A projekt fő célja olyan új típusú, aszparaginsav alapú polimer gyógyszerhordozó mátrixok előállítása, amelyek alkalmazásával bizonyos betegségtípusok esetén a jelenleginél hatékonyabb és a betegek számára kényelmesebb kezelés válik lehetővé. A projekt egyik fő iránya poliaszpartamid alapú, nanoszálas mátrixok előállítása, melynek első lépéseként széles kémiai összetétel tartományon meghatározzuk a polimerek szerkezet-tulajdonság összefüggéseit, így a szubsztituensek hatását az oldhatóságra, oldódási sebességre, az üvegesedési hőmérsékletre és a termikus stabilitásra. Ezen összefüggések ismeretében állítunk elő egy szemészeti terápiában használt, vízérzékeny hatóanyag leadására alkalmas mátrixot elektrosztatikus szálképzéssel. A projekt másik fő irányát mukoadhezív polimerek szintézise adja. Olyan tiol és diszulfid tartalmú polimereket állítunk elő többféle kémiai úton, melyek vizes oldata a nyálkahártya mucin fehérjéivel erős kölcsönhatást mutat – ezt reológiai és adhéziós mérésekkel igazoljuk –, így várhatóan kötődnek a szem nyálkahártyáján is. Az ígéretes polimerekből hatóanyag-leadási kísérletekkel választjuk ki a hatékony formulákat, melyek pontos kémiai összetételét, tárolási körülményeit, adagolási módját a projekt során dolgozzuk ki. A projekt során lehetőség van a polimerek citotoxicitás vizsgálatára, így a projektet követően a valódi potenciállal rendelkező gyógyszerformák fejlesztése indulhat el.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

A jelenleg alkalmazott gyógyszerformák esetén sok esetben csak korlátozott terápiás hatékonyság érhető el a hatóanyag gyenge biohasznosulása miatt, és bizonyos esetekben a hatóanyag bevitele nem kényelmes a beteg számára. A kutatásunk kiinduló hipotézise, hogy aszparaginsav alapú polimerek egyszerű kémiai reakciókban történő funkcionalizálásával és környezetbarát feldolgozásával olyan új gyógyszerformák állíthatóak elő, melyek két, előre meghatározott terápiás megoldás esetében a jelenleg kapható gyógyszerformáknál jobb terápiás hatékonyságot tesznek lehetővé.
Poliaszpartamid típusú származékok esetén széles összetétel tartományon meghatározzuk, hogy a polimerek oldhatósága és üvegesedési hőmérséklete hogyan befolyásolható a kémiai összetétellel. Vizsgáljuk az elektrosztatikus szálképzéssel előállított mátrixok feldolgozási körülményeinek hatását a morfológiára és az oldódási sebességre. Meghatározzuk a szintézis során alkalmazható zöld oldószerek körét. Választ keresünk a polimerek tiol tartalmának és a bioadhézió erősségének kapcsolatára, megvizsgáljuk, hogy a különböző kémiai úton bevitt tiolcsoportok és az egyéb funkciós csoportok hatással vannak-e az adhézióra. A meghatározott szerkezet-tulajdonság összefüggések alkalmazásával kiválasztjuk a későbbi gyógyászati alkalmazások szempontjából ígéretes polimereket. A kiválasztott polimerek toxicitásának alapján értékeljük a mátrixok alkalmazhatóságát szabályozott hatóanyag-leadás céljára. A projekt végén javaslatot teszünk a jövőben terápiás célra várhatóan felhasználható polimerekre és a vonatkozó feldolgozási eljárásra.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

A projekt során az aszparaginsav alapú polimerek széles körét állítjuk elő, mely lehetővé teszi, hogy ezen polimerek szerkezet-tulajdonság összefüggéseit részletesen megismerjük, és tervezhetővé tegyük meghatározott tulajdonságú poliaszpartamidok és poliaszparaginsavak szintézisét. Az elért eredmények tudományos jelentősége, hogy az aszparaginsav alapú polimerekre és a belőlük előállított mátrixokra a jelenleg korlátozott mértékben rendelkezésre álló ismereteket jelentősen bővítjük, elsősorban a kémiai szerkezet hatását határozzuk meg az oldhatóságra, az üvegesedési hőmérsékletre valamint a bioadhézióra. Különböző környezetbarát oldószerekben oldódó poliaszpartamid származékokat állítunk elő, melyekből meghatározott terápiás célra alkalmazható mátrixokat hozunk létre elektrosztatikus szálképzéssel. Az előállított hordozók a rifampicin, illetve ahhoz hasonló hatóanyagok esetén várhatóan a jelenlegi készítményekhez képest nagyobb terápiás hatékonyságot biztosítanak. Poliaszparaginsav és poliaszpartamid származékok esetében felderítjük a tiol tartalom és a polimer szerkezet mukoadhézióra gyakorolt hatását, melynek felhasználásával erős adhéziót mutató, vizes közegű hordozókat állítunk elő. Ezen ismeretek általánosíthatók, az ismeretek birtokában olyan hordozók állíthatók elő, melyek a nyálkahártyán megnövelt tartózkodási idejüknek köszönhetően elnyújtott hatóanyag-leadást, így jó hatóanyag-hasznosulást biztosíthatnak. A projektben a szem antibiotikumos kezelésére fókuszálunk. A tervezett mátrixoknak köszönhető hatékonyság-növeléssel a betegek életminősége és a terápiás komfort nagymértékben javulhat. A bejuttatott gyógyszerek jobb hasznosulása pedig mind környezeti, mind gazdasági szempontból előnyös. A gyógyszerhordozók területén a leginkább elterjedt poliakrilát alapú rendszerekkel szemben nyilvánvaló előnyünk a tervezett rendszerek várható biológiai lebonthatósága és az egyszerűbb, környezetbarát szintézisút. Az általunk vizsgált polimerek szisztematikus kémiai módosítására nincs szakirodalmi példa, így a már ismert aszparaginsav alapú polimerekből felépülő térhálós mátrixokhoz képest az általunk javasolt kutatási irány egyedülálló a tématerületen.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

A piacon jelenleg elérhető gyógyszerformák (tabletták, filmtabletták, tapaszok, injekciók stb.) alkalmazása során a hatóanyag hasznosulása sokszor korlátozott, amely a szükségesnél nagyobb dózisú hatóanyag bevitelét teheti szükségessé, ez pedig mellékhatásokhoz és a betegek komfortérzetének csökkenéséhez vezet. A gyógyszerhordozók alapanyagainak előállítása sok esetben jelentős környezetterheléssel jár, amely a hatóanyagok rossz hasznosulásával együtt felesleges környezetszennyezést és gazdasági veszteséget is okoz.
Célunk egy, a szervezettel összeférhető makromolekulából kiindulva, olyan új hordozók kifejlesztése, melyekkel a gyógyszeres terápiák hatékonysága növelhető. A projekt során két jól meghatározott gyógyszertípus esetén javaslatot teszünk a hatékonyság növelésére, jól ismert hatóanyagok felhasználásával. Egyrészt a szemészeti terápiában alkalmazott, de erősen vízérzékeny hatóanyag jó hasznosulását lehetővé tevő szilárd gyógyszerformát készítünk. Másrészt, a szem bakteriális fertőzésének kezelésére vagy megelőzésére alkalmas antibiotikumot elnyújtott módon leadó, a szem felületén megnövekedett tartózkodási idejű cseppkészítmény alapjait szeretnénk megteremteni. A célok eléréséhez egy természetes aminosav, az aszparaginsav jelenti a kiindulási anyagot, mely várhatóan biztosítja a belőle felépülő kémiai szerkezetek szervezettel való kompatibilitását és az alacsony környezetterhelést, és így valódi alternatívát jelenthet a hatóanyag-leadás területén a jelenleg elérhető megoldásokhoz képest.

Summary of the research and its aims for experts
Describe the major aims of the research for experts.

The main goal of the project is to synthesize aspartic acid based polymer matrices for drug delivery which may provide better therapeutic efficiency and improved patient compliance compared to those of currently available formulations. One of our main topics is the synthesis of nanofibrous polyaspartamide matrices and in the first step of this topic we explore the structure-property correlations in a wide range of composition, thus the effect of substituents on the polymer solubility, dissolution rate, glass transition temperature and thermal stability will be investigated. Based on the knowledge gained here, we prepare polymer matrices for the release of a water-sensitive drug used in ocular therapy. Further topic of our project is the synthesis of mucoadhesive polymers. The aqueous solutions of thiol containing polymers synthesized via various pathways presumably interact with the mucin glycoprotein. The interaction will be proven by rheological experiments and measurement of adhesion. We assume that polymers showing such interaction will adhere to the mucosal membrane of the eye. The polymer matrices with promising characteristics will be evaluated by drug release measurements in each subproject. Chemical composition, storage conditions and the method of administration will be elaborated during the project. The most promising polymers of the project will be investigated by cytotoxicity tests. The development of potential formulations may begin as the continuation of this project.

What is the major research question?

Currently available drug formulations provide only limited therapeutic efficiency because of the poor bioavailability of the active compound which deteriorates patient compliance in several cases. The hypothesis of our research is that the functionalization of aspartic acid based polymers in simple chemical reactions enables us to prepare new types of drug delivery formulations which provide better efficiency in two well-defined therapeutic fields than the currently available formulations.
We determine for polyaspartamides in a wide composition range whether the solubility and glass transition temperature can be controlled by chemical modification of the polymers. The effect of processing conditions on morphology and dissolution rate will be investigated in the case of electrospun matrices. We choose the possible range of green solvents of electrospinning. We determine the correlation between the thiol content and the strength of bioadhesion of the polymer and investigate if the thiol groups immobilized by various synthetic pathways and the presence of other side groups affect the adhesion. From the determined structure-property relationships the promising polymers will be chosen for medical applications. The potential of these polymers will be further evaluated by cytotoxicity tests. At the end of this project, we make suggestions on polymers and related processing methods with possible therapeutic applications.

What is the significance of the research?

A wide range of aspartic acid based polymers will be synthesized in our project to explore the structure-property relationships of these polymers and we utilize this knowledge to synthesize polymers with programmed properties. The lack or limited availability of such correlations on aspartic acid based polymers and matrices gives the scientific importance of the work. We will certainly extend the general knowledge on the effect of chemical structure on solubility, glass transition temperature and strength of bioadhesion. We will synthesize polyaspartamides soluble in different green solvents and prepare electrospun matrices of these polymers for pre-defined therapeutic fields. We expect improved therapeutic efficiency of as-prepared formulations in the case of rifampicin or similar active compounds. The effect of thiol content on mucoadhesion of poly(aspartic acid)s and polyaspartamides will also be determined and water soluble formulations with strong adhesive properties will be prepared. These formulations may provide prolonged residence time on mucosal surfaces, thus sustained release of the encapsulated drug can be achieved, which ensures better bioavailability. In this project we focus on the ocular antibiotic therapy (e.g. using ofloxacin). Such formulations can greatly improve the quality of life and patient compliance due to increased efficiency. The better bioavailability results in environmental and economic benefits. The definite advantage of our polymers compared to commercially available polyacrylates is their expected biodegradability and the shorter, environmentally friendly synthesis. The systematic chemical modification planned in this project lacks from the literature, thus compared to the already known aspartic acid based cross-linked polymer matrices, our research direction is completely unique in the field.

The therapeutic efficiency of drug molecules is limited using most of the commercially available formulations (tablets, tablets with film coating, patches, injections etc.). This necessitates an increased dosage resulting in unwanted side effects and decrease of patient compliance. A serious environmental load is caused during the preparation of drug formulations which, taking into account also the poor efficiency of drug, results in environmental pollution and economic loss.
We aim at the development of new drug formulations from a macromolecule which is compatible with the human body and can increase the therapeutic efficiency. The project focuses on two well-defined therapeutic fields utilizing well-known drug molecules. We will develop a solid formulation to deliver a strongly water sensitive drug molecule to the eye. We will synthesize also a liquid formulation with prolonged residence time at the surface of the eye. It can be used to treat bacterial infections with antibiotics. A natural amino acid, aspartic acid will be used as starting material of each formulation which ensures the compatibility of the developed chemical structures with the body and low environmental risk thus these formulations can be actual alternatives to the currently available solutions.

Aszparaginsav alapú polimerek széles körét állítottuk elő, mely lehetővé tette a szerkezet-tulajdonság összefüggések megismerését, így tervezhetővé vált meghatározott tulajdonságú poliaszpartamidok és poliaszparaginsavak szintézise. Leírtuk ciszteaminnal módosított poliaszpartamidok összetétel- és pH-függő szabályozott lebomlását. Poliaszpartamidokból nanoszálas mátrixokat állítottunk elő oldószeres elektrosztatikus szálképzéssel. A poliaszpartamidok kémiai szerkezete jelentős hatással volt a gyógyszerhatóanyagok felszabadulási sebességére. Az adagolási formák mukoadhezív tulajdonságainak ismerete döntő fontosságú hatóanyag-leadó rendszerek fejlesztésében. Feltártuk a különböző funkciós csoportokat tartalmazó szintetikus poliaszpartamidok kémiai összetételének hatását a polimer-mucin kölcsönhatásokra. Szintetikus, nyálkahártya helyettesítő hidrogél szubsztrátumot fejlesztettünk. Tiolált poliaszpartamidokból kétkomponensű in situ gélesítő rendszer állítottunk elő. Az ofloxacin elnyújtott felszabadulását figyeltük meg a hidrogélből, ami alátámasztja a kifejlesztett biopolimerek potenciálját szemészeti alkalmazásokban. A hagyományos szemcseppek gyenge biohasznosulása javítható mukoadhezív tiolált polimer tartalmú készítmények alkalmazásával, amelyek hosszabb tartózkodási időt biztosítanak, mivel a tiolcsoportok diszulfid kötéseket hozhatnak létre a nyálkaréteg ciszteinben gazdag szegmenseivel. A PASP-származékok ígéretes segédanyagoknak bizonyultak szemcseppek fejlesztéséhez.

A wide range of aspartic acid based polymers was synthesized to explore the structure-property relationships, and we utilized this knowledge to synthesize polymers with programmed properties. We described the composition- and pH-dependent controlled degradation of cysteamine-modified polyaspartamides. Nanofibrous matrices from polyaspartamides were prepared by solvent electrospinning. The chemical structure of polyaspartamides had a significant effect on the release rate of active pharmaceutical ingredients. The evaluation of mucoadhesive properties of dosage forms is crucial in the development of drug delivery systems. The effect of the chemical composition of synthetic polyaspartamides with various functional side groups on polymer-mucin interactions was explored. A synthetic mucosa-mimetic hydrogel was also developed. In situ gellable polymers have potential applications as injectable formulations in drug delivery and regenerative medicine. A two-component in situ gelling system of thiolated polyaspartamides synthesized. A prolonged release of ofloxacin was observed, supporting the potential of the developed biopolymers in ophthalmic drug delivery. Poor bioavailability of traditional eye drops might be overcome by using mucoadhesive thiolated polymer dosage forms providing a longer residence time as thiol groups can form disulfide linkages with cysteine-rich subdomains of the mucus layer. PASP derivatives were found to be promising excipients for developing eye drops.
