A modern, tudományos alapokon nyugvó orvoslás ellenére, a központi idegrendszert érintő patológiás kórképek gyakran eredményeznek komoly életminőség-rontó neurológiai tüneteket. A központi idegrendszer homeosztázisának fenntartásában meghatározó szerepet tölt be a neurovaszkuláris egység. Az endotél sejtek és periciták mechanobiológiája, fokozott mechanikai igénybevételnek való kitettségük miatt, kiemelkedő jelentőséggel bír. Az agyi endotélsejtek széles kutatási skálájához viszonyítva a periciták mechanobiológája kevéssé feltárt terület.
Az agyszövet sikeres kolonizációjában aktívan részt vesznek a tumor sejtek mikrokörnyezetének sejtes elemei melyek közül a periciták szerepe nem teljesen tisztázott. Eddig számos mechanizmus felmerült a tumor disszemináció és metasztatikus organotropizmus kapcsán azonban kevés információ áll rendelkezésünkre a tumor sejtek mikrokörnyezetének mechanobiológiájáról. Továbbá fontos lenne tisztázni ebben milyen szerepet játszanak a tumor sejtek által termelt extracelluláris vezikulák.
A neurovaszkuláris egységet alkotó sejtek strukturális, morfológiai valamint nanomechanikai vizsgálata képezi kutatásaink központi témáját, Raman spektroszkópiával kiegészített atomerő mikroszkópiai módszerekre alapozva. Kisérleteinkhez nagy felbontású morfológiai és rugalmasság térképek, valamint egyedi sejtes erő spektroszkópiát kiegészítve jelölésmentes kémiai képalkotást és hiperspektrális jellemzést is alkalmazunk. Ezen módszerekkel alapvető összefüggések birtokába juthatunk a neurovaszkuláris egység fiziológiás és patológiás körülmények közötti mechanobiológiájáról.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Raman spektroszkópiával kiegészített atomerő mikroszkópia segítségével a neurovaszkuláris egység sejtes elemeinek morfológiai, strukturális valamint nanomechanmikai jellemzői közötti alapvető összefüggések feltárását tűztük ki célul. A periciták nanomechanikája illetve a neurovaszkuláris egység biofizikája patológiás folyamatokban kevéssé feltárt területek. Megértésükhöz és leírásukhoz a következő kérdéseket szeretnénk megválaszolni:
1. Milyen nanomechanikai tulajdonságokkal rendelkeznek a periciták fiziológiás és patológiás folyamatokban? Feltételezéseink szerint gyulladásos mediátorok valamint tumor sejtek képesek befolyásolni a periciták mechanikai tulajdonságait, mely részben magyarázatot adna funkciózavaraik okára a fenti folyamatokban.
2. Jelölésmentes kémiai képalkotással hiperspektrális információk alapján azonosíthatóak-e tumor sejtek és az általuk termelt extracelluláris vezikulák?
3. Képesek-e tumor sejtek által termelt extracelluláris vezikulák módosítani a tumor sejt és endotél sejt között kialakuló adhézió mértékét? Feltételezéseink szerint ezen vezikulák jelentősen befolyásolják az endotélsejtek biofizikai paramétereit, mely hatással lehet tapadási tulajdonságaikra is.
Jelen projektben tervezett sikeres kísérletek eredményei nemcsak új fundamentális tudáshoz vezethetnek, hanem a diagnosztika, terápia valamint gyógyszerfejlesztés hatékonyságának növeléséhez is hozzájárulhatnak.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

A központi idegrendszert érintő rosszindulatú sejtburjánzások többsége metasztatikus eredetű. Az áttétek kialakulásának és terjedésének komplex folyamata nem teljesen tisztázott. A rosszindulatú tumorok agyi áttétei igen rossz prognózissal és korlátozott terápiás lehetőséggel bírnak. A burjánzásnak indult daganatsejtek eredeti helyükről leggyakrabban a véráram vagy nyirokkeringés útján eljutnak a szervezet más részeibe is, ott kitapadnak, s egymástól függetlenül, de egymással párhuzamosan növekedésnek indulnak. Ebben a soklépésű folyamatban a neurovaszkuláris egység kiemelt szerepet játszik, mely gyakran gyulladásos mediátorok jelenlétében sérülékenysége miatt csak részben képes megakadályozni a tumor sejtek bejutását az agyba. A tumor sejtek által termelt extracelluláris vezikulák kulcsszerephez juthatnak ezen folyamatok sikerességében, bár ezen jelengégek ok – okozati összefüggései nagyrészt feltáratlanok. Egyedi sejt-sejt kapcsolatok nanomechanikai aspektusai szorosan összefonódnak a molekuláris szintű folyamatokkal, ennélfogva egyre hangsúlyosabb szerephez jutnak az erre irányuló kutatásokban. Egyedi sejt-adhéziós, affinitás valamint viszkoelaszticitás mérésekkel közelebb kerülhetünk a jelenség megértéséhez, ami potenciális farmakológiai vagy terápiás lehetőségeket is magában hordozhat.

A kutatás összefoglalója, célkitűzései laikusok számára

A központi iedegrendszer magasan szervezett védelmi vonalát alkotja a neurovaszkuláris egység. Elsődleges funkciója a vér-agy gát kialakítása és fenntartása, mely egyidejűleg biztosítja tápanyagok bejutását illetve káros anyagok bejutásának blokkolását az agyszövetbe. Eme magasan szervezett egység részleges vagy teljes funkcióvesztése komoly neurológiai tünetekhez vezet.
Nem csak lokálisan, de periférián kialkuló patológiás folyamatok is jelentős mértékben hozzájárulhatnak a barrier funckió csökkenéséhez. Mivel a véráram felől állandó mechanikai igénybevételnek illetve kémiai orstromnak van kitéve, kiemelkedő jelentőséggel bír a barrier sejtes elemeinek mechanobiológiai ismerete. Számos cerebrovaszkuláris kórfolyamat esetében a neurovaszkuláris egység integritása és ezzel összefüggő patológiás folyamatok viszonya nem teljesen tisztázott. A neurovaszkuláris egység kettős szerepe neuropatológiás kórképekben – a tünetek kiváltó oka és küvetkezményeként is – nem csak gát de célpont is lehet a központi idegrendszert célzó gyógyszerek esetében. Nem csak egyéni de szorosan összefonódó sejtes elemei jelentős szerephez juthatnak neurológiai kórképek kezelésében és megelőzésében egyaránt.

Despite of modern medicine, brain related pathologies often cause life impairing profound neurological symptoms. The neurovascular unit plays crucial role in the maintenance of the proper homeostasis of the central nervous system. Endothelial cells and pericytes are the most exposed to mechanical stresses their mechanobiology is of primordial importance. Little is known about the mechanobiology of the pericytes.
Successful colonization of the brain involves active participation of all cellular components at the tumor microenvironment. In addition to immune cells, cancer associated fibroblasts and endothelial cells, pericytes are key components of the newly established tumor microenvironment, whose role in brain colonization is largely unknown. Although several mechanisms were proposed for tumor dissemination, homing and metastatic organotropism little is known about the mechanobiology and the role of tumor derived extracellular vesicles in these processes.
Our research is focused on structural, morphological and nano-mechanical investigation of cells of the neurovascular unit, using atomic force microscopy combined with Raman spectroscopy. High resolution morphology, stiffness mapping and single cell force spectroscopy complemented with label free chemical imaging and hyperspectral characterization will be involved to reveal fundamental features about the mechanobiology of the neurovascular unit in physiological and pathological conditions. A more detailed characterization and description regarding the mechanobiology of the neurovascular unit points towards development and application of more effective drugs in theranostic of neurological disease.

What is the major research question?
Describe here briefly the problem to be solved by the research, the starting hypothesis, and the questions addressed by the experiments.

Using atomic force microscopy combined with Raman spectroscopy, our main aim is to explore and elucidate the connections among the morphology, structure, mechanics and biological function of the neurovascular unit in health and disease. Little is known about the nanomechanics of pericytes and there is a major caveat in our understanding how pathological conditions affects biophysical properties of the neurovascular unit. Therefore, we would like to address the following specific aims:
• First specific aim is the nanomechanical characterization of pericytes under physiological and pathological conditions. We hypothesize that inflammatory mediators and tumor cells are able to influence mechanical properties of this cell type which may explain at least partly functional changes seen under the above mentioned conditions.
• Second specific aim is to hyperspectrally characterize tumor cells and their secreted extracellular vesicles using label-free chemical imaging. We hypothesize that we will be able to clearly identify extracellular vesicles of different cellular origin.
• Third specific aim is the quantification of adhesive properties between endothelial and tumor cells induced by specific tumor-derived extracellular vesicles. We expect that tumor derived vesicles will considerably alter biophysical properties of endothelial cells.
Besides revealing new fundamental characteristics of the neurovascular unit the results expected to emerge from the planned experiments might lead to the development of new theranostics in neurovascular unit-related disease.

What is the significance of the research?

Most of the neoplasms found in the central nervous system are of metastatic origin. Metastasis formation is a complex and not entirely understood process. The poorest prognosis and the most feared complications are associated to brain metastases. Due to the lack of classical lymphatic drainage, in the process of brain metastases formation the haematogenous route is of primordial importance. In many cases the dissemination of tumor cells is taking place in the presence of inflammatory cytokines, which may imbalance the outcome of organ colonization in the favor of invading cells. The exact details on the cause and effect are partially solved. Extracellular vesicles might play key role in pre-metastatic niche formation and might be involved in metastatic organotropism of different tumor types. The exact details and mechanisms behind these processes are largely unknown.
Label-free chemical imaging has emerged as a non-destructive and non-invasive tool to intracellularly detect administered drugs without exogenous labelling. Amongst the most promising applications one can find the ability of discrimination between normal and tumor cells not only in vitro, but in vivo as well. The need for development of new analytical techniques to study extracellular vesicles has led to the ascent of Raman spectroscopy in this field.
Nanomechanical manifestations are strongly interconnected with biochemical functions, therefore alterations in pathological conditions are strong indicators of underlying processes. Besides revealing new fundamental knowledge, the results based on the experiments planned in this research proposal, might lead to new development in neurovascular unit-related disease theranostic and improvement of brain targeting drug efficacy.

The neurovascular unit is a highly regulated defense line of the central nervous system. Simultaneously, this unit is responsible to block harmful substances and carry nutrients to the brain tissue. Failure or any damage of the highly orchestrated unit might result in profound neurological symptoms.
The most important function of the unit is to build up and sustain the blood-brain barrier. Pathologies occurring at the periphery can have sever effect on the integrity of the barrier as well. As being under constant mechanical stress and chemical assault from the blood stream, detailed knowledge concerning the mechanobiology of every constituent of the barrier is of primordial importance. In many conditions when cerebrovascular abnormalities have been noted, the relationship between the breakdown of the blood-brain barrier and the pathologies is not entirely clear. The bi-faced function of the neurovascular unit in neuropathology – both in response and contributing factor to disease – indicate that the neurovascular unit is a target as well not only obstacle in drug delivery to the central nervous system. Targeting individual or strongly interconnected components of the unit holds significant promise for treatment and prevention of neurological disease.

A központi idegrendszer homeosztázisának fenntartásában kiemelkedő szerepet játszik a neurovaszkuláris egység. Ennek felépítésében kulcsszerep jut az agyi ereket bélelő endotélsejtek illetve őket körbefogó pericitáknak, melyek mechanobiológiája a véráram felől állandó mechanikai igénybevétel miatt kulcsfontosságú. Célunk a neurovaszkuláris egység mechanobiológiájának részletesebb feltárása volt fiziológiás és patológiás folyamatokban. Kimutattuk, hogy tumor sejtek által termelt extracelluláris vezikulák befolyásolják ezen sejtek tapadási tulajdonságait agyi endotélsejtekhez. Agyi metasztázisból visszaizolált adenokarcinóma sejtek tapadási mintázata eltér parentális sejtekétől. Ezen jelleg, a neurovaszkuláris egység több eleme esetén is megfigyelhető. A metasztatikus tulajdonságot több nanomechanikai jellemzőben is kimutattuk. Metasztázist képző tumor sejtek nanomechanikai jellemzése értékes információt szolgáltathat az áttétképződés sok-lépcsős folyamatának részletesebb megértésében, anti-metsaztatikus szerek, akár étrend kiegészítők kifejlesztésében.

The neurovascular unit plays crucial role in the maintenance of the proper homeostasis of the central nervous system. Endothelial cells and pericytes are the most exposed to mechanical stresses, therefore their mechanobiology is of primordial importance. Our goal was to investigate the mechanobiology of the neurovascular unit by comparing its components’ mechanical characteristics in physiological and pathological conditions. We have successfully measured the nanomechanical characteristics of invading tumor cells in contact to brain endothelial cells in presence of tumor cell pre-released extracellular vesicles. Furthermore, adhesive characteristics of breast adenocarcinoma cells were investigated towards several components of the neurovascular units. Metastatic potential was shown to have nanomechanical fingerprints and detailed characterization of adhesive parameters were conducted. All the obtained results might have important contribution towards development and application of more effective anti-metastatic drugs or dietary aspects.
