A pályázat során az ecetmuslica (Drosophila melanogaster) hematopoietikus kompartmentumaiban zajló vérsejtképződés szabályozását tanulmányoztuk. Olyan modern módszereket alkalmaztunk a vérsejt differenciálódás, illetve a kompartmentumok integritásának vizsgálatára, mint az in vivo fotokonverzió, a kettős sejtvonaljelölés, a vérsejtek in vivo követése, illetve a gépi tanulás. Jellemeztük a korábban azonosított hdc gén mutációjának fenotípusát, kifejeződését a fejlődés során, valamint azonosítottuk az általa kódolt fehérje lehetséges együttműködő partnereit. Eredményeink szerint a Hdc alapvető szerepet játszik a lárva központi nyirokszervében található hematopoietikus őssejtek fenntartásában. A lárva szesszilis vérsejtjeinek dinamikáját elemezve megállapítottuk, hogy a központi nyirokszervvel ellentétben a szesszilis szövet nem valódi vérsejtképző niche. Kettős sejtvonaljelölés használatával azonosítottuk azokat a szignál transzdukciós utakat, melyek sejt-autonóm, illetve nem-sejt-autonóm módon szerepet játszanak a szesszilis szövet integritásának fenntartásában, illetve a lamellociták differenciálódásának szabályozásában. Kifejlesztettünk egy ex vivo módszert, mellyel lehetővé vált a plazmatociták lamellocitákká történő transzdifferenciálódásának valós idejű követése. Gépi tanulás alkalmazásával nagyszámú sejt sorsát vizsgáltuk a transzdifferenciálódás során. Ezen kísérletek segítséget nyújthatnak a transzdifferenciálódás molekuláris szintű szabályozásának megértéséhez.

During the last four years, we studied the regulation of blood cell differentiation in the hematopoietic compartments of the fruit fly, Drosophila melanogaster. In our studies, we applied state of the art methods, such as in vivo photoconversion, double cell lineage tracing, in vivo tracking of blood cells and machine learning to approach the regulation of blood cell differentiation and of the integrity of the hematopoietic compartments. We characterized the expression domain, the developmental regulation, the mutant phenotype and the interacting partners of the previously identified hdc gene. We found that Hdc plays an essential role in the stem cell maintenance in the lymph gland, one of the hematopoietic compartments of the larva. We analyzed the blood cell dynamics in another larval hematopoietic compartment, the sessile tissue. We found that unlike the lymph gland, the sessile tissue is not a real hemocyte niche. Using double cell lineage tracing, we identified the signaling pathways that contribute to the regulation of sessile tissue integrity and lamellocyte differentiation in either a cell-autonomous or a non-cell-autonomous manner. We developed a new method for culturing plasmatocytes ex vivo, and to monitor their differentiation into lamellocytes in real time. The application of deep learning enabled a high-throughput characterization of the transdifferentiation event, which may serve as a basis for the molecular analysis of this differentiation process.
