Édesvízi és tengeri kutatások egyaránt kimutatták, hogy a fitoplankton által megkötött szén jelentős része (~50%) a „mikrobiális hurkon” (microbial loop) halad át, melyben a heterotróf baktériumok asszimilálják az oldott szerves szénvegyületeket és juttatják el a heterotróf és mixotróf egysejtűekhez illetve a magasabb trofitási szintekhez. E kitüntetett anyagforgalmi jelentőség ellenére vízi ökoszisztémákban a heterotróf bakterioplankton működéséről csak nagyon kevés információval rendelkezünk. Különösen szembetűnő ez a hiány a litorális rendszerek esetében, ahol a szerves szénvegyületek döntő hányada a fitoplankton helyett a vízi makrofitonoktól származik és ahol a bakterioplankton anyagforgalmi jelentősége is alig ismert. A javasolt kutatási téma a heterotróf bakterioplankton makrofitonokkal borított sekély tavakban betöltött szerepének megismerése révén szándékozik ezt az űrt kitölteni. Célunk annak a megértése, hogy a különböző eredetű (emerz, szubmerz és úszó levelű makrofitonok) és összetételű szerves szénvegyületek hogyan befolyásolják a bakterioplankton abundanciáját, produkcióját és metabolizmusát a litorális ökoszisztémákban. A heterotróf bakteriális produkció és a vízi elsődleges szervesanyag termelés (fitoplankton + makrofitonok) közötti összefüggés feltárása a projekt másik fő feladata. Célunk továbbá annak megismerése, hogy a vízben oldott szerves szénvegyületek eredete és összetétele hogyan befolyásolja a bakterioplankton funkcionális csoportjainak összetételét, valamint hogy az eltérő szervesanyag kínálat milyen hatást gyakorol a bakterioplankton diverzitására.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

A vízben oldott szerves szénvegyületek kulcsszerepet töltenek be a vízi ökoszisztémákban, elsődleges tápanyagforrásként szolgálva a vízben élő baktériumok többsége számára. A litorális rendszerekben, ahol az oldott szerves szénvegyületek döntő része nem a planktonikus elsődleges termelőktől (fitoplankton), hanem a vízi makrofitonoktól származik, a bakterioplankton anyagforgalmi jelentősége csak alig ismert. Irodalmi adatok alapján a vízi makrofiton borítottság növekedése nem csak a vízben oldott szervesanyagok mennyiségének növekedését jelenti, hanem a szervesanyag készlet összetételének és biológiai hozzáférhetőségének változását is, a környezeti tényezőktől és a vízi makrofitonok típusától függően. A makrofiton borítottság növekedése emiatt nem csak a baktériumok mennyiségének és aktivitásának növekedését eredményezheti, hanem a bakterioplankton diverzitását is jelentően megváltoztatja. Mindemellett nem hagyhatjuk figyelmen kívül a vízi makrofitonok környezetmódosító hatását sem, amely a vízalatti fényviszonyok megváltoztatása révén a fotoheterotróf baktériumok számára kedvező feltételeket teremt. Az előzőek figyelembevételével a tervezett kutatás fő kérdése, hogy a különböző eredetű (emerz, szubmerz és úszó levelű makrofitonok) és összetételű szerves szénvegyületek hogyan befolyásolják a bakterioplankton szerepét és diverzitását (taxonómiai, funkcionális és metabolikus összetétel) a litorális ökoszisztémákban. Célunk továbbá annak megismerése, hogy a vízi makrofitonok szelektív fényabszorpciója hogyan befolyásolja a bakterioplankton struktúráját (fotoautotróf/fotoheterotróf/heterotróf életmód).

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. A litorális régióban zajló metabolikus folyamatok összetettsége jelentősen megnehezíti a vízi ökológiai rendszerek működésének megértését. A javasolt terepi és laboratóriumi munka nagymértékben hozzájárul a litorális rendszerek szénkörforgalmának, különösen azok mikrobiológiai aspektusainak megértéséhez. Rávilágít arra, hogy ezek az itt zajló folyamatok mi módon függenek a makrofitonok típusától, bomlásától, illetve más biotikus és abiotikus tényezőktől. A tervezett kutatások révén megismerjük a bakterioplankton produkciójának jelentőségét a litorális rendszerekben. Emellett a projekt eredményei tisztázni fogják a baktériumok, az elsődleges termelés és a szervesanyag készlet közötti viszonyokkal kapcsolatos ellentmondásos nézeteket. A baktériumok számára a szerves anyag hozzáférhetőségét elsősorban annak kémiai természete határozza meg, utóbbi pedig nagymértékben függ a makrofiton közösséget alkotó fajoktól és/vagy növénytípusoktól. A várható eredmények segítenek megérteni a nagyobb tavak parti övében zajló anyagcsere folyamatok jelentőségét, emellett a tervezett kutatások nélkülözhetetlen információval szolgálhatnak vizeink ökológiai modellezéséhez is.

A vízben lebegő baktériumok (bakterioplankton) kicsiny méretük ellenére a vízi táplálékhálózat fő komponensei, meghatározó szerepet töltenek be a szén biogeokémiai ciklusában. A bakterioplanktonnak és a szabályzó környezeti tényezőknek megismerése elengedhetetlen a vízi ökoszisztémák működésének megértéséhez. A bakterioplankton a vízben oldott szerves szénvegyületeket használja fel anyagcseréje során. Utóbbiak származhatnak a vízben lebegő algáktól (fitoplankton) csakúgy, mint a vízinövényektől, illetve egyéb külső forrásból (pl. szennyvíz bevezetésből). Amíg azokban a vízterekben (mély tavak, tengerek), ahol az algák képezik az elsődleges termelés döntő hányadát, a bakterioplankton anyagforgalmi jelentősége jól ismert, addig a vízinövényekkel borított vízterekben (sekély tavaink jelentős része, nagy tavak parti régiói) a bakterioplanktonról csak igen kevés információval rendelkezünk. Éppen ezért a teljes tavi anyagforgalom szempontjából nem elhanyagolható a bakterioplankton szerepének megismerése a vízinövényzettel borított területeken. A tervezett kutatás célja a bakterioplankton mennyiségének, dinamikájának, anyagforgalmi szerepének, funkcionális és taxonómiai diverzitásának megismerése különböző növényzetű sekély tavakban.

Freshwater and marine studies have both demonstrated that a significant portion (~50%) of carbon fixed by phytoplankton is passed through the so-called microbial loop, in which the dissolved organic carbon (DOC) is assimilated by heterotrophic bacteria and channelled to heterotrophic/mixotrophic protists and higher trophic levels. In contrast with the remarkable ecological significance of the heterotrophic bacterioplankton, there is a considerable lack of information on its detailed function in different aquatic habitats. This gap of knowledge is especially apparent in macrophyte-dominated littoral systems, where a considerable amount of DOC originates from aquatic macrophytes and the importance of the bacterioplankton is largely unknown. With this end in view, the primary aim of the project is to elucidate the role of the heterotrophic bacterioplankton in macrophyte-covered shallow lakes. Our aim is to determine how the variation in DOC source (emergent- submergent and floating-leaved macrophytes) and composition (different DOC forms) influence bacterial abundance, production and metabolism in littoral aquatic systems. We want to assess how heterotrophic bacterial production relates to total primary production (phytoplankton + macrophytes) in littoral systems. We intend to determine how different DOC sources and composition of DOC influence the composition of functional groups of bacterioplankton. We also intend to determine whether DOC content and composition has a significant influence on bacterial diversity.

What is the major research question?

Dissolved organic carbon (DOC) is a key regulator of aquatic ecosystems, and the primary substrate for numerous aquatic bacteria. However, in littoral ecosystems, where non-planktonic primary producers, such as aquatic macrophytes are substantial, the role of bacterioplankton is gravely under-researched. Based on literature data, the increase of macrophyte dominance does not only mean increasing DOC supply, but also causes changes in the composition and biological availability of the DOC pool depending on the type of macrophytes and various environmental factors. The increase in macrophyte dominance, therefore, can not only affect bacterial activity and abundance, but also inevitably modifies the diversity of the bacterioplankton. Moreover, the structuring effect of aquatic macrophytes cannot be neglected either, which creates a favourable environment for the growth of photoheterotrophic bacteria through modifying underwater light conditions. Considering the above, the main research question of the project is how the source and composition of DOC (originating from emergent, submergent and floating-leaved macrophytes) influence the role and diversity (taxonomic, functional and metabolic composition) of the bacterioplankton in littoral systems. Our further aim is to understand the effect of selective light absorption by aquatic marcophytes on bacterioplankton structure (photoautotrophic/photoheterotrophic/ heterotrophic life forms).

What is the significance of the research?

Incorporating the littoral zone into our understanding of whole-lake metabolism is complicated by the variety of littoral metabolism sources. Therefore the proposed sampling and experimental work will significantly contribute to understanding the complex web of relationships in the carbon cycling of littoral systems, its microbiological aspects in particular, and it will also reveal the dependence of these relationships on macrophyte decomposition and various other biotic and abiotic factors. The results of the project will shed light on the importance of heterotrophic bacterioplankton production and clear up the current contradictory views on how it is linked to the primary production and the DOC pool of the littoral zone. The availability of decomposing organic matter to bacteria is determined by its chemical nature, which highly depends on what types and/or species represent the macrophyte community. The expected results would also contribute to understanding the significance of the metabolic processes in the littoral zone of larger lakes, and provide invaluable information for the modelling of aquatic ecosystems.

Despite their small size, bacteria floating in the water (bacterioplankton) are a major component of the aquatic food web, and play a fundamental role in the biogeochemical carbon cycle. Elucidating the relationship between bacteria and the environmental factors regulating their activity is crucial in understanding the functioning of aquatic ecosystems. Bacterioplankton is fundamentally influenced by the quantity and composition of organic substances dissolved in the water. These substances, which are utilized by bacteria during their metabolism, can originate from algae floating in the water (phytoplankton) as well as aquatic macrophytes or external sources (e.g. sewage discharge). While in waters dominated by algae as major primary producers (deep lakes and oceans) the significance of bacterioplankton in nutrient cycling is well-known, in the case of macrophyte-dominated waters (reed, etc.; majority of shallow lakes, littoral shore zone of large lakes) there is a considerable lack of information in this respect. The role of the bacterioplankton in the macrophyte-covered littoral zone, therefore, cannot be left out of account with respect to the whole-lake carbon metabolism. Considering the above, the aim of the planned research is to determine the abundance and dynamics of the bacterioplankton, its role in carbon cycling, and its functional and taxonomic diversity in macrophyte-covered shallow lakes.
