A posztdoktori pályázat keretében végzett munkám során kerestem azokat a tényezőket, amelyek elősegíthetik a beültetett idegi őssejtek túlélését a kifejlett agyszövetbe. Az elért eredmények: 1. Kifejlett egerek agykérgében és hippocampus-ában, in vitro elődifferenciáltatás után, beültetett NE-4C idegi őssejtekből, csak asztroglia-sejtek fejlődtek jelentős számban. 2. Vizsgálataink azt mutatták, hogy az NE-4C sejtek in vitro idegi differenciációja során a fejlődő központi idegrendszer különböző régióira jellemző transzkripciós faktorok széles skálája aktiválódik. Az elkötelezetlen NE-4C őssejtek, tehát széles fejlődési potenciállal rendelkeznek. Az érett NE-4C idegsejtek között a központi idegrendszer fő transzmitter fenotípusát képviselő GABA-erg és glutamaterg neuronok egyaránt előfordulnak. 3. In vitro vizsgálataink azt mutatják, hogy a retinsav, amelyről ismert, hogy számos in vitro rendszerben az idegsejt irányú differenciációt növeli, az asztrocita-fejlődés késői szakaszában gátolja az asztroglia sejtek kialakulását. A kifejlett központi idegrendszer olyan szöveti környezettel rendelkezik, amelyből hiányoznak az idegi őssejtek, idegi progenitorok és idegsejt-előalakok nagy számú differenciációját, túlélését és integrációját elősegítő tényezők. Ugyanez a környezet az asztrogliasejtek differenciációját / túlélését megengedi. A fentiek szerint, hogy az idegsejtek integrációját differenciációját elősegítsük a befogadó szövetet kell megfelelő módon változtatni.
Results in English
During the three-year postdoc period, my research was focused on the clarification of some conditions promoting the survival and neural tissue-type differentiation of implanted neural stem cells inside the mature CNS. Results: After a short-term pre-differentiation in vitro, NE-4C neural stem cells can develop into astrocytes inside the mature CNS, while do not differentiate to neurons. In the course of the in vitro neural differentiation of NE-4C cells, a wide spectrum of regionally restricted transcription factors, those expressed in non-overlapping areas of the CNS, get activated. Therefore, the NE-4C neural stem cells have a developmental potential to develop multiple, positionally determined neural cell types. Accordingly, NE-4C neurons can acquire GABAergic and glutamatergic phenotypes, as well. The data indicate that the neurotransmitter phenotype of neurons is determined by environmental factors. According to our in vitro data, retinoic acid, a known inducer of neuronal differentiation, impairs the formation of the astroglial phenotype. The glia formation is inhibited by retionic acid in some late phases of gliogenesis. The environment of the mature brain tissue does not provide factors promoting the survival and tissue integration of non-differentiated neuronal progenitors or precursors, but allows the development of astrocytes. In order to elaborate methods for the replacement of decayed neurons, the host-environment has to be characterized and modified.
