Itt írja le a kutatás fő célkitűzéseit a témában jártas szakember számára.

A növényi mikrobiom, valamint a növényi mikrobiom metabolomra, ill. a növényre való kapcsolata egyre intenzívebb kutatási terület, a fenntartható mezőgazdaság egyik alapjának tartott megközelítés. Jelen pályázat egy jelentős regionális termék, a torma poszt-harveszt viselkedésének és elicitációjának, valamint mikrobiom és metabolom szintű variabilitásának vizsgálatát tűzi ki célul.
A kérdések megválaszolásához klasszikus (mikrobiális izolátumok létrehozása, vegyületek megtisztítása) és a legmodernebb módszertan (metabolomika, adatbányászat) elemeit is fel kívánjuk használni.
A projektben definiálni tervezzük a torma tipikus mikrobiomját, valamint részlegesen az e fajokban előforduló természetes vegyületeket. Bizonyítani kívánjuk, hogy a torma endofitonok valamely módon alkalmazkodtak a tormában jelen lévő kémiai komponensekhez. Meg kívánjuk vizsgálni a hozzáférhető termesztett változatok közötti metabolom szintű különbséget. Továbbá, a tormagyökér poszt-harveszt változásainak vizsgálatát tervezzük, a gyökér viselkedését elicitációs modellekbe bevonva. Az elicitáció során stressz hatások (többek között endofiton inokuláció) segítségével kíséreljük meg a bioaktív komponensek koncentrációjának növelését. A főbb jelenségeket főbb termesztett fajtákra lebontva is vizsgáljuk.
A projekt nem csak értékes alapkutatási eredményeket hozhat. A mikrobiom-metabolom korrelációk vagy a poszt-harveszt eredmények termesztés-, ill. tárolástechnológiába való átültethetőségét is valószínűsítjük.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

A munka fő hipotézisei a következőek.
1. A tormagyökér mikrobiomjának definiálása. A kutatás során klasszikus módszerekkel igyekszünk a tormában élő endofiton flóra (mindenekelőtt fonalas gombák) variabilitását fajszinten definiálni, ezen felül egyes érdekes fajok kémiai-funkcionális karakterizálását is tervezzük (bioaktív hatóanyagaik izolációja, mirozinázok karakterizálása).
2. Kimutatható-e az endofitonok növényi metabolomhoz való adaptációja? Egyrészt a növényi metabolitok endofitonokra kifejtett hatását vizsgáljuk más növényből származó endofitonokat vagy egyéb fonalas gombákat kontrollként felhasználva, másrészt a növényi metabolom, ill. a növényben előforduló vegyületek biotranszformációs termékeit keressük.
3. Van-e szignifikáns kémiai variancia a torma változatok között? A széles körben elterjedt, termesztésbe vett fajták metabolomjának különbségeit vizsgáljuk, majd adatbányászati modellekben keressük meg azokat a termesztési paramétereket, amelyek befolyásolhatják azt.
4. Milyen posztharveszt változásokat mutat a tormagyökér? A tormagyökér posztharveszt elicitációs vizsgálatokban stresszhatásokkal vagy endofiton inokulációval kíséreljük meg a fokozott bioaktív vegyület-tartalom elérését. A vizsgálatok során proteomikai és metabolomikai változásokat is követünk.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

A növényi mikrobiom – metabolom interakció, a posztharveszt elicitáció és a posztharveszt kolonizáció mind ígéretes, feltörekvő kutatási területek. Az eredmények az endofitonok posztharveszt stádiumban betöltött jelentősége, ill. alkalmazahtósága kapcsán nagy jelentőségűek, várhatóan alapvető hatással rendelkező dologról van szó.
Az adatbányászati módszerekkel immár olyan jelenségek feltárása is lehetséges, amelyre egy évtizeddel korábban még alapvetően nem volt lehetőség – konkrétan a metabolom – termesztési paraméter korrelációk vizsgálatának eredménye kapcsán várható érdeklődés a szakma részéről.
A projektet tervező csapat erősségei a komplexitás (sok megközelítés), valamint a korszerű (metabolomika, adatbányászat) és klasszikus módszertan összhangja. A projekt további erőssége, hogy kifejezetten a növényre vonatkozó szakmai tapasztalat találkozik forró kutatási témával és a tormatermesztés körüli aktuális, praktikus kérdésekkel. Igen fontos, hogy kapcsolataink révén megfelelő és stabil hozzáférésünk van a különféle torma vonalakhoz, ill. a termesztéssel, tárolással kapcsolatos know-how-hoz is.
Az eredmények hatással lehetnek a fajták további szelekciójára, szerencsés esetben azonosíthatnak olyan tényezőket is, amelyek alapvetően befolyásolják a tormagyökér piacképességét. Ezen felül, valószínűsítjük a poszt-harveszt eredmények tárolás-technológiai adaptációjának lehetőségét, amely egyrészt növeli a hozzáadott értéket, másrészt eddig kevésbé értékesnek tekintett torma sarzsok piacképességének fokozását is lehetővé teheti. Így az eredmények a régió számára hasznos alkalmazott kutatás alapjául is szolgálhatnak a későbbiekben.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

Plant microbiome and its effects on the plant metabolome or its interaction with the plant is a trending research topic, which is considered one of the main bases of sustainable agriculture. The current grant aims to study the post-harvest changes, elicitation, the variance of the microbiome and the metabolome of a regionally important product, horseradish.
The methods that are used to answer the questions include classical (isolation of microbes, and purification of natural products), as well as the most modern ones (metabolomics, data mining).
In the current project, we aim to define the typical microbiome of horseradish roots, and partially characterize the metabolites present in them. We aim to prove that horseradish endophytes are to some extent adapted to the chemical environment in the plant. We aim to assess differences between widely available horseradish varieties at the metabolome level. Also, we aim to study the post-harvest changes of horseradish roots in different elicitation models. During elicitation, the living plant is stressed (among others, by inoculation with endophytes) so as to increase the amount of bioactive constituents. Main phenomena will be examined on the available varieties separately.
The project can result in other results beyond those for basic research. We think that the results of the microbiome-metabolome correlation studies, and the post-harvest evaluations can be integrated into crop production or storage technology as well.

What is the major research question?
Describe here briefly the problem to be solved by the research, the starting hypothesis, and the questions addressed by the experiments.

The work is centered around the following hypotheses:
1., Define the horseradish root endophyte microbiome. During the work, we plan to define the variability of the horseradish microbiome at the species level (mainly filamentous fungi) by classical isolation methods. We also plan the chemical-functional characterization of some of the species (isolation and characterization of bioactive constituents, characterization of myrosinase enzymes).
2., What adaptation do endophytes show to the plant metabolome? We either examine the effects of plant constituents on the endophytes using non-plant endophytes or other fungi as controls, or, check for biotransformation products of plant metabolites by endophytes.
3., Is there a significant chemical variance among horseradish varieties? We examine the variance of the metabolome of the widely available varietites, then, try to find culture parameters that influence it.
4., What post-harvest changes can be observed in horseradish roots? In post-harvest elicitation studies we aim to increase the amount of bioactive constituents by stress or endophyte inoculation. We also aim to quantify changes in the metabolome and the proteome.

What is the significance of the research?

The plant microbiome - metabolome interaction, the post-harvest elicitation and the postharvest colonization are all promising, trending research topics. The results on the role and application of endophytes in the post-harvest stage have high significance, the effect of the endophytes is expected to be fundamental.
With the data mining techniques, there is now a possibility to find such phenomena, that was impossible a decade ago. In particular, interest is expected about the results of the metabolome – culture parameter correlation study.
The project team's main strengths are complexity (many approaches), and the harmony of high-end methods (metabolomics, data mining) and classical methods. Another strength of the project is that horseradish-specific experience meets a hot topic, and practical questions that come from actual production of the crop. It is also important, that through our partners in the industry, we have stable access to different varieties, and know-how about the production, storage of the horseradish roots.
The results can impact selection of varieties, and, in a lucky case, such parameters can be identified that fundamentally influence the market value of the horseradish roots. What is more, we also think that the approaches used in the post-harvest study can be applied to the storage technology, which increases added value, and may increase value of horseradish batches of less quality. So, the results can also serve as a basis for later applied research in the region.

The methods that are used to answer the questions include classical, as well as the most modern ones (e.g. data mining).

We aim to prove that microbes living in horseradish are somehow adapted to the chemical compounds of the plant. We aim to assess differences between widely available horseradish varieties with respect to the beneficial components. Also, we aim to study the post-harvest changes of horseradish roots by stressing the roots (among others, by inoculation with endophytes) to force it to produce higher amounts of bioactive components. Main phenomena will be checked on the available varieties separately.
The project can result in other results beyond those for basic research. We think that the results of the microbiome-metabolome correlation studies, and the post-harvest evaluations can be integrated into crop production or storage technology as well.

A projektben a tormagyökér, annak hatóanyagai és a benne élő fonalas gombák közötti interakciót vizsgáltuk. Izolálással számos gomba vonalat izoláltunk tormagyökérből és további taxonokat írtunk le molekuláris biológiai módszerekkel. Mikroszkópos, szelektív festéses módszerrel igazoltuk, hogy az általunk izolált endofiton gombák valóban képesek a tormagyökér belső szöveteinek kolonizációjára. Talajgombákat, mint kontroll csoportot alkalmazva igazoltuk, hogy a tormagyökér glükozinolát tipusú hatóanyagait az endofitonok képesek elbontani, hasznosítani, és a növény által termelt antifungális izotiocianátokkal szemben is ellenállóbbak, mint a talajgombák. Számos endofiton illékony vegyület profilját elsőként jellemeztük. Ezen felül ígéretes eredményeket értünk el a tormafajták közötti kémiai variabilitás jellemzésében, ill. a torma metabolom talajtípus-függésének vizsgálatában.

The project aimed to characterize the interaction between the horseradish bioactive constituents and the endophytic fungi living inside the plant. We successfully isolated several fungal clones from horseradish roots, and detected the presence of additional taxa by molecular biological methods. We have proven the ability of the horseradish endophytes to colonize the inner tissues of horseradish roots by selective staining and microscopy. Using soil fungi as a control group, we have proven that endophytes are efficient decomposers and users of the glucosinolates produced by the plant. We have also shown that the endophytes are more tolerant to the antifungal isothiocyanates produced by the plant, compared to the soil fungi. We published the volatile organic constituent profile of several endophytic fungi for the first time. Our experiments also resulted in promising results in defining the chemical variability of different horseradish varieties, as well as the correlation of the plant metabolome and soil type.
