A jelen pályázat célja organokaszkád reakciók fejlesztése és szintetikus alkalmazása királis építőelemek felépítésében, majd ezen elemek alkalmazása indolvázas alkaloidok mintázatfelismerésen alapuló és biomimetikus totálszintézisében. Az organokaszkád reakciók egy új irányvonalat képviselnek az aszimmetrikus organokatalízis területén, amelyek segítségével egy lépésben változatos és bonyolult szerkezetek alakíthatók ki regio-, enantio- és diasztereoszelektív módon. További előnyei a módszernek, hogy könnyen hozzáférhető kiindulási anyagokat használ, és a reakciók egyszerűen kivitelezhetőek, így bonyolult és többlépéses szintéziseket lehet kiváltani velük.
A pályázat további célja, hogy az építőelemek előállításához szükséges reakciókra olyan eljárást dolgozzunk ki, amelyek később egyszerűen beépíthetőek új, potenciális biológiai aktivitással rendelkező vegyületek szintézisének tervébe. Ilyen fejlesztés lenne például egy olyan hatékony allil oxidációs eljárás kidolgozása, ami nem igényli a mérgező szelén-dioxid alkalmazását.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

A tervezett kutatás során olyan kémiai reakciókat kívánunk vizsgálni, amelyek egy szintetikus lépésben több átalakítás elvégzésére is képesek. Az ilyen organokaszkád reakciók alkalmazhatóságát és előnyeit szeretnénk bemutatni a mintázatfelismerésen alapuló totálszintézisben. A kaszkádreakcióban előállított építőelemeket indolvázas alkaloidok totálszintéziséhez tervezzük felhasználni. Emellett azt is vizsgálni szeretnénk, hogy a módszer mennyire alkalmas akár több grammos méretben is biológiailag aktív anyagok szintézisére.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

A tervezett kutatás során kifejlesztett módszerek segítségével összetett vegyületek állíthatók elő regio-, enantio- és diasztereoszelektíven. Az organokatalitikus és organokaszkád reakciók ezért gyorsan és jelentősen kibővítik a kémiai teret, nagy változatosságot eredményeznek, így hatékony eszközei lehetnek a kvantitatív szerkzet-hatás összefüggés kutatásoknak.
Másrészről az előállított építőelemek segítségével megvalósítható totálszintézis új szintetikus stratégiát nyithat meg a biológiai aktivitást mutató indol alkaloidok előállítására. Így könnyen szerteágazó természetes vegyületekhez hasonló molekulakönyvtárak hozhatók létre, melyek jó kiindulópontként szolgálhatnak a gyógyszerkémiai kutatásokban.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

A gyógyszerhatóanyagok jelentős része szerves molekulákból áll. Az anyagok jelentős részét szerves kémiai szintézis során állítják elő, amely általában sok reakciólépést foglal magában. A tervezett kutatás során olyan folyamatok vizsgálatát tűztük ki célul, amelyek képesek egy lépés alatt bonyolult szerkezetek irányított létrehozására. Így nehezen előállítható anyagok válnak könnyen hozzáférhetővé, emellett csökken a keletkező hulladék mennyisége is.
A módszer másik előnye, hogy nem alkalmaz átmenetifémet, a katalizátorok minden esetben megújuló forrásból származó alapanyagokból készülnek. Ily módon az általunk vizsgált reakciók alkalmasak lehetnek egy fenntartható kémiai eljárás kidolgozására is, amely nagyban segít a gyógyszerhatóanyagok előállítását.

Summary of the research and its aims for experts

The aim of the present project is to develop organocascade reactions for the synthesis of chiral building blocks, and to demonstrate the application of these building blocks in biomimetic and pattern recognition type total synthesis of indole alkaloids. The organocascade reactions, emerged as a new trend in the field of asymmetric organocatalysis, are powerful tools in constructing complex molecules in regio-, enantio- and diastereoselective manner. Further advantages of this methodology are the use of cheap, easily accessable starting materials and the simple reaction conditions, therefore it can replace several steps from a long synthesis plan.
The further aim of the project is to modify reactions required for the construction of the building blocks in order to establish a methodologies that are easily applicable in the synthesis of new, potentially bioactive molecules. An example for this development is the modification of the allylic oxidation which omits the use of highly toxic selene dioxid.

What is the major research question?
Describe here briefly the problem to be solved by the research, the starting hypothesis, and the questions addressed by the experiments.

In this project, we plan to investigate chemical reactions in which several transformations are realized in one synthetic step. We would like to show the applicability and advantages of these organocascade reactions in the pattern recognition type total synthesis of indole alkaloids. Moreover, we plan to realize the synthesis plans in a multigram scale for biologically active molecules.

What is the significance of the research?
Describe the new perspectives opened by the results achieved, including the scientific basics of potential societal applications. Please describe the unique strengths of your proposal in comparison to your domestic and international competitors in the given field.

The methodologies planned to be developed during this research are to be used for the construction of complex and regio-, enantio- and diastereoselective molecules. Therefore, the organocatalytic and organocascade reactions expand the chemical space in a rapid and efficient manner, producing diversity which can be exploited in the quantitative structure-reactivity relationship studies.
On the other hand, the total synthesis with these new building blocks could open a new synthetic strategy to the biologically active indole alkaloids. Moreover, we can synthesize diverse natural-product-like molecular libraries, which are good starting point for medical drug research.

Most of the currently used drugs are organic compounds. Greater part of these compounds are created by means of chemical synthesis, which usually consists of several distinct steps. Our research aims to investigate chemical methods which are capable to carry several transformations in one synthetic step, creating complex structures. As a result, hard to obtain compounds become easily accessible, and the amount of waste is decreased.
Other benefit of the process is that it does not require the use of metals, the catalysts are prepared from renewable resources. In this way, the reactions can be used for the planning of a sustainable chemical processes, which facilitate drug production.

Kutatásaink során királis építőelemeket állítottunk elő újonnan kifejlesztett enantioszelektív organokatalitikus módszerekkel, továbbá az így létrehozott építőelemet Aspidosperma alkaloidok szintézisére használtuk fel. Kutatómunkánk első részeként diszubsztituált királis tietánok szintézist valósítottuk meg sztereoszelektív organokatalitikus sztereoablációval, valamint anti, anti-polipropionát sztereotriádokat állítottunk elő méretnövelhető módon bifunkcionális organokatalízist alkalmazó deszimmetrizációval. Kutatásaink során iteratív Michael-Michael-aldol organokaszkád reakció segítségével telítetlen aldehidekből kiindulva királis ciklohexanon származékokat képeztünk, telítettlen ketonokból pedig Michael-Michael reakciósorral nyílt láncú sztereotriádokat hoztunk létre enantio-, és diasztereoszeleketív módon. Kutatómunkánk második részeként Aspidosperma alkaloidok sztereoszelektív előállítására egy divergens és kaszkád reakciókon alapuló szintézis utat terveztük. Ezen megközelítés kulcs intermedierjét (amelyet a Stork-féle szintézis központi elemének analogonja) több grammos méretben állítottuk elő. Ebből a triciklusos építőelemből megvalósítottuk a (-)-minovincine, (-)-aspidospermidine szintézisét, valamint a (-)-aspidofractinine formális szintézisét.
Results in English
During our research programme, we developed new enantioselective organocatalytic methods to synthesise chiral building blocks, which have been used in the synthesis of Aspidosperman type alkaloids. In the first part of our research, we assembled disubstituted thietanes with stereoselective organocatalytic stereoablation, moreover, anti, anti-polypropionate stereotriads were synthesised via bifunctional organocatalytic desymmetrisation. Using iterative Michael-Michael-aldol cascade reaction, cyclohexene derivatives were synthesised from unsaturated aldehydes. Furthermore, we extended this methodology to unsaturated ketones and assembled acyclic stereotriads via diastereoselective, iterative Michael-Michael reactions. In the second part of our research programme, we planned a divergent and cascade based synthesis of Aspidosperman type alkaloids. We designed an advanced intermediate, based on Stork's classical approach, and synthesised it in multigram scale. From this tricyclic intermediate, we performed a concise total synthesis of (-)-minovincine, (-)-aspidospermidine and formal synthesis of (-)-aspidofractinine.
