Az MTA SZBK kutatói klorofill fluoreszcencia tranziensek vizsgálatával bizonyítékot találtak arra, hogy a második fotokémiai rendszer (PSII) konformációs állapotai különböznek a sötétadaptált, egyetlen telítési flash-sel gerjesztett és fényadaptált állapotaikban. A konformációváltozások szerkezeti háttere azonban nem ismert. Mindezek tisztázására szisztematikus spektroszkópiai vizsgálatokra (klorofill fluoreszcencia, cirkuláris dikroizmus, Fourier transzfrom infravörös spektroszkópia) van szükség PSII mikrokristályokon, ’core’ komplexeken, PSII membránokon valamint vad típusú és pigment- és lipid-mutáns cianobakteriális sejteken és membrán preparátumokon. A mikrokristályokon szabadelektron lézeres Röntgen (X-FEL) és neutron diffrakciós vizsgálatokat is tervezünk.
Kínai és japán kutatók nemrég X-FEL technika alkalmazásával meghatározták a PSII atomi felbontású szerkezetét, amely mentes a Röntgen-sugarak okozta károsodásoktól. A fehérje krisztallográfia nélkülözhetetlen a biológiai rendszerek funkcióinak megértésében. A PSII működése közben azonban a szerkezet, így az optikai tulajdonságok és a gerjesztési dinamika is konformációs változásokra lehet képes. Ezért a projekt során megteremtjük a kísérleti feltételeket a szerkezetváltozások detektálásához mikrokristályokon X-FEL és neutron diffrakció segítségével. A különböző mintákon végzett spektroszkópiai vizsgálatok összefogó képet fognak adni a PSII-ről szerzett szerkezeti ismereteinkről, ill. a fényindukált konformációs változások részleteiről. A feltételezett funkcionális változások jellemzésére, ill. a fény- és sötétadaptált reakciócentrum állapotok karakterizálására fluoreszcencia élettartam méréseket tervezünk.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Fotoszintetikus szervezetekben a fényenergia befogása és elektrokémiai szabad energiává alakítása a fotokémiai reakciócentrum (RC) fehérje-komplexekben történik, amelyek redox komponenseket és pigment molekulákat tartalmaznak, és meghatározó szerepük van az elektrontranszfer szabályozásában. Az elsődleges töltéselválasztás hatására a PSII-ben erős elektromos tér alakul ki, ami képes lehet a RC-mal kölcsönhatásba lépni és lassú, hőmérsékletfüggő konformációs változásokat elindítani és stabilizálni a RC fényadaptált állapotában. Ezek a konformációs átmenetek segíthetik a RC-on belüli másodlagos elektrontranszfer lépéseket, melyek így jelentős hatással lehetnek a fotoszintézis reakcióinak teljes energetikájára és dinamikájára. A második fotokémiai rendszer reakciócentrumának konformációváltozásaira hőmérsékletfüggő fluoreszcencia-hozam mérések közvetett bizonyítékot szolgáltattak, azonban a szerkezetváltozások természete és fiziológiai jelentősége nem tisztázott. Vizsgálataink ezen kérdések megválaszolására irányulnak.
Munkánk során PSII mikrokristályokon, ’core’ komplexeken, PSII membránokon, valamint vad típusú és pigment- és lipid-mutáns cianobakteriális membrán preparátumokon végzünk spektroszkópiai (klorofill fluoreszcencia, cirkuláris dikroizmus, Fourier transzform infravörös spektroszkópia) méréseket, és - mikrokristályokon - szabadelektron lézeres Röntgen szerkezetvizsgálatokat és neutron diffrakciós méréseket tervezünk. A feltételezett funkcionális változások jellemzésére, ill. a fényadaptált és sötétadaptált RC állapotok karakterizálására fluoreszcencia élettartam méréseket tervezünk.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

Az utóbbi évtizedek intenzív kutatásai lehetővé tették a fotoszintetikus fehérje komplexek nagyfelbontású szerkezeti meghatározását, amely fontos lépést jelentett a fotoszintézis molekuláris mechanizmusának felderítésében. A komplexek szerkezetváltozásainak természetéről és fiziológiai jelentőségéről azonban igen hiányosak az ismereteink. Vizsgálataink ezen probléma sarkalatos kérdéseit célozzák. A feladat lényege, hogy PSII mikrokristályokon, core komplexeken és cianobakteriális membránokon azonosítsuk a konformációváltozásokat. A különböző mintákon végzett vizsgálatok képet fognak adni a PSII fényindukált konformációváltozásainak természetének részleteiről. A konformációs változások és az elektrontranszfer utak megismerésével pontosabb képet kaphatunk a növények szoláris energia hasznosításáról, amely akár az alternatív energiaforrások kutatásában is hasznos alapot képezhet.
Ezen kívül célunk olyan kísérleti körülmények kidolgozása, amelyek kompatibilisek a későbbiekben tervezett szerkezetvizsgáló technikákkal. Ennek fő célja, hogy előkészületeket tegyünk fejlett szerkezetvizsgáló módszerek alkalmazására. A kutatás kutatóközpontban zajlik, tehát lehetőség nyílik egyetemi hallgatók (MSc, PhD) bevonására is.
Az SZBK laboratóriumainak infrastruktúrája szilárd alapot nyújt a tervezett kutatáshoz, rendelkezik a biológiai minták preparálásához és szerkezetének és funkcióinak jellemzéséhez szükséges feltételekkel. A Fotoszintetikus Membrán Csoport laboratóriumai teljesen felszereltek a minták előállításához és jellemzéséhez (preparatív eszközök, spektroszkópiai, termolumineszcencia és fluoreszcencia tranziens mérések), valamint rendelkezik egy a cirkuláris (lineáris, anizotróp cirkuláris) dikroizmus méréséhez szükséges továbbfejlesztett spektropolariméterrel, amely az adott feltéttel egyedi felszereltségű berendezés.
A kutatás fontos része az SZBK-ban nemrégiben Dr. Brockhauser Sándor vezetésével megalapított Röntgen-krisztallográfia Laboratórium, ahol a sérülékenységük miatt nehezen szállítható mikrokristályok előállíthatóak és jellemezhetőek. Brockhauser Sándor közreműködésére a neutron-diffrakciós kísérletekben is számíthatunk. Jian –Ren Shen X-FEL kísérleteihez a mérési protokollt szolgáltatjuk.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

A projekt alapvető célkitűzése új ismeretek szerzése az oxigénfejlesztésért felelős fotokémiai rendszer, a PSII fotokémiai reakciócentrumának szerkezetéről és funkcióiról, ill. különböző – korábban részleteiben nem azonosított – szerkezeti és funkcionális állapotairól. A fényenergia befogása és elektrokémiai szabadenergiává alakítása a reakciócentrum fehérje-komplexen belül történik. A fotoszintézis fényreakciói nagymértékben függnek a fotoszintetikus egység szerkezetétől; ezek az egységek nagyfokú változatosságot mutatnak és gyorsan reagálnak a környezeti tényezők változásaira. Vannak bizonyítékok, melyek azt mutatják, hogy a PSII konformációs állapotai különböznek a sötét- és fényadaptált állapotaikban – ezen konformációs változások szerkezeti háttere azonban nem ismert. Ennek tisztázása érdekében szisztematikus spektroszkópiai vizsgálatokra (klorofill fluoreszcencia, cirkuláris dikroizmus, Fourier transzform infravörös spektroszkópia) van szükség különböző PSII mintákon és preparátumokon.
A proteinek kristályos állapotukban megtartják funkcionális szerkezetüket, gerjesztésüket követően azonban a szerkezet konformációs változásokra lehet képes. Ezért munkánk során mérési protokollt dolgozunk ki szabadelektron lézeres Röntgen szerkezetvizsgálatokhoz és neutron diffrakciós mérésekhez mikrokristályokon – kristálytani módszerekkel is azonosítandó a konformációváltozások természetét. A különböző mutáns membránokon végzett spektroszkópiai vizsgálatok összefogó képet fognak adni a konformációs változások fiziológiai jelentőségéről.

By investigating chlorophyll fluorescence transients, coworkers at BRC have found evidence that the conformational states of Photosystem II (PSII) vary in dark-adapted states, after excitation by a single-turnover saturating flash and upon multiple-turnover excitation. However, the structural background of these conformational changes is not known. In order to clarify these, systematic spectroscopic investigations (chlorophyll fluorescence, circular dichroism, FTIR) are needed on PSII microcrystals, ’core’ complexes, PSII membranes and wild-type, pigment- and lipid-mutant cyanobacterial cells and membrane preparations; on microcrystals X-ray free-electron laser (X-FEL) and neutron diffraction experiments will also be prepared.
Recently, Chinese and Japanese researchers have determined the atomic-resolution crystal structure of PSII by using X-FEL, which avoids damages caused by X-rays. Protein crystallography is crucial in understanding the functions of biological systems. After excitation, however, the structure, thus the optical features and the excitation dynamics might undergo conformational changes. During our work we will elaborate measuring protocols for detecting the light-induced PSII reaction center (RC) conformational changes by X-FEL and neutron diffraction experiments. The spectroscopic parameters measured on different samples will provide a detailed overview about the different structural and functional states of PSII; in particular on the dark- and light-adapted RC conformational states. We are planning fluorescence lifetime measurements to characterize the hypothetical functional changes and the dark- and light-adapted RC states.

What is the major research question?

In photosynthetic organisms, the capture of light energy and its transformation into electrochemical free energy occurs within the RC complex. The protein matrix containing the redox components and photosynthetic pigments can play a crucial role in the regulation of the electron transport. The strong electric field in Photosystem II generated by the primary charge separation can interact with the RC proteins initiating a slow, temperature-dependent conformational change to the light-adapted state. These conformational transitions promote the secondary electron transfer steps within the RC complex, thus they can have considerable effect on the energetics and dynamics of the reactions of photosynthesis. Temperature-dependent fluorescence yield measurements provided indirect evidence on conformational changes in PSII reaction center. However, the nature and the physiological significance of the structural changes have not been clarified. The aim of our investigations is to answer these questions.
During our work we will perform spectroscopic investigations (chlorophyll fluorescence, circular dichroism and Fourier transform infrared spectroscopy) on PSII microcrystals, ’core’ complexes, PSII membranes and wild-type, pigment- and lipid-mutant cyanobacterial cells and membrane preparations; on microcrystals X-ray free-electron laser and neutron diffraction experiments will also be prepared. We are planning fluorescence lifetime measurements to characterize the hypothetical functional changes and the dark- and light-adapted reaction center states.

What is the significance of the research?

The intensive researches of last decades enabled the determination of photosynthetic protein complexes’ high resolution structures, which was an important step in exploring the molecular mechanisms of photosynthesis. However, our knowledge about the nature and physiological significance of the structural changes of these complexes is still rudimentary. The essence of the task is the identification of conformational changes of PSII microcrystals, ’core’ complexes, PSII membranes and cyanobacterial cells and membrane preparations. The measurements performed on different samples will provide an overview about the details of the nature of light-induced conformational changes of PSII. By the exploration of the conformational changes and electron transfer pathways we can gain a more detailed picture about the solar energy utilization of plants, which can also be beneficial in alternative energy source researches.
Another aim of our work is to develop such experimental conditions which are compatible with the planned structure-investigating techniques. The main goal is to be prepared to apply advanced structure-investigating methods. The research will be completed at the academic research centre, thus there are many possibilities of the involvement of university students (MSc, PhD).
The infrastructure of the laboratories in Biological Research Centre provides a solid basis for the planned research progress. The Laboratory of Photosynthetic Membranes is equipped for biological sample preparations and characterization of the structure and functions of the prepared samples (spectroscopic instruments, thermoluminescence, fluorescence transient measurements). The Laboratory also possesses an advanced spectropolarimeter for circular dichroism (also linear- and anisotropic circular dichroism) measurements which makes it unique with the given condition.
Important part of the research is the recently established X-ray Crystallography Laboratory in BRC by Dr. Brockhauser Sándor, where the sensitive microcrystals can be prepared and investigated in place, and we can count on his cooperation with the neutron diffraction measurements. We will also elaborate the measuring protocol for Jain-Ren Shen’s X-FEL investigations.

The fundamental aim of our work is gaining new information about the structure and function and different – which have not yet been identified – structural and functional states of Photosystem II (PSII) and its photochemical reaction center (RC). The capture of light energy and its transformation into electrochemical free energy occurs within the RC complex. The light reactions of photosynthesis are greatly dependent on the structure of the photosynthetic unit, which shows high diversity and has a quick response to the change of the environmental factors. Evidences indicate that the conformational states of PSII vary in dark- and light-adapted states, however the structural background of these conformational changes is not known. In order to clarify these, systematic spectroscopic investigations (chlorophyll fluorescence, circular dichroism and Fourier transform infrared spectroscopy) of PSII samples and preparations are needed.
In most cases proteins retain their functional structure in crystal state. After excitation, however, the structure might undergo conformational changes. Hence, during our work we will elaborate measuring protocol(s) for detecting the light-induced PSII reaction center conformational changes in microcrystals by X-ray free-electron laser and neutron diffraction experiments. The spectroscopic parameters measured on different mutant membranes will provide an overview about the physiological significance of the conformational changes.

A második fotokémiai rendszer (PSII) a fotoszintetikus apparátus azon komponense, amely képes a fényenergia abszorpciójára, gerjesztett állapotában redoxpotenciálja révén a víz oxidálására, valamint transzmembrán elektrokémiai potenciál grádiens létrehozására. Munkánk megkezdésekor a PSII funkcionális változásait tükröző tranziens, a változó klorofill-a fluoreszcencia (Fv), értelmezése ellentmondásokkal terhelt volt. Korábban atomi-felbontású krisztallográfia segítségével a PSII akceptor oldala és vízbontó komplexe körül konformációváltozásokat azonosítottak; felmerült a kérdés, hogy hasonló szerkezetváltozások fellépnek-e az Fv során. A pályázat során felfedtük, hogy a korábbi – széles körben elfogadott – modell-értelmezéssel ellentétben az Fv nem a stabil töltésszétválasztásból ered; továbbá, az Fv/Fm paraméter nem azonos a PSII kvantumhatásfokával. Ezen kívül rávilágítottunk, hogy a PSII egy korábban nem azonosított, ún. fény-adaptált töltés-szétválasztott állapotot is felvesz az eddig ismert nyitott és zárt állapotokon kívül. Egy másik fontos megállapításunk, hogy ez az állapot csak lépcsőzetesen érhető el, kellően hosszú várakozási időkkel az egyedi gerjesztések között. Kimutattuk azt is, hogy ezek a várakozási idők erősen függenek a hőmérséklettől és a PSII natív lipid környezetétől. Feltevésünk szerint ezeket a folyamatokat a PSII reakciócentrumában keletkező tranziens és stacionárius lokális elektromos terek és dielektromos relaxációs folyamatok határozzák meg.

Photosystem II (PSII), embedded in the thylakoid membranes of oxygenic photosynthetic organisms, catalyses the light-induced water oxidation and generates a transmembrane electrochemical gradient. At the start of our project, the model describing the origin of variable chlorophyll-a fluorescence (Fv), reflecting the functional activity of PSII, was controversial. Atomic resolution crystallography experiments have revealed conformational changes accompanying the reactions around the acceptor side and water splitting complex of PSII. This raised the question if Fv contains any component arising from similar structural changes. During the project we have shown that, in contrast to the earlier – widely accepted – model, Fv does not originate directly from the stable charge separation event, and the Fv/Fm parameter cannot be equated with the quantum efficiency of PSII. Further, we identified the previously unknown charge-separated light-adapted state of PSII, in addition to its open and closed states. Another important observation made by us is that this state can be generated only in a stepwise manner, with sufficiently long waiting times between the excitations. We have also shown that these waiting times depend largely on the temperature and on the native lipid environment of PSII. According to our interpretation, these processes are determined by the stationary and transient local electric fields and dielectric relaxation processes occurring in the reaction center of PSII.
