A humán nm23 géncsalád tagjai nukleozid-difoszfát kinázokat (NDPK) kódolnak. A 10 humán NM23 izoforma (H1-H10) közül az NM23-H1 volt az elsőként azonosított metasztázis inhibitor. Az intenzív biokémiai kutatások ellenére sem ismertek azok a mechanizmusok, amelyek a metasztázis szuppresszor hatás kifejtéséhez hozzájárulnak. Célunk ezeknek a mechanizmusoknak, valamint a NDPK-ok expresszióját szabályozó jelátviteli folyamatoknak a megismerése.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Caenorhabditis elegans modell rendszeren és invazív emlőkarcinóma sejtvonalon végzett előzetes kísérleteink azt mutatják, hogy a nematóda NM23 homológ NDK-1 a sejtmigrációt valamint az apoptotikus testek bekebelezését a citoszkeleton átrendezésén keresztül, a CED-10/Rac-tól downstream befolyásolja. Célunk, hogy meghatározzuk azokat a kölcsönható fehérjéket, amelyek segítségével az NDK-1/NM23 az apoptotikus testek bekebelezésére, valamint a citoszkeleton szerveződésére hat. Kiemelten vizsgáljuk az NDK-1/DYN-1 interakciót. Az NDK-1 apoptotikus testek bekebelezését segítő funkcióját humán mieloblast-monocita eredetű sejtvonalakon is teszteljük. Ezen kívül olyan géneket kívánunk azonosítani, amelyek az NDK-1 kifejeződését befolyásolják: NDK-1::GFP transzgenikus nematódák EMS mutagenezisét követően megváltozott expressziót mutató vonalakat keresnénk, amelyekben az NDK-1-et szabályozó faktorok szenvedtek mutációt. Ezáltal lehetővé válik az upstream szabályozó faktorok meghatározása.
A projekt egy másik részében az NM23 fehérjével, mint esetleges biomarkerrel foglalkozunk. E pályázat keretében colorectalis daganatos betegek szérumában határoznánk meg az NM23 fehérje szintjét. Fő kérdésünk, hogy a szérum NM23 szint ebben a tumor típusban korrelál-e a tumor előrehaladott stádiumával (vagyis a metasztázis megjelenésével).

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Az nm23 géncsalád tagjai és az interakciós partnereik potenciális gyógyszer célpontok. Számos ráktípusban (pl. melanóma valamint mellrák) az NM23 alacsony expressziója a rossz prognózis jele. Az NM23 biológiai funkciójának (apoptózisban, sejtmigrációban betöltött szerepének), valamint szabályozásának megismerése hozzájárulhat az áttétképződés folyamatának jobb megértéséhez, távlatilag esetleges új terápiás alkalmazás kifejlesztéséhez.
Vizsgálatainkban arra is választ keresünk, hogy használható-e a szérum NM23 fehérje biomarkerként.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

Ez a projekt az elsőként felfedezett metasztázis inhibitor gén, az nm23 biológiai funkcióinak megértésével foglalkozik. A metasztázis inhibitorok az áttétképződés bizonyos szakaszaiban játszanak szerepet, a metasztázist gátolják, míg a primer tumorok kialakulására nincsenek hatással. Az NM23 funkcióit fonálféreg genetikai modellrendszeren illetve sejtvonalakon vizsgáljuk. Eddigi eredményeink azt mutatják, hogy az NM23 féreg homológja, az NDK-1 befolyásolja a sejtvándorlást valamint a programozott sejthalállal elpusztult sejtek eltávolítását segíti. Mindkét említett folyamat kulcsfontosságú lehet az áttétképződés megakadályozásában, ezért célunk, hogy olyan NDK-1/NM23-mal kölcsönható fehérjéket határozzunk meg, amelyek ezekben a folyamatokban részt vesznek. Máig sem ismert, hogy mellrák esetében milyen faktor gátolja az NM23 expresszióját. Ezért céljaink közé tartozik azoknak a géneknek a felderítése, amelyek az NDK-1/NM23 szabályozását végzik.
Vizsgálatainkban arra is választ keresünk, hogy használható-e a szérum NM23 fehérje biomarkerként.

Members of the nm23 gene family encode nucleoside diphosphate kinases (NDPKs). Amongst the ten human nm23 paralogs (nm23-H1-H10) nm23-H1 was the first identified metastasis inhibitor gene. Despite its medical significance, the molecular mechanisms by which NM23 suppresses the metastatic potential of cancer cells are still unclear. Our goal is to better understand these mechanisms, in addition we also intend to uncover upstream factors which regulate the expression of NDPKs.

What is the major research question?
Describe here briefly the problem to be solved by the research, the starting hypothesis, and the questions addressed by the experiments.

Our preliminary experiments performed on the tractable genetic model organism Caenorhabditis elegans and invasive breast carcinoma cell lines show that the nematode NM23 homolog NDK-1 influences cell migration and apoptotic engulfment through rearrangement of the cytoskeleton, downstream of CED-10/Rac. Our aim is to determine proteins interacting with NDK-1/NM23 to better understand, how (in which complexes) NDK-1 is able to reorganize the cytoskeleton and promote apoptotic engulfment. We especially focus on the NDK-1/DYN-1 interaction. We will also test how NDK-1 promotes apoptotic engulfment in human mieloblast-monocyte cell lines. Moreover, we intend to identify factors, which regulate the expression of NDK-1. Nematodes transgenic for NDK-1::GFP will be mutagenized by EMS, then worms with altered GFP expression will be screened and selected. The selected nematodes carry mutations in upstream regulators of NDK-1/NM23.
In another part of the project we ask whether serum NM23 protein can be used as a biomarker. We would determine the level of NM23 in the serum of patients suffering in colorectal carcinoma (CRC). Our main question is whether elevated NM23 in the serum correlates with advanced stage disease (e.g. presence of metastases).

What is the significance of the research?

Members of the NM23 family and their interaction partners are potential drug targets. In certain tumour types (such as breast cancer or melanoma) reduced NM23 level is associated with bad prognosis. Exploring biological functions of NM23, such as its role in cell migration and apoptosis, and uncovering its upstream regulation could contribute to better understanding of the metastatic processes. In the future, perhaps these potential results could lead to new therapeutic approaches.
We also intend to examine whether serum NM23 can be used as a biomarker in colorectal carcinoma.

Summary and aims of the research for the public

The focus of this project is understanding the biological functions of the first identified metastasis inhibitor, nm23. Metastasis inhibitors suppress metastasis, they are involved in differents steps of the metastatic cascade, but they do not interfere with primary tumour growth. We examine the functions of NM23 using the nematode as a genetic model organism and we also work on different cell lines. Preliminary results show that the nematode counterpart of NM23, NDK-1 influences cell migration and promotes removal of dying cells. Both processes mentioned above may be crucial in metastasis inhibition, therefore our goal is to identify proteins interacting with NDK-1 in these processes. Today it is still unknown, what kind of factor suppresses the expression of NM23 in breast cancer. Therefore we also intend to uncover genes that regulate ("switch on or off")ndk-1/nm23.
We also intend to examine whether serum NM23 can be used as a biomarker in colorectal carcinoma.

Különböző modell rendszerekben végzett kísérleteink azt mutatják, hogy az NDK-1 apoptotikus sejtek eltakarítását serkentő szerepe evolúciósan konzervált, mivel egér és humán homológjai (NM23-M1 valamint NME1, azaz NM23-H1) egyaránt a fagocitózist segítő esszenciális faktorok. A dynamin/NDPK kooperáció globális szerepet játszik a fagoszóma kialakulásában. Az el nem takarított apoptotikus sejtek jelenlétét összefüggésbe hozták gyulladással járó betegségekkel. Az apoptotikus testek elégtelen eltávolítása következtében kialakuló gyulladás hozzájárulhat a tumorprogresszióhoz. Mivel az NME1 és homológjai segítik az apoptotikus sejtek fagocitózisát a dynamin GTPáz GTP ellátásán keresztül, hozzájárulnak a gyulladás elkerüléséhez, ezáltal a tumorprogresszió ellen hathatnak. Vagyis a fenti mechanizmus új kontextusba helyezi az NME1 metasztázis inhibitor aktivitását. Az NDK-1 evolúciósan konzervált interakciós partnereinek meghatározása lehetőséget ad új NM23-mal kölcsönható fehérjék azonosítására is. Egy menekítő NDK-1::GFP konstrukciót hordozó transzgenikus törzs segítségével co-IP-t, majd tömegspektrometriát végeztünk, és a glutamát-dehidrogenázt (GDH-1) azonosítottuk, mint új NDK-1-gyel kölcsönható fehérjét. A GDH enzim, amelyet a GTP negatívan szabályoz, glutamátból alfa-ketoglutarátot állít elő, amely a TCA ciklusban használódik fel. Ez az eredmény az NDPK-k szerepét tehát összekötheti a citromsav ciklussal. A GDH-1/NDK-1 interakciót tovább vizsgáljuk.
Results in English
Our experiments in three different models show that the positive effect exerted on apoptotic clearance by NDK-1 is evolutionarily conserved, as mouse and human NDPK homologues, NM23-M1 and NME1 (NM23-H1), respectively, are essential factors promoting phagocytosis. Our study provides evidence that the Dynamin/NDPK cooperation is a global mechanism in phagosome formation. The presence of uncleared apoptotic cells have been associated with different diseases that involve inflammation, autoimmunity and cancer. Inflammation that arises as a result of insufficient removal of apoptotic cells, damages organs and might also favour tumour growth and progression. Thus, we can propose a novel explanation for the metastasis inhibitor activity of NME1: as this group I NDPK promotes phagocytosis of apoptotic cells through fuelling Dynamin GTPases by GTP, it contributes to avoid inflammation. By identifying evolutionarily conserved interaction partners of NDK-1, we have the opportunity to detect novel NM23 interactors. We performed co-IP analyses coupled to mass spectrometry using a transgenic strain carrying a rescuing translational NDK-1::GFP construct, and obtained glutamate dehydrogenase (GDH-1) as the most promising candidate. GDH, which provides alpha-ketoglutarate for the TCA cycle by converting glutamate, is negatively regulated by GTP. These results might link NDPKs to TCA cycle function. This novel interaction will be further investigated.
