A 2002-2006 közötti időszakban végzett kutatómunkánk során olyan felismerésre jutottunk, amely lehetővé teszi az indolváz 4., 5., 6. és 7. helyzetében helyettesitők regioszelektív kialakitását az irodalomban nem ismert, preparativ szempontból rendkivül hasznos módon. A felismerés lényege a 2-etoxikarbonil-indol-4-, 5-, 6- és 7-metánszulfonsavak sajátos, új reakciója, melynek során a szulfonsav csoport a szulfonsavklorid képzés körülményei között klórra cserélhető. Igy a 4-, 5-, 6- és 7-klórmetil-indolok egyszerü preparativ módszerrel, közel kvantitativ termeléssel állithatók elő. Vizsgáltuk a 4-, 5-, 6-, 7-klórmetil-indol-2-karbonsavészterek nukleofil szubsztitúciós reakcióit. Ennek során 4-, 5-, 6-, 7-cianometil-, 4-, 5-, 6-, 7-dialkilaminometil- és 4-, 5-, 6-, 7-hidroximetil-indolokat állitottunk elő. Utóbbiak oxidációja 4-, 5-, 6-, 7-formilindolokat adott. Az indol -elektronszerkezetét közelítő rezonancia határszerkezetekből kiindulva egyszerű magyarázatot adtunk a 4-és 6-, valamint az 5- és 7-regioizomerek különböző sebességgel történő hidrolízisére (metanolízisére). Felismertük, hogy a 4-, 5-, 6-, 7-dialkilaminometil-indolok acetilkloriddal, 20oC-on, kvantitativ termeléssel a megfelelő klórmetil-indolokká alakulnak. Sikerült megvalósítanunk a dialkilaminometil-indolok előállítását a Japp-Klingemann reakció és a Fischer-féle indolszintézis együttes alkalmazásával, dialkilaminometil-anilinekből kiindulva is.

The regioselective synthesis of 4-, 5-, 6- and 7-substituted indoles as reactive synthetic intermediates is an important aspect in the chemistry of this heterocycle. We found that the 2-ethoxycarbonyl-1H-indole-4-, 5-, 6- and 7-methanesulfonic acids could easily be transformed into 4-, 5-, 6- and 7-chloromethyl-1H-indole-2-carboxylates, respectively, using typical conditions for the formation of sulfonyl chlorides. The elimination and replacement of the sulfonic acid group by chlorine of these methanesulfonic acids is actually so facile that the sulfonic acid group may be regarded as a kind of latent chlorine atom. We have developed a facile route to these indolemethanesulfonic acids by the Fischer indolisation of hydrazones obtained from the respective (aminophenyl)methanesulfonic acids through the Japp-Klingemann reaction. The 4-, 5-, 6- and 7-chloromethyl-1H-indole-2-carboxylates provided easy access to the respective cyanomethylindoles, dialkylaminomethylindoles and hydroxymethylindoles oxidized to formylindoles. The dialkylaminomethylindoles which are the regioisomeric analogues of gramine, are valuable synthetic intermediates, too: they yielded the respective (chloromethyl)indoles with acetyl chloride at rt within a few minutes in quantitative yields. These new indolic intermediates may serve as convenient starting materials for the synthesis of a number of alkaloids and compounds of pharmaceutical interest.
