A Candida parapsilosis (Cp) a harmadik leggyakoribb invazív candidiasist okozó élesztőgomba. Annak ellenére, hogy a faj előfordulási gyakorisága világszerte növekvő tendenciát mutat, a patogén által kiváltott védekezési mechanizmusokról csak kevés ismeretünk van. Kutatócsoportunk elsők között kezdett foglalkozni a Cp által kiváltott immunválasz tanulmányozásával. Eddigi eredményeink fontos különbségekre derítettek fényt a Cp, és a gombás fertőzések leggyakoribb modellorganizmusa, a C. albicans által indukált védekezési mechanizmusokat illetően. Jelenlegi célunk a Cp elleni immunválasz molekuláris mechanizmusainak vizsgálata, amelyeknek megértése elengedhetetlen az új és hatékonyabb antifungális terápiák kifejlesztéséhez. Célunk a Cp által kiváltott természetes és adaptív immunitás vizsgálata. Munkánk során vizsgálni fogjuk az immunsejtek és a Cp között létrejövő molekuláris kölcsönhatásokat, különös tekintettel a patogén immunológiai felismerésére és az általa indukált inflammaszóma-aktivációra. A Cp által kiváltott immunválasz molekuláris hátterét különböző gén knock-out egér modellek segítségével is vizsgáljuk. Kutatási projektünk kivitelezését kooperációban tervezzünk a Mihai G. Netea által Nijmegenben (Hollandia) vezetett kutatócsoporttal, amely a legkiválóbb laboratórium Európában a molekuláris immunológia területén. Ez az együttműködés egyedülálló lehetőséget biztosít számunkra szaktudásunk bővítéséhez az antifungális immunológia területén, valamint lehetővé teszi, hogy a legújabb technikákat saját laboratóriumunkban is alkalmazzuk. A nemzetközi együttműködés hozzájárul hallgatóink képzéséhez is, segítve őket, hogy a tudományterület kiváló kutatóivá váljanak.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Kutatási tervünk fő célkitűzése a Candida parapsilosis által kiváltott immunválasz molekuláris hátterének vizsgálata. Míg a C. albicans elleni immunválaszt az elmúlt évek folyamán átfogóan tanulmányozták, a C. parapsilosis által kiváltott védekezési mechanizmusokról csak kevés információnk van. Kutatási projektünk legfontosabb kérdései a következők: (1) a C. parapsilosis immunológiai felismerésében részt vevő mintázatfelismerő recetorok azonosítása, (2) annak tisztázása, hogy képes-e (és ha igen, hogyan) a C. parapsilosis aktiválni az inflammaszómát (azt a makromolekuláris komplexet, amely az IL-1b gyulladásos citokin érett formájának kialakításáért felelős), (3) az adaptív Th sejtes válasz szabályozásának vizsgálata C. parapsilosis fertőzés folyamán. Kísérleteink során minden esetben össze kívánjuk hasonlítani a C. parapsilosis és a C. albicans által kiváltott immunválasz molekuláris hátterét, előzetes eredményeink ugyanis azt mutatják, hogy fontos különbségek vannak a két faj által kiváltott immunválaszban. Az egyik legfontosabb eredményünk azt bizonyítja, hogy jelentős különbség van a C. parapsilosis és a C. albicans által indukált Th17 differenciációban. Továbbá, legújabb eredményeink azt mutatják, hogy a C. parapsilosis sokkal alacsonyabb IL-1b termelést indukál, mint a C. albicans, ami arra utalhat, hogy az inflammaszóma aktivációja sokkal kevésbé hatékony C. parapsilosis fertőzés folyamán. A jelen kutatási projekt keretein belül zajló kísérletek hozzájárulhatnak majd az említett különbségek molekuláris hátterének tisztázásához, illetve a C. parapsilosis elleni immunitás teljesebb megértéséhez.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Számos globális felmérés jelzi, hogy az utóbbi időben világszerte nő a gombás fertőzések száma. A Candida fajok által okozott invazív fertőzések magas morbiditással és mortalitással járnak, ezért komoly egészségügyi problémát jelentenek. Noha egyre több új antifungális szer áll rendelkezésünkre, ezek hatékonysága gyakran limitált a rezisztens törzsek terjedése miatt. Másrészt, az antifungális terápia többnyire igen költséges és gyakran jár komoly mellékhatásokkal, ezért nagy szükségünk van hatékonyabb gombaellenes gyógyszerekre. A gazda-patogén kölcsönhatások megértése a gombás fertőzések folyamán igen fontos az új terápiák kifejlesztése szempontjából. Ennek ellenére, noha a C. albicans elleni immunválasz feltérképezésében nagy előrelépés történt az utóbbi időben, a nem-albicans Candida fajok által kiváltott védekezési mechanizmusokról igen kevés információ áll rendelkezésünkre. Fő célkitűzésünk a C. parapsilosis – amely a C. albicans-tól eltekintve az egyik klinikai szempontból legfontosabb Candida faj – által kiváltott immunválasz molekuláris hátterének megértése. Noha a két faj azonos genus-ba tartozik, morfológiájukban, gyógyszer rezisztenciájukban és virulenciájukban is jelentősen különböznek. Az utóbbi időben általunk és mások által folytatott tanulmányok kimutatták, hogy jelentős különbségek vannak a C. albicans és a C. parapsilosis elleni immunválaszban. Fő célunk ezeknek a különbségeknek a kimerítő vizsgálata a legújabb rendelkezésre álló laboratóriumi technikák segítségével. Fontos kiemelni, hogy a Candida fajok elleni immunválasz megértése nemcsak az antifungális immunitásról szerzett tudásunkat fogja növelni, hanem hozzájárulhat majd az általános immunológiai ismereteink bővítéséhez is. A jelen kutatási projekt keretein belül olyan alapvető kérdéseket is vizsgálni fogunk, mint a gyulladásos válasz szabályozása, gazda-patogén kölcsönhatások, illetve a kórokozók immunválaszt befolyásoló képessége. A kutatások során nyert információ elvezethet minket más fertőzéses megbetegedések molekuláris hátterének a megértéséhez is, végső soron pedig hozzájárulhat a hatékonyabb gyógyszeres terápiák kifejlesztéséhez.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

Az utóbbi évtizedek folyamán világszerte megnövekedett a gombafertőzések száma. A fertőzések többségéért opportunista patogének felelősek, amelyek az immunrendszer legyengülése (pl. immunszuppresszív terápia, rákos megbetegedés vagy AIDS) esetén okoznak megbetegedést. A Candida fajok, amelyek enyhe bőrfertőzéseket, valamint súlyos szisztémás megbetegedéseket (invazív candidiasis) egyaránt okozhatnak, az opportunista gombafertőzések leggyakoribb okozói. Az új antifungális terápiák bevezetése ellenére a Candida fertőzéseket magas, 20-30 %-os halálozási arány jellemzi, ami jól jelzi az új gombaellenes szerek, illetve kombinált terápiák kifejlesztésének szükségességét. Noha az invazív candidiasis leggyakoribb okozója a C. albicans, újabban jelentősen megnövekedett a nem-albicans Candida fajok előfordulási gyakorisága. A C. parapsilosis különösen az újszülöttekre jelent veszélyt, és gyakran hozható összefüggésbe csecsemőkori halálozásokkal. Ennek ellenére, noha a C. albicans elleni immunválaszról jelentősen bővültek ismereteink az utóbbi évek folyamán, a C. parapsilosis által kiváltott védekezési mechanizmusokról még kevés információ áll rendelkezésünkre. Csoportunk korábban bizonyította, hogy jelentős különbségek vannak a C. albicans és a C. parapsilosis által kiváltott immunválaszban. Jelen pályázat keretein belül a C. albicans és a C. parapsilosis elleni immunválasz molekuláris hátterét vizsgáljuk. A C. parapsilosis által kiváltott immunválasz pontosabb megértése fényt deríthet arra, hogy miért fogékonyabbak az újszülöttek a gomba által okozott fertőzésre, valamint hozzájárulhat a jövőben a hatékonyabb gombaellenes terápiák kidolgozásához is.

Summary of the research and its aims for experts
Describe the major aims of the research for experts.

Candida parapsilosis (Cp) is the third most common species causing invasive candidiasis and its prevalence has been increasing worldwide. However, little is known about the host defense mechanisms induced by this pathogen. Our research group was among the first ones investigating the immune responses against Cp. Importantly, our recent findings revealed crucial differences between the host responses induced by Cp and the prototypic fungal pathogen C. albicans. Our current purpose is to investigate the molecular mechanisms underlying Cp-specific immunity. Importantly, understanding the host response upon Cp is absolute necessary for the development of better antifungal therapies. Our aims include the investigation of both innate and adaptive immune responses against Cp. We are going to study the molecular interactions between immune cells (monocytes and macrophages) and Cp, with special focus on immune recognition and the activation of inflammasome. Furthermore, the molecular background of immune responses during Cp infection is going to be studied with the help of different gene knock-out mouse strains. Importantly, our whole research project will be carried out in cooperation with the laboratory of Mihai G. Netea (Nijmegen, The Netherlands), one of the most excellent teams in Europe in the field of molecular immunology. This cooperation is a unique opportunity for us to improve our expertise in antifungal immunology and establish new state-of-the-art techniques in our laboratory. In the long term, the international cooperation will also contribute to the training of talented students, helping them to become prominent researchers of the field.

What is the major research question?

The main aim of our research project is to investigate the molecular background of immune responses upon Candida parapsilosis infection. While the immune responses against C. albicans has been studied extensively during recent years, we have little information about the host defense against C. parapsilosis. The most important aims of our research project are the following: (1) to investigate which pattern recognition receptors are involved in the immune sensing of C. parapsilosis, (2) to examine whether and how C. parapsilosis is able to induce the activation of inflammasome – the macromolecular complex responsible for the processing of the pro-inflammatory cytokine IL-1b, (3) to investigate how adaptive T helper cell responses are modulated during C. parapsilosis infection. Importantly, we wish to compare the molecular background of host defense mechanisms induced by C. parapsilosis and C. albicans, as our previous findings indicate that there are crucial differences between the immune responses evoked by the two species. One key finding was that there is a marked difference in the capacity of C. parapsilosis and C. albicans to induce Th17-differentiation. Furthermore, recent results show that C. parapsilosis is a much weaker inducer of IL-1b than C. albicans, implicating that the activation of inflammasome may be less effective upon infection with C. parapsilosis. The experiments conducted in the frames of this research project will allow us to find the molecular explanation of these differences as well as will help to fully elucidate C. parapsilosis-specific immunity.

Global surveillance programs indicate that the number of fungal infections has been increasing worldwide. Invasive infections caused by Candida species are associated with high morbidity and mortality, and therefore represent an important public health problem. Although there are several new antifungal drugs available to treat Candida infections, their efficiency is still limited due to the emergence of resistant strains. On the other hand, antifungal therapies represent a high cost and may have severe side effects, underlining the need for better antifungal treatments. Understanding the host-pathogen interactions during fungal infections is crucial for the development of more efficient antifungal treatments. However, although there has been a great progress in the understanding of the immune response against the prototypic fungal pathogen C. albicans, we have little information about host defense mechanisms against non-albicans Candida species. Our main purpose is to study the molecular background of host defense mechanisms against C. parapsilosis, which is one of the most clinically important Candida species apart from C. albicans. Importantly, although related, the two species are different in their cellular morphology, antifungal drug susceptibility and virulence. Recently, several studies (conducted by us and others) have indicated that there are crucial differences between the immune response induced by C. albicans and C. parapsilosis. Our main research aim is to extensively characterize these differences, using the new, state-of-the-art methods. Understanding the immune response against Candida species will not only increase our knowledge about antifungal immunity, but it will contribute to understanding of immunology in general. In the frames of this research project, we are going to gain answers to questions regarding basic inflammatory responses, interactions between microbes and the host and the capacity of pathogens to modulate the host’s defense mechanisms. This information is highly valuable because it may lead to the understanding of the molecular basis of other infectious diseases as well, and, eventually, to the development of more efficient therapies.

Summary and aims of the research for the public

During the past few decades, there has been a great increase in the number of fungal infections worldwide. The majority of fungal infections are caused by opportunistic species, which only cause disease when the immune system of the host is weakened (e. g. in the case of immunosuppressive therapy, cancer or AIDS). Candida species are the leading cause of fungal infections, being responsible for both mild mucosal infections and severe systemic disease (invasive candidiasis). Despite the novel antifungal therapies, Candida infections have a high mortality rate of 20 to 30 %, highlighting the need for more efficient antifungal drugs or combined therapies. Furthermore, although the majority of candidiasis cases is due to C. albicans, the prevalence of non-albicans species has largely increased over the past decades. C. parapsilosis is of special concern in critically ill newborns and young children, and is often associated with neonatal mortality. However, although our knowledge about the immune responses during C. albicans infection has increased significantly, little information is available about C. parapsilosis-specific immunity. Recently, we have shown that there are important differences between the host response to C. albicans and C. parapsilosis infection. In the frames of this research project, we are going to investigate and compare the molecular background of host defense mechanisms induced by C. albicans and C. parapsilosis. The better understanding of immune response against C. parapsilosis may reveal why newborns are highly susceptible to C. parapsilosis infection, and hopefully contribute the development of more efficient antifungal therapies in the future.

A kutatási projekt keretein belül a fertőzés során kialakuló immunválaszt vizsgáltuk Cp fertőzések során. Megállapítottuk, hogy az antifungális immunválasz egyik kulcsfontosságú komponensének az inflamaszómának szerepe van a Cp elleni immunválaszban is. A veszély jelek hiánya azonban az inflamaszóma kevésbé hatékony és lassabb aktiválódását okozza Cp fertőzések során. Az immunválasz hatékonyabb vizsgálata érdekében kifejlesztettünk egy újszülött egereket alkalmazó fertőzési modellt. Kimutattuk, hogy a gomba sejtfalának N-kötött mannozilációja alapvetően képes befolyásolni a patogén virulenciáját és a gazda által kiváltott veleszületett immunválaszt. Megállapítottul továbbá, hogy ellentétben a C. albican-szal a C. parapsilosis elleni immunválasz során a béta glükán felismerő C tipusú lektin receptornak nincs alapvető szerepe a hatékony gazdaválasz kialakításában. Kifejlesztettünk egy alternatív fertőzési modelt ahol ecetmuslica vad tipusú és toll útvonalban mutáns egyedeit felhasználva vizsgáltuk a az N mannozilációban károsdott Cp mutánsok immunológiai felismerését és a gazda által kiváltott immunválaszt.

Candida parapsilosis (Cp) is the third most common species causing invasive candidiasis and its prevalence has been increasing worldwide. However, little is known about the host defense mechanisms induced by this pathogen. In this project, we investigated the host response upon Cp infection. Our results suggest that similar mechanisms are involved in inflammasome activation upon C. parapsilosis and C. albicans infection, but the slower production of “danger signals” leads to delayed inflammasome activation following C. parapsilosis stimulation. . Additionally, we developed a neonatal model of disseminated candidiasis for C. parapsilosis infections. Our results demonstrate that N-linked mannosylation significantly affects the virulence and the specific steps of innate immune response against this pathogen in the wild-type mouse model of systemic candidiasis. Interestingly, in contrast to C. albicans, our observations disclose that the recognition of wild-type C. parapsilosis is independent of the Dectin-1 receptor. As a “mini host” we used Drosophila toll pathway mutants and we demonstrated that using this alternative, invertebrate model is appropriate for the investigation of fungal virulence, as we were able to effectively reproduce our previous results: the N-mannosylation deficient och1∆/∆ strain has decreased virulence in vivo.
