A projekt során FRET módszereket fejlesztettünk fehérjék asszociációjának jellemzésére: - Megoldottuk a GFP variánsok közötti FRET mérések kvantitatívvá tételét és három fehérje kölcsönhatásának vizsgálatát FRET-tel - A projekt e részének legfontosabb eredménye a homoklaszterek kvantitatív jellemzésére kidolgozott módszer mellyel becslést lehet adni az egy homoklaszterben levő molekulák számára. Az eljárás felhasználásával megállapítottuk, hogy az ErbB2 nyugvó sejteken nagyfokú homoasszociációs hajlammal rendelkezik (50-100 ErbB2-ből álló csoportokat alkot), melyek stimuláció hatására szétesnek. Az ErbB1 ezzel ellentétesen viselkedik. Az ErbB2 klasztermérete fordítottan arányos a fehérje aktiváltsági szintjével, és valószínűleg a növekedési faktorok által kiváltott ErbB1 vagy ErbB3 aktivációhoz szükséges ErbB2 raktározási helyéül szolgál A projekt másik felében megállapítottuk, hogy a CD44 az ErbB komplexum részét képezi. A kölcsönhatásnak több biológiai hatása van: - A CD44 ligand hialuronsav hozzájárul a trastuzumab (ErbB2 ellenes antitest) epitópjának maszkírozásához - Az EGF és a heregulin a sejtek motilitásának fokozásával járó CD44 sheddinget vált ki - Legfontosabb eredményként megállapítottuk, hogy a trastuzumab ADCC reakció révén fejti ki hatását, és hogy a trastuzumab rezisztencia kialakulásában az epitóp maszkírozás jelentős szerepet játszik, hiszen a hialuronsav által alkotott mátrix kiépülésének gátlásával a trastuzumab rezisztencia kialakulása gátolható

In the project we developed FRET methods for the characterization of protein associations: - We solved the problem of quantifying FRET between GFP variants and measuring the association of 3 proteins at the same time - The most important result of this part of the project is the method developed for the quantitative measurement of the number of molecules in a single homocluster. Using the approach we determined that ErbB2 has a profound propensity for homoassociation in quiescent cells; it forms clusters containing 50-100 ErbB2 which is disassembled upon growth factor stimulation. ErbB1 behaves in the opposite way. The homocluster size of ErbB2 was inversely proportional to its activation state. ErbB2 homoclusters most likely serve as a reservoir of inactive ErbB2 molecules ready to be recruited by growth factor activated ErbB1 or ErbB3. In the second part of the project we determined that CD44 is an integral part of the ErbB complex. This interaction has several biological consequences: - the ligand of CD44, hyaluronan, contributes to the masking of the epitope of trastuzumab (anti-ErbB2 antibody) - EGF and heregulin induce CD44 shedding leading to enhanced cellular motility - Most importantly, trastuzumab induces its anti-tumor effect by ADCC, and one of the reasons of trastuzumab resistance is epitope masking, since blocking the development of the extracellular matrix formed by hyaluronan inhibited trastuzumab resistance.
