A kutatás összefoglalója, célkitűzései szakemberek számára
Itt írja le a kutatás fő célkitűzéseit a témában jártas szakember számára.

A kemoterápiás kezelések során alkalmazott gyógyszerek okozta súlyos mellékhatások és a fellépő rezisztencia olyan új fémtartalmú vegyületek tervezését és fejlesztését motiválják, melyek jó hatékonyságuk mellett kellő szelektivitást és így alacsony szisztémás toxicitást mutatnak. A hatóanyagok racionális fejlesztési és optimalizálási folyamata erősen igényli, hogy egy rákellenes fémkomplex oldatbeli viselkedésértől és farmakokinetikai tulajdonságairól minél többet tudjunk. Ebben a pályázatban új fémkomplexek előállítása és széleskörű jellemzése mellett összefüggéseket keressünk a meghatározott termodinamikai, kinetikai paraméterek, fizikai-kémiai tulajdonságok, redoxi aktivitás, fehérjékhez és DNS-hez való kötődési képességük, a kristályszerkezeti, valamint a biológiai aktivitási adatok között. A kapott eredmények jelentős mértékben hozzájárul(hat)nak a bioaktivitás optimalizálásához, mivel fontos kapcsolatot teremtenek a szilárd fázisú karakterizálás és az in vitro, in vivo aktivitás mérések között. A pályázatban különböző organoruténium-, organoródium-komplexek, és tioszemikarbazonok fémkomplexeinek tanulmányozása mellett olyan vegyületeket is bevonunk a vizsgálatainkba, melyek multidrog-rezisztencia szelektivitást mutatnak és jelentős a fémion-kötő képességük. Az eredmények segíteni fogják ezen fémkomplexek és nemfémes rákellenes vegyületek biofluidumokban való viselkedésének, a farmakokinetikai tulajdonságaik, a hatásmechanizmusuknak és a mellékhatások kialakulásához vezető kémiai folyamatoknak a megértését.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Új rákellenes szerek fejlesztésére még mindig szükség van, mert számos ráktípus mutat rezisztenciát a jelenleg rendelkezésre álló gyógyszerekkel, köztük a Pt-tartalmúak, szemben. A hatékonyabb fémtartalmú gyógyszerek kifejlesztéséhez fontos, hogy jobban megértsük a farmakokinetikai viselkedésüket és a hatásmechanizmusukat. Azonban a fémkomplexek a vizes fázisban való oldódásuk és biomolekulákkal való kölcsönhatásuk révén sokkal nagyobb mértékben változtathatják meg eredeti formájukat, mint a hagyományos gyógyszerek. Ez is az oka annak, hogy legtöbbször nincs elég információ rákellenes komplexek biospeciációjára vonatkozóan. Ebben a pályázatban célunk új fél-szendvics fémorganikus (Ru, Rh), és tioszemikarbazonok komplexeinek előállítása és jellemzése. Vizsgálni fogjuk oldatbeli stabilitásukat, fehérjékkel való kölcsönhatásukat, melyeknek hatása van a farmakokinetikai tulajdonságokra, a mellékhatásokra és az aktivitásukra. Az egyensúlyi folyamatokat, a fehérjékhez való kötődést különböző technikák, mint pl. pH-potenciometriás, spektroszkópiai (UV-látható, fluorimetria, NMR, EPR) és elválasztási (ultraszűrés, CZE) módszerek kombinált használatával fogjuk tanulmányozni. A komplexek antiproliferatív aktivitását is meg fogjuk határozni. Az eredmények részletes elemzésével fel szeretnénk tárni, hogy milyen fizikai-kémiai és farmakológiai tulajdonságoknak van szerepe a biológiai hatékonyság kialakításában. Számos multidrog-rezisztencia szelektivitást mutató vegyületben (tioszemikarbazonok, 8-quinolinolok) található kelátképző csoport. Szeretnénk feltárni, hogy milyen szerepet játszhat a hatásmechanizmusukban a szervezetben megtalálható fémionokkal való komplexképzésük.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

A fémtartalmú daganatellenes vegyületek fejlesztése fontos kutatási irányvonalat jelent, melynek célja a mellékhatások csökkentése és a jobb gyógyászati tulajdonságok elérése. Ugyanakkor a fémkomplexek klinikai vizsgálatokba csak ritkán és lassan jutnak el, mely gyakran a hatásmechanizmusra és az oldatbeli viselkedésre vonatkozó információ hiánya miatt van. Általában a biológiailag aktív fémkomplexeket részletesen jellemzik szilárd vagy oldatfázisban, de az egyensúlyi folyamatok, a stabilitás és a biofluidumok komponenseivel való kölcsönhatások tanulmányozására gyakran nem kerül sor. A rákellenes fémkomplexek biospeciációs jellemzése meglehetősen hiányos a szakirodalomban, és az a fajta kombinációja a különböző műszeres módszereknek és adatfeldolgozásnak, amit a pályázatban szeretnénk alkalmazni igen ritka. Számos multidrog-rezisztencia szelektivitást mutató vegyület fémion-kötő tulajdonságának szerepe a hatásmechanizmusukban nem tisztázott, és pályázatunkban ezt is vizsgálni fogjuk. A várható eredmények és a megelőző vizsgálataink során kapottak alapján szeretnénk egy olyan adatbázist felépíteni, mely egymással összehasonlítható paramétereket fog össze, mint pl., a különböző fizikai-kémiai tulajdonságok, oldatbeli stabilitás, fehérjék, feltételezett targetmolekulák és modelljeik felé mutatott affinitási adatok és antiproliferatív aktivitás. Ezen adatok korrelációs elemzése hozzájárul a biológiai hatékonyságban és mellékhatásokban megfigyelhető különbségek jobb megértéséhez. Így munkánk támogatni fogja a hatékonyabb gyógyszerfejlesztései folyamatot a preklinikai fázisban. Kutatóhelyeinken kiváló infrastruktúra biztosított ahhoz, hogy végrehajtsuk a tervezett vizsgálatinkat, a pályázat résztvevői és a nemzetközi partnerek is elismert szakemberek, erős publikálási aktivitással.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

A rákos betegségek világszerte a vezető halálok egyike; a rosszindulatú daganatok okozta elhalálozás folyamatosan emelkedik, és 2024-ben várhatóan 19 millió embert fog érinteni. Így az új daganatellenes szerek fejlesztése még mindig nagyon fontos feladat, mely erősen ösztönzi olyan vegyületek előállítását, melyek hatékonyak, de sokkal jobban tolerálható mellékhatásokat okoznak, mint a gyógyászatban jelenleg használt hatóanyagok. Nagyszámú, különböző szerkezetű rákellenes fémkomplexek előállítását írták már le, de az előrelépéshez minél több információra van szükség ezen vegyületek emberi szervezetben való viselkedéséről, mely azonban meglehetősen limitált az irodalomban. Ebben a pályázatban új fémtartalmú vegyületek előállítását tervezzük és részletesen fogjuk jellemezni az oldatkémiai, szerkezeti, redoxi tulajdonságaikat, valamint a vérben és sejten belül megtalálható egyes fehérjékkel, DNS-sel való kölcsönhatásukat és rákellenes hatásukat humán rákos sejteken is vizsgálni fogjuk. Megpróbáljuk megérteni egyes multidrog-rezisztencia szelektivitást mutató vegyületek létfontosságú fémionokkal való kölcsönhatásának szerepét a hatásmechanizmusukban. Eredményeink és az egymással összehasonlítható adatokból felépített adatbázis elemzése hozzájárul, ahhoz, hogy jobban megértsük a vizsgált rákellenes vegyületek viselkedésének különbségeit és hasonlóságait, mely segíti a gyógyszerek fejlesztését, és támogatja a preklinikai és klinikai vizsgálatokat.

The serious side effects and resistance of anticancer drugs used in chemotherapy have motivated the design and development of novel metal-based compounds that combine good efficacy, selectivity and low systemic toxicity. The rational drug development and optimization process strongly require information about the solution behavior, pharmacokinetics properties of the metallodrugs. In this proposal in addition to the preparation and characterization of the various features of selected metal-based compounds using a broad spectrum of instrumentation, we will search for intrinsic multi-component based correlations between the determined thermodynamic/kinetic parameters, physico-chemical properties, redox activity, protein and DNA binding ability, crystallographic data and the biological effectiveness. The gained knowledge strongly contributes to the optimization of the bioactivity as it provides an important link between the solid phase characterization and the in vitro/in vivo activity measurements. The project is focused on various organoruthenium, organorhodium complexes, and various metal complexes of thiosemicarbazones in addition to compounds possessing multidrug resistance-selective antiproliferative activity and significant affinity towards essential metal ions. The results help in elucidation of the fate of these metal complexes and non-metallic anticancer compounds in the biofluids in order to understand the pharmacokinetic properties, the mechanism of actions and the (bio)chemical reactions which are responsible for the unwanted side effects.

What is the major research question?

Novel alternative metal-based anticancer compounds are still desperately sought due to the fact that many high-grade cancers show resistance to most of the current available treatments, including Pt drugs. For the more efficient drug development we have to understand better the pharmacokinetic profile, the mechanism of action. However, these processes are more complicated compared to the conventional drugs, since the metal complexes may undergo alterations via dissolution in water, interaction with biomolecules. This is the reason why only limited information is available about the biospeciation of antitumor metallodrugs in most cases. In this project we aim to develop and characterize novel half-sandwich organometallic (Ru, Rh) complexes and various metal complexes of thiosemicarbazones concerning their solution speciation, interaction with proteins, which can play crucial role in their pharmacokinetic properties, side effects or their final effect. These properties, equilibrium processes and protein binding sites and strength will be studied by the combination of various techniques such as pH-potentiometry, spectroscopic (UV-vis, fluorometry, NMR, EPR) and separation (ultrafiltration, CZE) methods. The antiproliferative activity of the complexes will be also measured. Based on the analysis of the results we aim to reveal what kinds of physico-chemical and pharmacological properties are crucial for the biological effectiveness. Many multidrug resistance-selective drugs (thiosemicarbazones, 8-quinolinols) possess chelating groups, and the possible role of their complexation with essential metal ions in the mechanism of action will be also addressed in the project.

Development of metal-based antitumor compounds aiming reduced side effects and better pharmaceutical properties is in the main stream of current endeavours. However, the unsatisfactory rate of the translation of metallodrugs to clinical trials often roots on the missing adequate information on solution speciation and mechanism of action. In the general practice, biologically active complexes are meticulously characterized in the solid or solution states, but study of their solution equilibria, stability and interactions with endogenous components of biofluids in aqueous systems is often neglected. The biospeciation analyses of the antitumor metal complexes are considerably incomplete in the literature and the kind of combination of various instrumental methods with data processing, what we offer here, is unique in the research field. As well as the role of the metal ion binding properties of a series of multidrug resistance-selective compounds in their mechanism of action is unclear and will be investigated. The results obtained from the studies conducted within the project in addition to our previous ones are aimed to built up a database containing comparable parameters on various physico-chemical properties, solution stability, affinities towards proteins, putative targets (such as DNA), their model compounds and antiproliferative activity. Analysis of the multi-component based correlations between these data will contribute to a better understanding of the differences in biological effectiveness, side effects; therefore our work will support a more efficient drug selection process in the advanced preclinical phase. To perform this multifaceted investigation the top notch infrastructure is provided at the host research units and the members of the research team and the international partners are recognized scientists with strong publication records.

Cancer is a leading cause of death globally, and mortality due to malignant tumours is projected to rise to over 19 million people in 2024. Therefore alternative antitumor drugs are still desperately sought, which inspires the research passion for the discovery of novel compounds with improved effectiveness and more tolerable side effects compared to the current drugs used in medication. A large number of metal complexes with various structures have been developed and reported as antitumor agents, but the pharmaceutical success strongly requires information about the fate of these metallodrugs in the human body, which is rather limited in the literature. In the project we will develop and study various metal-containing compounds regarding their solution chemical, structural, redox properties, their binding affinities towards selected blood and cellular proteins, DNA in addition to their anticancer properties screened in human cancer cells. We will attempt to describe the role of essential metal ions in mechanism of action of selected compounds showing significant preference for targeting multidrug resistant cancer cells. Our results and the analysis of the collection of large number of comparative data will contribute to understanding of the differences and similarities of these anticancer compounds and help the drug development process, support preclinical and clinical studies.

A hatásosabb, kedvezőbb mellékhatás-profilú és jobb gyógyszerkémiai tulajdonságú rákellenes gyógyszerhatóanyagok kifejlesztése fontos kutatási irányvonalat jelent. Ugyanakkor a fémtartalmú vegyületek klinikai vizsgálatokba lassan jutnak el, melynek gyakori oka a hatásmechanizmusra és az oldatbeli viselkedésre vonatkozó korlátozott információ. Ebben a pályázatban nemcsak sikeresen állítottunk elő különböző típusú rákellenes félszendvics fémorganikus-, tioszemikarbazon- és kinolinol-alapú fémkomplexeket, melyek között volt jó szelektivitást mutató és multidrog rezisztens rákos sejteken is aktív vegyület, hanem számos összefüggésre mutattunk rá a jellemzett fizikai-kémiai, oldategyensúlyi, biospeciációs jellemzők és a rákellenes hatás között. A termodinamikai, kinetikai, szilárd- és oldatfázisra vonatkozó szerkezeti és biológiai adatok korrelációs elemzése hozzájárult az érzékeny és a rezisztens rákos sejtek felé mutatott citotoxicitásban megfigyelt különbségek jobb megértéséhez. Ezen ismeretek egyértelműen támogatják a hatékonyabb gyógyszerfejlesztései folyamatot a preklinikai fázisban. Az öt éves pályázat 43 tudományos közleménnyel zárult, melyek összesített hatástényezője 186.

Development of anticancer drugs aiming reduced side effects and better pharmaceutical properties is in the main stream of current endeavours; however, the unsatisfactory rate of the translation of metal-containing compounds to clinical trials often roots on the limited information on their mechanism of action and behavior in solution. In this project, we have not only successfully developed various types of anticancer half-sandwich organometallic, thiosemicarbazone and quinolinol-based metal complexes, among which there were representatives with good selectivity and activity on multidrug resistant cancer cells, but we also could point out correlations between the characterized physico-chemical, solution equilibrium and biospeciation properties and the anticancer effect. Our correlation analysis of the thermodynamic, kinetic, solid- and solution-phase structural and biological data contributed to a better understanding of the observed differences in cytotoxicity shown towards sensitive and resistant cancer cells. This knowledge clearly supports a more efficient drug development process in the preclinical phase. The five-year project ended with 43 scientific publications, with a summed impact factor of 186.
