A prolaktin-releasing peptid (PrRP) számos, központi idegrendszeri funkcióját tanulmányoztuk. A tejelválasztásban a PrRP-t nem találtuk fontosnak. Krónikus, de nem akut stressz után a PrRP expresszió megnőtt, az adaptáció során megváltozott szabályozási mechanizmusra utalva. A PrRP expresszióban és stresszre adott válaszreakciójában nem-és genotípus-függő változásokat találtunk. Nőstény patkányok agytörzsi PrRP-termelő sejtjeiben ösztrogén receptort (alfa típust, bétát nem) mutattunk ki, mely magyarázhatja a hímekénél magasabb PrRP expressziót és enyhébb PrRP stressz-választ. Vazopresszin-hiányos Brattleboro patkányokban magasabb agytörzsi PrRP mRNS expressziót észleltünk, ami a kontrolloktól eltérően akut stresszre az A2 katekolaminerg sejtcsoportban fokozódott. Agytörzsi, organotipikus szövetkultúrán végzett in vitro kísérletekben ugyanezen a területen a vazopresszin agonista a PrRP mRNS expressziót gátolta. Számos PrRP-pozitív idegvégződést találtunk a dorsolateralis hipotalamuszban, szinaptikus kontaktusokat létesítve melanin-koncentráló hormon-(MCH) és orexin A-termelő idegsejtekkel. Az MCH sejtek legtöbbje Fos immunpozitivvá vált 72h REM alvásmegvonást követő 3h pihenés hatására. Az orexin A pozitív sejtek nagy része aktiválódott 12h hideg-stresszt követő 3h pihenés után. Az aktivált MCH-és orexin A-pozitív neuronok közül sok állt közeli kontaktusban PrRP-pozitív végződésekkel. Munkánk során sokakat érintő metodikai problémákat is megoldottunk, eredményeit publikáltuk.

Several aspects of prolactin-releasing peptide’s (PrRP) functions in the CNS were studied. Role of PrRP was not confirmed in lactation. PrRP expression elevated after chronic but not after acute stress indicating a switch in the regulatory mechanisms during the adaptation. Gender- and genotype-specific differences were found in the PrRP expression and in the PrRP response to stress. Estrogen alpha, but not beta receptor was demonstrated in the medullary PrRP neurons of females that may explain the females’ higher PrRP expression and less sensitive PrRP reaction to chronic stress. Vasopressin-deficient Brattleboro rats showed higher medullary PrRP mRNA expression then the controls, and unlike controls acute stress evoked PrRP mRNA expression in the A2 cell group. In vitro experiments on brainstem organotypic slice cultures showed inhibitory effect of vasopressin agonist on PrRP mRNA expression in the same area. In the dorsolateral hypothalamus PrRP-positive terminals established synaptic contacts with melanin-concentrating hormone (MCH)- and orexin A-like neurons. Most of the MCH neurons were Fos-positive after 72h paradoxical sleep deprivation followed by 3h rebound. A great percent of the orexin A neurons were activated by 12h cold followed by 3h rebound. Many of the activated MCH and orexin A positive neurons formed close contacts with PrRP-positive terminals. During the work we encountered methodical problems, with widespread interest that were also solved and publicized.
