A termesztett gombák, mint a csiperke (Agaricus bisporus), laskagomba (Pleurotus ostreatus) és shiitake (Lentinula edodes) zöldpenészes megbetegedéséért a Trichoderma fonalasgomba-nemzetség bizonyos képviselői tehetők felelőssé, melyek világszerte egyre gyakrabban okoznak komoly gazdasági károkat a gombatermesztésben. Magyarországon a csiperke zöldpenészes betegségét a Nyugat-Európából már ismert Trichoderma aggressivum f. europaeum faj okozza, míg a laskagomba esetében a nemzetség más képviselői, a Trichoderma pleuroti és a Trichoderma pleuroticola okozzák a problémát - utóbbi emellett a csiperke és shiitake termesztésében is előfordul.
A kutatás fő céljai
- a termesztett gombák, és a gombatermesztésben zöldpenészt okozó három Trichoderma faj fluoreszcensen jelölt törzsei közötti kölcsönhatások vizsgálata a mikoparazitizmus zöldpenész kialakulásában feltételezett szerepének tanulmányozása céljából,
- a peptaibolok termelése útján megvalósuló antibiózis zöldpenész kialakulásában feltételezett szerepének vizsgálata,
- a három, gombatermesztésben zöldpenészt okozó Trichoderma faj, a T. aggressivum f. aggressivum (csiperke zöldpenész), T. pleuroti (laskagomba zöldpenész) és T. pleuroticola (a laskagomba és a csiperke termesztésében egyaránt előforduló zöldpenész) összehasonlító genomszekvencia-elemzése, valamint
- különböző Trichoderma – termesztett gomba kölcsönhatások összehasonlító transzkriptomikai elemzése.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

A szakirodalomban a mai napig nagyon kevés információ lelhető fel a termesztett gombák zöldpenészes megbetegedését okozó Trichoderma fajok patogenitási mechanizmusairól. Az élőhelyért és tápanyagokért folytatott kompetíció mellett a feltételezett mechanizmusok között szerepel a sejtfalbontó enzimek (kitinázok, glukanázok, proteázok) hatása révén megvalósuló mikoparazitizmus és az antifungális vegyületek termelése révén megvalósuló antibiózis. A zöldpenészes betegség genomikai hátteréről és transzkriptomikai jellegzetességeiről sem áll rendelkezésre szakirodalmi információ. A tervezett projekt keretein belül mélyebb betekintést kívánunk szerezni a zöldpenészt okozó Trichoderma fajok patomechanizmusáról, szubsztrátspecificitásáról és gazdaspektrumáról.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. A gfp-jelölt Trichoderma törzsekkel végzett kísérletek eredményei megmutatják, hogy a nemzetség biológiai védekezésben alkalmazható törzseinek növénypatogén gombákra gyakorolt hatásához hasonlóan a közvetlen mikoparazitizmus részét képezi-e a Trichoderma termesztett gombák megtámadása során bevetett fegyverarzenáljának.
A gombapatogén Trichoderma törzsek által termelt peptaibolok termesztett gombákra gyakorolt hatásának vizsgálatából kapott eredmények mélyebb betekintést nyújtanak az antibiózis zöldpenész kialakulásában betöltött szerepébe.
A termesztett gombákra káros Trichoderma fajok genomszekvenciájának ártalmatlan Trichoderma fajok genomszekvenciáival történő összehasonlítása feltárja a zöldpenészt okozó Trichoderma fajok specialitásait, információkkal szolgál evolúciós fejlődésükről és magyarázatot nyújt az ökológiai elterjedésükben és gazdaspektrumukban mutatkozó különbségekre.
Az összehasonlító transzkriptomikai elemzés lehetővé teszi a termesztett gombák megtámadásában feltehetően szerepet játszó Trichoderma gének, valamint a védekezési reakcióban feltehetően szerepet játszó termesztett gomba gének azonosítását, ezáltal megteremti a zöldpenésszel szembeni védekezés új célpontjainak azonosítását és új stratégiáinak kidolgozását.
A kutatásból származó eredmények nívós nemzetközi tudományos szakfolyóiratokban kerülnek publikálásra, illetve hazai és nemzetközi konferenciákon szóbeli előadások és poszterek formájában bemutatásra. Különös figyelmet fordítunk az eredmények gombatermesztők körében történő terjesztésére.

Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

A Trichoderma néven ismert penészgombacsoport egyes képviselői okozzák a termesztett gombák (pl. csiperke, laskagomba) úgynevezett zöldpenészes megbetegedését, mely világszerte komoly gazdasági károkat okoz a gombatermesztőknek. Bizonyos Trichoderma penészek csak a csiperkét, vagy csak a laskagombát támadják, míg mások mindkét gomba termesztésében képesek zöldpenészes betegséget okozni. Kutatásunk célja a termesztett gombák és a Trichoderma penészek közötti kölcsönhatások vizsgálata annak érdekében, hogy tisztázzuk a parazitizmusnak, valamint a gátló vegyületek Trichoderma általi termelésének szerepét a zöldpenész kialakulásában. Feltérképezzük ezen túl három, zöldpenészt okozó Trichoderma-típus teljes genetikai állományát és azonosítjuk a különböző Trichoderma – termesztett gomba kölcsönhatások során működő géneket.

Certain members of the filamentous fungal genus Trichoderma are responsible for the green mould disease of commercially produced mushrooms like the white button mushroom (Agaricus bisporus), oyster mushroom (Pleurotus ostreatus) and shiitake (Lentinula edodes), and are more and more frequently causing severe economical damages to mushroom production around the globe. The causal agent of the green mould disease in the case of A. bisporus in Hungary is Trichoderma aggressivum f. europaeum, a species already known from Western Europe, while the causal agents of the problem in the case of P. ostreatus are other species of the genus, Trichoderma pleuroti and Trichoderma pleuroticola - the latter being also reported to occur in the cultivation of white button mushroom and shiitake.
The main aims of the research are
- the examination of the interactions between cultivated mushrooms and fluorescently labelled Trichoderma strains of the three species causing green mould disease in mushroom cultivation in order to clear the hypothesized role of mycoparasitism in green mould disease development,
- the examination of the hypothesized role of antibiosis by the production of peptaibols during the development of green mould disease,
- comparative genome sequence analysis of the three Trichoderma species causing green mould diseases in mushroom cultivation: T. aggressivum f. aggressivum (causing green mould of Agaricus bisporus), T. pleuroti (causing green mould of P. ostreatus) and T. pleuroticola (causing green mould in both P. ostreatus and A. bisporus cultivation), and
- comparative transcriptome analysis of various Trichoderma - cultivated mushroom interactions.

What is the major research question?

Informations available in the literature about the pathogenicity mechanisms of Trichoderma species causing green mould diseases of cultivated mushrooms are still very limited. Besides the competition for space and nutrients, hypothesized mechanisms include mycoparasitism realized by the action of cell wall degrading enzymes (chitinases, glucanases, proteases) and antibiosis by the production of antifungal substances. In the literature there is also a lack of information about the genomic background and transcriptomic features of the green mould disease development. Within the frames of the project we aim to get deeper insight into the patomechanism, substrate specificity and host range of Trichoderma species causing green mould disease.

What is the significance of the research?

Results from the experiments with gfp-labelled Trichoderma strains will show whether – similarly to the case of biocontrol strains of the genus on plant pathogenic fungi – direct mycoparasitism is a part of the weaponry of Trichoderma during its attack on cultivated mushrooms.
Results from studying the effect of peptaibols produced by mushroom pathogenic Trichoderma strains on cultivated mushrooms will provide more insight into the role of antibiosis during the green mould disease development.
The comparative genome analysis of Trichoderma species harmful and harmless to cultivated mushrooms will show the specialties of green mould-causing Trichoderma species, provide information about their evolutionary development and reveal potential reasons for the differences between green mould-causing Trichoderma species in their ecological distribution and host range.
The comparative transcriptome analysis will reveal Trichoderma genes possibly involved in the attack of the cultivated mushrooms and mushroom genes possibly involved in the defence reaction, thereby enabling the identification of potential targets and elaboration of new strategies for green mould control.
Results deriving from the research will be published in the form of articles in highly-ranked international scientific journals as well as distributed among the participants of national and international conferences in the form of oral and poster presentations. Special attention will be drawn to the dissemination of the results among mushroom producers.

Certain members of the mould group named Trichoderma are causing the so called green mould disease of commercially produced mushrooms (e.g. champignons, oyster mushroom), which causes severe economical losses to mushroom producers world-wide. Certain Trichoderma moulds are attacking only champignons or only oyster mushroom, while others cause green mould disease to both of them. Our research aims the examination of interactions between cultivated mushrooms and Trichoderma moulds to clear the role of parasitism and the production of inhibitory compounds by Trichoderma during the green mould development. Furthermore, the entire genetic material of three Trichoderma types causing green mould disease will be mapped and the genes functioning during various Trichoderma - cultivated mushroom interactions identified.

Elvégeztük 3, gombatermesztésben zöldpenészt okozó Trichoderma faj (T. aggressivum - csiperke zöldpenész, T. pleuroti és T. pleuroticola - laskagomba zöldpenész) teljes genomjának szekvenálását (3 NGS platformon), összeszerelését, funkcionális annotációját, és zöldpenész-specifikus géneket azonosítottunk. Prediktáltuk az NRPS-gének peptaibol-termékeit és összehasonlítottuk a T. aggressivum és T. pleuroti által termelt hypomurocinokkal és tripleurinokkal. aMD-szimulációkkal vizsgáltuk egy általunk felfedezett tripleurin 3D-szerkezetét és érési tulajdonságait. Trichoderma törzsek termesztett gombák elleni agresszivitásának vizsgálatára optimazilált, képelemzésen alapuló módszer alkalmazásával megállapítottuk, hogy gfp-jelölt törzsek valószínűleg nem alkalmasak a zöldpenész-fertőzések gombatermesztésben történő nyomonkövetésére. A T. pleuroti és laskagomba között kivitelezett konfrontációs tesztekből izolált RNS transzkriptomikai elemzése a kölcsönhatás során differenciálisan expresszálódó Trichoderma gének azonosításához vezetett. Az észak-amerikai csiperke zöldpenész-típust (T. aggressivum f. aggressivum) elsőként detektáltuk Európában, és csiperketermesztésből elsőként tudósítottunk a T. decipiens fehérpenészes kártételéről. További zöldpenészt okozó ágenseket azonosítottunk a nemzetség Harzianum kládjából. Zöldpenész elleni védekezés céljára ígéretes baktériumokat szelektáltunk és teszteltünk növénykamrára optimalizált kísérleti gombatermesztésben és gombatermesztő üzemben.
Results in English
Full genomes of 3 Trichoderma spp. causing green mould disease in mushroom cultivation (Agaricus green mould T. aggressivum, Pleurotus green moulds T. pleuroti and T. pleuroticola), were sequenced on 3 NGS platforms, assembled, functionally annotated and green mould-specific genes identified. Peptaibol products of NRPS-encoding genes were predicted and compared with hypomurocins and tripleurins produced by T. aggressivum and T. pleuroti, respectively. 3D structure and folding properties of a newly discovered tripleurin were examined with aMD simulations. Results from an optimized image analysis-based method for the evaluation of the aggressivity of Trichoderma strains towards cultivated mushrooms revealed that gfp-labeled strains may not be applicable for monitoring green mould infections in mushroom cultivation. Confrontation assays performed with T. pleuroti and P. ostreatus for the purposes of RNA isolation and transcriptome analysis revealed Trichoderma genes differentially expressed during interaction with P. ostreatus. The North-American Agaricus green mould biotype T. aggressivum f. aggressivum was firstly detected from Europe, while T. decipiens was firstly reported to cause white mould in Agaricus production. Further green mould agents were identified in the Harzianum clade of genus Trichoderma. Bacteria promising for green mould control were selected and tested both in experimental, plant growth chamber-based mushroom cultivation as well as in a mushroom farm.
