A pályázat célja volt a D4 dopamin receptor gén 5’szabályzó régiójában a transzkripciót befolyásoló funkcionális illetve a neutrális polimorfizmusok elkülönítése, valamint a lehetséges transzkripciós faktor kötőhelyek, enhancer és silencer elemek felderítése. A DRD4 promóterben található –521 CT polimorfizmusról megállapítottuk, hogy - más kutatók korábbi eredményével ellentétben - érdemben nem befolyásolja a promóter aktivitását. A 120 bp-os ismétlődéses polimorfizmusról (120 bp VNTR) kimutattuk, hogy minél többször fordul elő a 120 bp szakasz a promóterben, annál kisebb a promóter aktivitása. A 120 bp VNTR tehát egyértelműen represszor elemként viselkedik. Tranziens transzfekciós vizsgálataink egy negatív moduláló elem jelenlétére utalnak a –1571 nt és a –799 nt szakaszon. Valószínű, hogy ez az elem a 120 bp VNTR. A RAR/RXR heterodimer jelentős mértékben hat a promóter aktivitására tranziens transzfekciós kísérletben. Ez felveti a reténsav szerepét a DRD4 gén aktiválásában a dopaminerg idegsejtek differenciációja során. A Dopamin receptor reguláló faktor kotranszfekciós kísérletekben háromszorosára növelte a promóter aktivitását. Feltehetően a promóterben található Sp1 kötőhelyek is szerepet játszanak a DRRF hatásmechanizmusában. A hipoxiás környezet képes fokozni a DRD4 gén expresszióját neuronális sejtvonalakon. Eredményeink segíthetik megérteni az emberi személyiség jegyek genetikai alapjait, illetve bizonyos pszichiátriai kórképek genetikai hátterének feltárását.

Efforts in this study focused on the polymorphisms of the 5’ regulatory region of D4 dopamine receptor gene. We investigated their possible role in transcriptional regulation and searched for other regulatory elements. Evaluating the effect of the –521 CT polymorhism, we observed no significant difference between the activities of the –521C and –521 T alleles. Transient expression analysis of allelic variants of the 120 bp VNTR revealed that increasing repeat numbers in the promoter resulted in decreasing transcriptional activities in neuroblastoma as well as retinoblastoma cells. The 120 bp VNTR acts as a negative modulator element. Other data also pointed to a putative repressor element located in the promoter region between nucleotides –1571 and –799. This element is probably the 120 bp VNTR. We found a putative RARE element and investigated it’s role in DRD4 expression in vitro. Cotransfection of the RAR/RXR heterodimer elevated the promoter activity more than sixfold. Suggesting that retinoic acid plays an important role in the DRD4 gene regulation during the dopaminergic neuronal differentiation. Dopamine receptor regulatory factor increased the activity of the DRD4 promoter threefold in transient transfection experiments. Probably DRRF acts through the Sp1 binding sites in the promoter. Hypoxia treatment can induce the DRD4 gene expression in several neuronal cell lines. This study contributes to the understanding the genetic components of the human personal traits and the genetic background of several psychiatric syndromes.
