Az aszkorbát (C-vitamin) az emberek számára esszenciális, növényi eredetű vitamin, amely számos sejtvédő és egyéb funkcióval rendelkezik. A gazdasági jelentőség miatt az aszkorbát bioszintézisét és élettani jelentőségét intenzíven kutatják, de ennek ellenére számos alapvető kérdés még tisztázásra vár.
Jelen OTKA-NN project célja, hogy az együttműködő fél, Dr. Alisdair R. Fernie (Max-Planck Institue of Molecular Plant Physiology, Potsdam-Golm) csoportjával közösen i) tanulmányozzuk az aszkorbát bioszintézisének szabályozását és a különböző bioszintézis-utak jelentőségét ii) megvizsgáljuk az általunk előállított aszkorbát-hiányos Chlamydomonas zöldalga mutánsokban az oxidatív stressz hatására kialakuló adaptációs folyamatokat, valamint tanulmányozzuk az aszkorbátnak a biohidrogén-termelés megindításában betöltött szerepét iii) aszkorbát-transzportereket azonosítsunk és jellemezzünk. Emellett hozzájárulunk az együttműködő fél által beadott DFG pályázat (“Ascorbate-signalling mediated influence on plastidial metabolism and photosynthesis”) sikerességéhez azáltal, hogy az általuk előállított aszkorbát-túltermelő mutánsok fotoszintézisét jellemezzük. A projekt alatt két PhD disszertáció elkészítése várható, és emellett a résztvevők tapasztalatszerzési lehetőséget kapnak a potsdami Max-Planck Intézetben.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

A projekt alapkérdései, céljai:
I. A különböző aszkorbát-bioszintézis utak élettani jelentőségének feltárása. Előzetes kísérleteink és a publikált eredmények azt mutatják, hogy az alternatív bioszintézis utak hozzájárulása stresszkörülmények és a növényi egyedfejlődés során változik. A kérdés megválaszolásához génexpressziós vizsgálatokat, fotoszintézis méréseket végzünk, valamint Dr. Alisdair Fernie csoportjával együttműködésben metabolomika segítségével meghatározzuk a bioszintézis-utak közbülső termékeit.
II. A aszkorbát bioszintézisének jellemzése Chlamydomonasban és az oxidatív stressz hatására kialakuló adaptációs folyamatok vizsgálata. Emellett VTC2 mutánsainkat felhasználva megvizsgáljuk az aszkorbát nemfotokémiai kioltásban (NPQ, Allorent et al., 2013) valamint a biohidrogén-termelés megindításában betöltött szerepét (Nagy et al. 2012).
III. Az aszkorbát transzporterek azonosítása a kloroplasztiszban és esetleg más sejtorganellumokban. Koexpressziós analízisből kiindulva, a lehetséges transzportereket T-DNS vonalakon, klorofill-a fluoreszcencia segítségével teszteljük. Ezt nem-vizes fázisú frakcionálás követi, majd izotóp jelölést felhasználva meghatározzuk a különböző sejtorganellumokban az aszkorbát mennyiségét.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

A növények aszkorbáttartalmának emelése a növényi sejtben betöltött számos védőfunkciója miatt gazdaságilag igen jelentős, és emellett az étrendünk legnagyobb részt növényi eredetű aszkorbátot tartalmaz. Bízunk benne, hogy eredményeink jelentősen hozzájárulnak az aszkorbát bioszintézisének és sejten belüli transzportjának jobb megismeréséhez és hosszú távon hozzá tudunk járulni magas aszkorbáttartalmú fajták előállításához, amelyek feltehetően jobb stressztűrő képességgel is rendelkeznek.
A Chlamydomonas nevű zöldalgában az aszkorbát bioszintézise és annak szabályzása igen kevéssé ismert. A Chlamydomonas reinhardtii nemcsak alapkutatás szempontjából jelentős, hanem a biohidrogén-termelő képessége miatt is intenzíven kutatott faj. Korábbi eredményeink azt mutatják, hogy az aszkorbát szerepet játszhat a hidrogéntermelés megindításában és az általunk előállított aszkorbát-deficiens mutánsok segítségével szeretnénk ezt a kérdést tisztázni.
Az aszkorbát bioszintézisének és szerepeinek vizsgálata ígéretes kutatási terület. Ebben a témában nagyszámú (kb. 170), gyakran magas impakt faktorú (6 feletti) folyóiratcikk jelenik meg minden évben, amelyek idézettsége is jelentős. A Chlamydomonas H2 termelésének kutatási területén is hasonló a helyzet, így az ezeken a területeken publikált eredmények jelentős érdeklődésre tarthatnak számot.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

Munkacsoportunkkal a potsdami Max-Planck Kutatóintézettel folytatott nemzetközi együttműködésre támaszkodva, a metabolomika, a biofizika és a molekuláris biológia széles eszköztárát felhasználva az aszkorbát (C-vitamin) bioszintézisét és élettani szerepét fogjuk vizsgálni. Kutatásaink eredményeképpen lehetővé válhat a zöldségfélék és gyümölcsök aszkorbát-tartalmának fokozása, így eredményeink közvetlenül hozzájárulhatnak az emberi egészség megőrzéséhez és ezen élelmiszerek jobb eltarthatóságához. Az aszkorbát-tartalom növelése révén a növények stressztűrő képessége és így produktivitásuk fokozható. Emellett korábbi eredményeink már bizonyították, hogy aszkorbát hozzáadásával a megújuló energia kutatásban intenzíven használt Chlamydomonas reinhardtii nevű zöldalga hidrogéntermelése jelentősen, többszörösére fokozható. Célkitűzéseink között szerepel a serkentés hatásmechanizmusának feltárása és a hidrogéntermelés fokozásának optimalizálása, amely környezetvédelmi szempontból is rendkívül jelentős lehet.

Ascorbate (Asc, also called vitamin C) is an essential vitamin for humans of plant origin, which fulfills various protective and other roles in the cells. Because of economic interest in increasing the Asc contents of plants and thereby their productivity, the metabolism and physiological roles of Asc are under intensive research, but still there are fundamental processes that are very poorly understood or perfectly unknown.
The primary objectives of this OTKA-NN project proposal is to target, in collaboration with the group of Dr. Alisdair Fernie at the Max-Planck Institute of Molecular Plant Physiology in Potsdam-Golm i) the regulation of Asc biosynthesis and the roles of the different biosynthesis pathways in plants, ii) oxidative stress responses of our newly-generated Asc-deficient Chlamydomonas transformants and the role of Asc in the initiation of biohydrogen production iii) Asc transport within plant cell and in particular, in the chloroplasts. Moreover, we will contribute to the project entitled “Ascorbate-signalling mediated influence on plastidial metabolism and photosynthesis” led by Dr. Alisdair Fernie, by characterizing the photosynthetic activities of Asc-overproducing mutants. Based this project, two PhD theses will be completed and the participants of this project will have the possibility to gain experience in the Max-Planck Institute of Molecular Plant Physiology in Potsdam-Golm.

What is the major research question?
Describe here briefly the problem to be solved by the research, the starting hypothesis, and the questions addressed by the experiments.

Our objectives:
I. Revealing the physiological significance of the various ascorbate biosynthesis pathways. Preliminary data and published results suggest that the contribution of the alternative ascorbate biosynthesis pathways is increased under stress conditions and that it also varies during plant development. In order to tackle this question, metabolite profiling, gene expression analysis and photosynthetic measurements will be carried out on Arabidopsis mutants affected in ascorbate biosynthesis.
II. Elucidating the route ascorbate biosynthesis in the green alga Chlamydomonas reinhardtii and investigating how the lack of ascorbate affects growth and photosynthesis under oxidative stress conditions and which defense mechanisms come into play when ascorbate is limiting. Moreover, using our VTC2 transformants, we will be able to shed light on the involvement of ascorbate in non-photochemical quenching (NPQ, Allorent et al., 2013) and in the role of ascorbate in the initiation of biohydrogen production (Nagy et al., 2012).
III. Identification of ascorbate transporters in higher plants. We would like to use co-expression analysis based on genes of the main ascorbate biosynthesis pathway. Once candidate genes are found, Arabidopsis T-DNA insertion lines will be screened using chl a fluorescence. This will be followed by non-aqueous fractionation and isotope feeding experiments to determine alterations in the organellar and cytosolic poolsizes.

What is the significance of the research?

Increasing the Asc (vitamin C) contents of plants is of high economic importance because of the various roles of Asc, and also when considering that plants are the primary sources of this vitamin in the human diet. We trust that our results will clarify important questions regarding the regulation of Asc biosynthesis and its transport within the plant cell, more specifically in the chloroplast. In the long run, our results may contribute to the development of crop varieties with higher Asc contents, with possibly enhanced productivity.
In the green alga, Chlamydomonas reinhardtii, very little is known about Asc biosynthesis and regulation. This alga is important not only for fundamental research, but it is being very intensively investigated because it is capable of producing significant amount of H2 gas. Our earlier results suggest that Asc may have a regulatory role in the initiation of biohydrogen production and with the help of our newly generated Asc-deficient Chlamydomonas transformants we aim at clarifying this issue.
Studying Asc biosynthesis and its roles in plants and green algae is a promising area of research. Articles in journals of high impact factor (above 6) regularly appear. Since it is an area of great interest (about 170 papers published per year), the most prominent papers receive large number of citations. The area of H2 gas production of Chlamydomonas is similar; Therefore the reward of making a significant progress in these research areas is definitely high.

Summary and aims of the research for the public
Describe here the major aims of the research for an audience with average background information. This summary is especially important for NRDI Office in order to inform decision-makers, media, and others.

Based on international cooperation with the Max-Planck Institute of Molecular Plant Physiology (Potsdam), we will investigate the biosynthesis and physiological roles of ascorbate (vitamin C), using various tools of metabolomics, biophysics and molecular biology. Our research will contribute to the generation of varieties with elevated ascorbate content, and thereby to the preservation of human health and better storage properties of fruits and vegetables. By increasing the ascorbate contents of crops, our research will also contribute to an enhanced stress tolerance and thereby improved plant productivity. Besides, our earlier results have shown that in the intensively studied green alga Chlamydomonas reinhardtii, the rate of biohydrogen production can be significantly increased by supplying the cultures with ascorbate. We aim at understanding the mechanism of this enhancement and further improving algal biohydrogen production; Thereby we expect that our results will be of significance in the field of renewable energy research as well.

Az aszkorbát (Asc, más néven C-vitamin) nélkülözhetetlen a növények és állatok sejtfunkcióihoz. Az Asc reaktív oxigénfajták semlegesítésében betöltött szerepe jól ismert, azonban még számos kérdés vár tisztázásra az Asc metabolizmusával, élettani szerepeivel és transzport folyamataival kapcsolatban, amelyek rendkívül fontosak a magas Asc-tartalmú növényfajták nemesítése szempontjából. Megállapítottuk, hogy a Smirnoff-Wheeler útvonal felelős az Asc-bioszintézisért, és a korábban javasolt alternatív Asc-bioszintézis útvonalak szerepe elhanyagolható a növényekben. Megállapítottuk azt is, hogy önmagában a reaktív oxigénformák nem vezetnek Asc felhalmozódásához magasabb rendű növényekben. A Chlamydomonas reinhardtii zöldalga fontos modellszervezet a különféle sejtfolyamatok tanulmányozására, és nagy biotechnológiai alkalmazási potenciállal rendelkezik. Bebizonyítottuk, hogy a zöldalgák csak környezeti stresszhatásra növelik Asc-termelésüket, a magasabb rendű növényekétől eltérő szabályozási mechanizmusok révén. A növényekhez képest egy másik fő különbség az, hogy a zöldalgákban a fotoprotektív nemfotokémiai kioltási mechanizmusok nem igényelnek Asc-ot. Bebizonyítottuk azt is, hogy az Asc fontos szerepet játszik a zöldalgák kénmegvonás által indukált biohidrogén-termelésében. Az Asc szállítását illetően megállapítottuk, hogy a kloroplasztisz burkolómembránja egy eddig ismeretlen Asc transzportert tartalmaz. Zöldalgákban pedig három feltételezett Asc-transzportert azonosítottunk.
Results in English
Ascorbate (Asc, also known as vitamin C) is essential for the cellular functions of plants and animals. The role of Asc in the scavenging of reactive oxygen species is well known, but many questions remain regarding the metabolism, physiological roles and transport processes of Asc, which are extremely important for breeding plant species with elevated Asc contents. We found that the Smirnoff-Wheeler pathway is responsible for Asc biosynthesis and the roles of previously proposed alternative Asc biosynthesis pathways in plants are negligible. It was also found that reactive oxygen species alone do not lead to the accumulation of Asc in higher plants. The green alga Chlamydomonas reinhardtii is an important model organism for studying various cellular processes and has great potential for biotechnological applications. We have demonstrated that green algae boost their Asc production only under environmental stress via regulatory mechanisms that are distinct from those of seed plants. Another major difference from the plants is that the photoprotective non-photochemical quenching mechanisms in green algae do not require Asc. We have also demonstrated that Asc plays a very important role in the sulphur deprivation-induced biohydrogen production of green algae. With regard to Asc transport, we have found that the chloroplast envelope membrane contains an Asc transporter that has not been previously known. In green algae, three putative Asc transporters were identified.
