A VCD-spektroszkópiát kis- és közepes méretű peptidek, valamint királis szerves molekulák [mint például β-laktámok és N-S(IV)-O axiális kötésrendszerű spiro-λ4-szulfánok] széles körében sikeresen alkalmaztuk konformáció vagy abszolút konfiguráció meghatározására. Kimutattuk, hogy β-aminosavat tartalmazó ciklotetrapeptidekben a β-aminosavegység jelenléte a szomszédos α-aminosavegységek körül γ-kanyart (C7) indukál, és csak nagyobb, kevésbé feszült ciklopenta- és hexapeptidek esetében jönnek létre a várt C6, C8 H-kötéses ún. pszeudo-γ-kanyar szerkezetek. Cisz-2-aminociklopentán-karbonsav alternáló enantiomerjeiből felépülő lineáris oligo-β-peptidek esetében VCD-spektroszkópiával stabilis H10/H12 hélix szerkezetet azonosítottunk. Az oldószer erős kölcsönhatása a vizsgált molekulával vagy a molekulák asszociációja megnehezítheti a spektrum értelmezését, ennek kiküszöbölésére az alacsony hőmérsékletű (8-12 K) mátrixizolációs VCD-technikát vezettük be, mely a majdnem kölcsönhatás-mentes környezet mellett egyidejűleg nagyobb spektrális felbontást is eredményez, így több konformer detektálását teszi lehetővé. Védett aminosavszármazékok vízzel való kölcsönhatását vizsgálva kimutattuk, hogy a VCD-spektroszkópia a kiralitástranszfer jelenség alapján alkalmazható királis vegyületek akirális molekulákkal (például oldószerrel) képezett komplexeinek kimutatására is.

VCD spectroscopy has been successfully applied in the determination of the conformation or absolute configuration of a wide range of small and mid-sized peptides, as well as other chiral organic compounds, such as β-lactams and spiro-λ4-sulfanes with N-S(IV)-O axial bond system. It has been shown that cyclic tetrapeptides containing β-amino acid residues form γ-turns (C7) centered at the neighboring α-amino acid units, while the expected pseudo-γ-turns (C6 or C8) are only present in larger, less strained cyclic penta- and hexapeptides. In linear oligo-β-peptides built up from alternating enantiomers of cis-2-amino cyclopentanecarboxylic acid residues formation of a stable H10/H12 helix structure was identified by VCD spectroscopy. Strong interaction between the studied molecule and the solvent or intermolecular associations may cause difficulties in the interpretation of spectra. To overcome this, we introduced the low-temperature (8-12 K) matrix isolation VCD spectroscopic technique which provides an almost interaction free environment and higher achievable spectral resolution at the same time, permitting the detection of a higher number of conformers. By studying the interaction of protected amino acids with water we showed that VCD spectroscopy, based on the chirality transfer phenomenon, can be applied in the detection of complexes between chiral substrates and achiral guest (e.g. solvent) molecules.
