált TPPP/p25 a „neomorphic moonlighting” fehérjék prototípusa, mivel fiziológiás és patológiás körülmények között eltérő funkciókat lát el. A TPPP/p25 a mikrotubuláris rendszer dinamikáját és stabilitását befolyásolja, tubulin acetilációt elősegítő és a mikrotubulusokat keresztkötő aktivitása által. A TPPP/p25 ezen jellemzői alapvető szerepet játszanak az olyan fiziológiás folyamatokban, mint a gliális sejtek differenciációja, melyek az idegsejtek nyúlványait körülvevő mielinhüvely fő alkotói. A TPPP/p25 nem fiziológiás kifejeződése a központi idegrendszer károsodásához vezet, sclerosis multiplexet (mielinhüvely betegség), gliómát (agyi tumor) vagy szinukleinopátiát, mint pl. Parkinson-kórt (TPPP/p25-SYN felhalmozódás zárványtestekben) okozva. A pályázatban a TPPP/p25 szerepét tárjuk fel különböző fiziológiás sejtvédő mechanizmusokban, az autofágiában, proteaszóma rendszerben és az aggreszóma képződésben. Másrészt azt tervezzük, hogy patológiás körülmények között (betegség esetén) a toxikus TPPP/p25-SYN komplex kialakulását megelőzzük és/vagy megsemmisítjük a már kialakult patológiás komplexeket specifikus ún. foldamerek molekulákkal. A projekt eredményes megvalósítása értékes adatokkal fog szolgálni ezen sejtvédő folyamatok mechanizmusairól és résztvevőiről, ezenkívül új utakat nyithat a Parkinson-kór kezelésére szánt gyógyszerek tervezésében.

The sensing, integrating and coordinating features of the microtubule (MT) system are of great importance in its diverse functions achieved by its complex ultrastructural arrays. This project focuses on the neomorphic moonlighting feature of TPPP/p25 originated from its associations with physiological (MT and tubulin deacetylases) and pathological (alpha-synuclein, SYN) partners. TPPP/p25 modulates the dynamics and stability of the MT network by its tubulin acetylation enhancing and MT bundling activities. The MT network acetylation is intimately involved in both autophagy and aggresome formation, as cytoprotecting mechanisms. We have shown that the over-expression of TPPP/p25 results in aggresome formation, its association with SYN is prone to protein aggregation, and the inhibition of the proteasome system enhances the TPPP/p25 level. We hypothesize that at physiological conditions TPPP/p25 is also involved in the autophagosome maturation process, thus it could coordinate the physiological functions of these cytoprotective systems. Accordingly our objectives are i) to characterize the effect of the multifarious TPPP/p25 on the biogenesis of autophagy in the absence and presence of SYN; ii) to characterize the effect of TPPP/p25 on the interplay between the autophagy and proteasome machinery; iii) to explore the role of the mutual interaction of TPPP/p25 and tubulin deacetylases in the autophagy and aggresome formation. When the coordinated functions of the cytoprotective systems are failed, then the association/dissociation of the TPPP/p25-SYN complex has to be achieved, thus we plan iv) to explore foldamer structure which could be potential anti-synucleinopathy drugs.

What is the major research question?

This project aims to characterize the relationship of the ultrastructural organization and multiple functions of the microtubule (MT) system at physiological and pathological conditions focusing on the role of the disordered, brain specific Tubulin Polymerization Promoting Protein (TPPP/p25). Up to now a number of information has been accumulated concerning i) its disordered structure, binding motives in distinct segments of the proteins; ii) its heterologous and homologous interactions at molecular and cell levels; iii) its occurrence in pathological inclusions characteristic for Parkinson’s disease and other synucleinopathies. The question we plan to answer within the frame of this project is tightly coupled to TPPP/p25, a unique disordered protein, which displays tubulin acetylation enhancing and MT bundling activities at physiological conditions. Our objectives are i) to characterize the effect of TPPP/p25 on the biogenesis of autophagy in HeLa, CHO and CG4 cells expressing TPPP/p25 ectopically, in an inducible manner and endogenously, respectively; ii) to characterize the interrelationship between the autophagy and proteasome machinery in the absence and presence of TPPP/p25 and/or SYN at different level of the acetylation of the MT network; iii) to establish the role of the ultrastructure of MT network determined by TPPP/p25-derived microtubule bundling and acetylation in the autophagy and aggresome formation. In pathological conditions TPPP/p25 is co-enriched and co-localizes with SYN forming brain inclusions, thus we plan to answer the question as well: how to prevent the formation of the TPPP/p25-SYN complex characteristic for synucleinopathies.

The microtubule (MT) system, a major constituent of the cytoskeleton, is intimately involved in both physiological and pathological processes. Its multiple functions are directed by macro- and micromolecular interactions as well as post-translational modifications. A recently discovered Microtubule Associated Protein denoted TPPP/p25 displays neomorphic moonlighting function due to its interactions with distinct partners at physiological and pathological conditions. In the present project we will explore the role of TPPP/p25 in the cytoprotecting mechanisms which can eliminate unwanted proteins, for example, the pathological TPPP/p25-SYN complex. This hypothesis seems to be supported by our previous data, namely that the TPPP/p25-induced tubulin acetylation is a critical factor in aggresome formation, a cytoprotecting mechanism which mimics the pathological inclusion formation. The aggresome could be eliminated by autophagy. Autophagy is a not-yet studied area concerning the involvement of TPPP/p25 which could be a new player in this complex process. In addition, TPPP/p25 is related to the proteasome (UPS) function, the inhibition of which enhances the intracellular level of TPPP/p25 leading to the formation of pathological TPPP/p25-SYN complex. Therefore, we plan to explore the autophagy-related activity of TPPP/p25 which could coordinate the cytoprotecting/cleaning function at physiological condition; at pathological conditions the formation of the toxic TPPP/p25-SYN complex is planned to prevent by peptidomimetic foldamer(s), as potential anti-synucleinopathy drug(s).

The multiple functions of microtubules, key element of cytoskeleton, are achieved by their complex structure derived by multiple interactions and posttranslationalal modifications. The microtubule associated TPPP/p25 is a prototype of the neomorphic moonlighting proteins originated from its distinct functions at physiological and pathological conditions. TPPP/p25 modulates the dynamicas and stability of the microtubule system via its tubulin acetylation enhancing and MT cross-linking activities. These features of TPPP/p25 play crucial roles in physiological processes such as differentiation of glial cell which are the major constitutes of the myelin sheath around the axons of neurons. The non-physiological expression of TPPP/p25 causes destruction of the CNS resulting in multiple sclerosis (a myelin sheath disease), glioma (brain tumor) or synucleionopathies (TPPP/p25-SYN enrichment in inclusions) such as Parkinson’s disease. Our project aims to explore the role of TPPP/p25 in the physiological cytoprotective mechanisms, such as autophagy, proteasome function and aggresome formation. On the other hand, we also plan to overcome the pathological (disease) situation by preventing the toxic TPPP/p25-SYN complex formation and/or by destruction of the already formed pathological compexes by highly specific foldamer agents. The successful evaluation of the project will provide valuable data for the players and mechanisms of these protecting events and may open new way to design drugs for treatment of Parkinson’s disease.

Az eukarióta sejtek érzékelő, integráló és koordináló sajátosságait a citoszkeleton mikrotubuláris hálózata, annak komplex ultrastruktúrája biztosítja; fehérjékkel/enzimekkel való hetero-asszociációja, valamint poszttranszlációs módosulásai teszik lehetővé a többrétű funkciók ellátását. A jelen projekt a munkacsoport által azonosított fehérje, a TPPP/p25 fiziológiás és patológiás funkciójának vizsgálatára fókuszál. Fiziológiás körülmények között, a „moonlighting” TPPP/p25 modulálja a mikrotubuláris hálózat dinamikáját és stabilitását a mikrotubulusokat kötegelő és acetiláció növelő aktivitásai révén. Az optimális intracelluláris TPPP/p25 szint elengedhetetlen a fehérje fiziológiás funkciójának betöltéséhez, így például az oligodendrocita sejtek differenciációjához, melyek meghatározó komponensei a mielin hüvelynek. Patológiás körülmények között a TPPP/p25 az alpha-szinukleinnel való kölcsönhatása révén oligomereket/aggregátumokat hoz létre, mely jellemző markerei a PD-nek illetve az MSA-nak, következésképpen potenciális gyógyszercélpontok. Ugyanakkor, a TPPP/p25-által vezérelt mikrotubulus hiperacetiláció a kontrolláltalan sejtosztódás, azaz a rákos folyamatok ellen hat. Ezen eredményeink rámutatnak a TPPP/p25 két, látszólag ellentmondásos sajátságára, nevezetesen anti-mitotikus hatás, valamint aggregációt elősegítő hatás kifejtésére képes, ami egybevág azon klinikai megfigyeléssel, hogy a Parkinson-kórban szenvedő betegek kissebb valószínűséggel lesznek rákosak.

The sensing, integrating and coordinating features of the eukaryotic cells are achieved by the complex ultrastructural arrays and multifarious functions of the cytoskeletal microtubule network. Microtubules play crucial roles in both physiological and pathological processes manifesting itself by their decoration with proteins/enzymes as well as by posttranslational modifications. This project focuses on the “regulatory functions by day and pathological functions at night” of a new microtubule associated protein, the TPPP/p25. Physiologically, the moonlighting TPPP/p25 modulates the dynamics and stability of the microtubule network by bundling microtubules and enhancing the tubulin acetylation due to the inhibition of tubulin deacetylases. The optimal endogenous TPPP/p25 level is crucial for its physiological functions, for example, to the differentiation of oligodendrocytes, which are the major constituents of the myelin sheath. Pathologically, TPPP/p25 forms toxic oligomers/aggregates with alpha-synuclein in neurons and oligodendrocytes in Parkinson’s disease and Multiple System Atrophy, respectively; and their complex is a potential therapeutic drug target. TPPP/p25-derived microtubule hyperacetylation counteracts uncontrolled cell division. All these issues reveal the anti-mitotic and alpha-synuclein aggregation-promoting potency of TPPP/p25, that is consistent with the finding that Parkinson’s disease patients have reduced risk for cancerous diseases.
