A pályázatban hazai és nemzetközi kutatócsoportokkal együttműködve vizsgáltunk olyan koordinációs (réz, cink, nikkel, ródium és ruténium tartalmú) vegyületeket amelyek szerkezeti sajátságainak feltárása a biológiai hatás megértése szempontjából fontos. Ehhez két nagyműszeres technikát, a szilárd kristályok esetén nagy szerkezeti pontosságot adó egykristály röntgendiffrakciót (SXRD) valamint a paramágneses vegyületek esetén igen érzékeny elektronspin rezonancia (ESR) spektroszkópiát alkalmaztuk. A paramágneses fémkomplexek esetén a két módszer együttes alkalmazásával a komplexek szerkezetét szilárd és oldat fázisban is meghatároztuk. Három biológiai jelentőségű tématerületen összesen 51 kristályszerkezetet határoztunk meg és körül-belül 20 paramágneses komplex oldatbeli ESR vizsgálatát végeztük el. Az egyik terület a kelátterápiában (fém-felhalmozódással járó betegségek kezelésében) használható vegyületek szerkezetvizsgálata réz(II) és cink(II) ionokkal. Egy másik területen rákellenes hatóanyagok céljára kifejlesztett új réz(II)-tioszemikarbazon, valamint „zongora-szék” elrendeződésű Ru(II) és Rh(III)-komplexek szerkezeti elemzését végeztük el. A harmadik terület a metalloenzimeket utánzó fém-komplexek szerkezetvizsgálata, ahol a szerkezet és a katalitikus aktivitás közötti összefüggések feltárása volt a cél.
Results in English
In cooperation with national and international research groups we have been investigated the structural properties of coordination (copper, zinc, nickel, rhodium and ruthenium) compounds with biological relevance to reveal structure-function relationship. Two high performance instrumental techniques have been used: single crystal X-ray diffraction (SXRD) which results in very precise structural data of single crystals and electron paramagnetic resonance spectroscopy (EPR) which is a very sensitive technique for paramagnetic complexes. In case of paramagnetic complexes both methods have been used resulting in structural data both in solid and solution phases. Working in three different biologically relevant research fields, we have been determined the crystal structure of 51 compounds and performed EPR study of more than 20 paramagnetic compounds. One of the research fields was the investigation of chelator ligands in the development of chelate therapy. Several anticancer ligands have also been studied with copper(II) and with „piano-stool” Ru(II) and Rh(III). In the third topic new metalloenzyme model complexes have been studied where the goal was to explore the relationship between the structure and the catalytic activity.
